We developed an analytical method using an on-line column-switching liquid chromatography with triple quadrupole mass spectrometry (LC/MS/MS) for quantifying multiple steroids in serum. Using the developed method, we evaluated the serum concentration of nine steroids (cortisol, corticosterone, cortisone, 11-deoxycortisol, 21-deoxycortisol, deoxycorticosterone, progesterone, 17α-OH-progesterone and aldosterone) in dogs with hyperadrenocorticism (HAC). Serum was mixed with stable isotope internal standards and thereafter purified by the automated column-switching system. The limit of detection ranged 2–16 pg/ml for nine steroids. In the baseline samples, five steroids (cortisol, corticosterone, cortisone, 11-deoxycortisol, and 17α-OH-progesterone) were detected in all dogs. The concentrations of cortisone, 11-deoxycortisol, and 17α-OH-progesterone in dogs with HAC (n=19) were significantly higher those in dogs without HAC (n=15, P<0.02). After the adrenocorticotropic hormone stimulation test, six steroids (cortisol, corticosterone, cortisone, 11-deoxycortisol, 17α-OH-progesterone, and deoxycorticosterone) were above the limit of quantification in all dogs. Cortisol, corticosterone, cortisone, and deoxycorticosterone concentrations of dogs with HAC were significantly higher than those of dogs without HAC (P<0.02). In addition, 11-deoxycortisol and 17α-OH-progesterone concentration was higher in dogs with HAC than in dogs without HAC (P=0.044 and P=0.048, respectively). The on-line column-switching LC/MS/MS would be feasible for measuring multiple steroids in dog serum. The results suggest that cortisone, 11-deoxycortisol, and 17α-OH-progesterone would be related to HAC. Further studies are warranted to assess the clinical feasibility of steroid profile in dogs with HAC. 相似文献

In this paper, perforated composite panel was combined with porous and resonance structures to investigate the influence on acoustic absorption and resilient properties. The perforated composite panel was fabricated based on highdensity flexible-foam via perforating and reinforcing with laminated hybrid nonwoven fabric. Effect of aperture size (AS) (ranging from 3 mm to 6 mm), perforation ratio (PR) (5 %, 10 %, 15 % and 20 %) and perforation depth (PD) (25 %, 50 %, 75 % and 100 %) on the compressive hardness, rebound resilience and acoustic absorption properties was explored. Multiply hybrid nonwoven fabric which was fabricated with low-melting point polyester (LMPET), flame-retardant polyester (FRPET) and recycled Kevlar fibers was utilized to reinforce the flexible composites and improve the acoustic property. Nonwoven that was fabricated with entangled LMPET fibers had porous structures which could reinforce the flexible foam and enhance the acoustic absorption properties. The result revealed that the continuity and supporting of porous flexible foam had directly influence the compressive hardness. The maximum hardness of the flexible-foam based perforated composites reached 420 N. The rebound resilience result showed that the sample had high resilient structure and the resilience was up to 48 %. The perforated flexible composites plate (PFP) with 4 mm-AS performed the highest acoustic absorption coefficient at 0.9. The acoustic absorption coefficient was higher than 0.8 in the frequency range from 800 to 1600 Hz and 1600 to 2400 Hz when perforated composites had 4 mm-AS at 5 % and 10 % perforation ratio. With the increase in perforation ratio, absorption peak moved from 3200 Hz to 4000 Hz. Hybrid nonwoven laminated layer help to broaden the frequency range of acoustic absorption of perforated high-density flexible foam based composites panel. In order to search for new bioactive substances from marine organisms, we have investigated the acetone extracts of the soft coral Sarcophyton ehrenbergi collected at San-Hsian-Tai, Taitong County, Taiwan. Chromatographic fractionation of the extracts of the octocoral S. ehrenbergi led to the isolation of three new cembranoids, (+)-12-ethoxycarbonyl-11Z-sarcophine (1), ehrenbergol A and B (2 and 3). The structures of these isolated metabolites were elucidated through extensive spectroscopic analyses. Our earlier studies indicated the high expression of a UV-damaged-DNA binding activity in zebrafish (Danio rerio) embryos at 12?h postfertilization (hpf). Two 30- to 35-kDa polypeptides homologous to the N-terminal lipovitellin 1 (Lv1) domain of the 150-kDa zebrafish vitellogenin 1 (zfVg1) were identified as the damage recognition factors in zebrafish extracts, and the metal-chelating agent 1,10-phenanthroline (OP) was found to inhibit the embryonic UV-damaged-DNA binding activity. This study further explored the DNA damage-sensing components in 12 hpf zebrafish extracts. UV-damaged-DNA binding proteins were enriched from zebrafish extracts by isoelectrofocusing. Both OP-sensitive and OP-stimulated, UV-damaged-DNA binding activities were detected in fractionated zebrafish extracts. Two-dimensional gel electrophoresis of proteins captured by an immobilized oligonucleotide carrying a UV-induced (6-4)photoproduct (6-4PP) revealed a 25-kDa polypeptide as the major 6-4PP-binding factor in an OP-stimulated fraction. Three 25-kDa factors that bound weakly to 6-4PPs were also isolated. The four polypeptides having pIs between 7.0 and 7.3 were unreactive to an anti-zfVg1 antibody targeting the Lv1 domain. Mass spectral analysis showed the appearance of amino acid sequences LPIIVTTYAK and IPEITMSK in all 25-kDa polypeptides and sequences exactly matching those contained in the four factors exist only in the C-terminal Lv2 domain of zfVg1, reflecting the origination of these factors from enzymatic cleavage of the Lv2 domain at slightly different positions. The OP-stimulated fraction produced a much stronger UV-dependent DNA incision activity in the presence than in the absence of OP, suggesting the association of these factors with DNA damage repair under metal-deficient conditions. 相似文献

In the present study, we examined the effects of climate on temporal variation in the abundance and geographical distribution of demersal fishes at both interannual and decadal scales in the Tsushima Warm Current region of the Japan Sea using single‐trawler fishery data (1972–2002). This single‐trawler fishery targeted multiple species with different geographical affinities and life history traits. Thus, these data were suitable for examining the biological mechanisms that underlie how species respond to climate variability. Our results indicate that the distributional changes of species in response to decadal climate variability are best explained by asymptotic length, which indicates that warming has greater negative effects on larger fishes in the Japan Sea. However, none of the variables examined (including geographical affinity, asymptotic length and age at maturation) could explain the shift in abundance at interannual or decadal scales. It should be noted that life history traits and geographical affinities only provide partial explanations of the responses of species to environmental variability, thereby suggesting that other factors, such as interactions among species, may be involved in mediating species responses. 相似文献

The aim of this work was to determine the enzymatic activity of phosphofructokinase (PFK), malate dehydrogenase (MDH) and isocitrate dehydrogenase (IDH) in boar spermatozoa and study their participation in bicarbonate‐induced capacitation and follicular fluid‐induced acrosome reaction. Enzymatic activity of these enzymes was determined spectrophotometrically in extracts of boar spermatozoa. Sperm suspensions were incubated in the presence of bicarbonate (40 mM), a well‐known capacitation inducer, or follicular fluid (30%), as an acrosome reaction inducer, and different concentrations of oxoglutarate, oxalomalate and hydroxymalonate, inhibitors of PFK, IDH and MDH, respectively. Capacitation percentages were determined by the fluorescence technique of chlortetracycline (CTC), and true acrosome reaction was determined by trypan blue and differential–interferential contrast, optical microscopy. The activity of PFK in boar spermatozoa enzymatic extracts was 1.70 ± 0.19 U/1010 spermatozoa, the activity of NAD‐ and NADP‐dependent IDH was 0.111 ± 0.005 U/1010 and 2.22 ± 0.14 U/1010 spermatozoa, respectively, and the activity of MDH was 4.24 ± 0.38 U/1010 spermatozoa. The addition of the specific inhibitors of these enzymes prevented sperm capacitation and decreased sperm motility during capacitation and inhibited the acrosome reaction (AR), without affecting the sperm motility during this process. Our results demonstrate the participation of PFK, IDH and MDH in bicarbonate‐induced capacitation and follicular fluid‐induced acrosome reaction in boar spermatozoa, contributing to elucidate the mechanisms that produce energy necessary for these processes in porcine spermatozoa. 相似文献

Soil carbon dioxide (CO2) respiration is one of the important soil health parameters that provides a general assessment of soil microbial activity and soil quality. Soil respiration rates, however, have not been widely applied in soil testing protocols mainly because the traditional methods are either inconvenient, technically cumbersome or too expensive. Currently, only two methods are available for a true real-time soil respiration rate determination (<2 h): the infrared gas analyzer (IRGA) and the microrespirometer (MR or MicroRes®) methods. We analyzed the real-time soil respiration rates of 20 soil samples from fifteen states after various periods of incubation using the IRGA method and the MR method. The measured soil respiration rates ranged from 0.4 µL CO2/h/g to 9.0 µL CO2/h/g. Both methods show precision in soil respiration determinations (CV = 12.7% and 11.9%, respectively). Comparison of the results between the IRGA and MR methods indicates high degrees of agreement (r2 = 0.914). This study shows that the MR method is a simpler and more cost-effective alternative for real-time soil respiration rate determinations. 相似文献

and secure protections are also emphasized gradually. To shield these electromagnetic wave radition jeopardizing people’s
health, in this study, stainless steel wires were the core yarn and bamboo charcoal polyester textured yarns were the wrapped
yarn. The bamboo charcoal polyester/stainless steel (BC/SS) complex yarns were manufactured using a rotor twister machine.
The BC/SS complex knitted fabrics were woven with the complex yarns employing a circular knitting machine. Three manufacture
parameters were the wrapped amount of the complex yarn (2 to 6 turns/cm), the lamination amount of the knitted fabrics (1
to 6 layers) and lamination angles of the knitted fabrics (0°/0°/0°/0°/0°/0°, 0°/45°/90°/−45°/0°/45°, and 0°/90°/0°/90°/0°/90°).
The knitted fabric exhibited the lowest surface resistance 32.3 Ω/sq. Optimum electromagnetic shielding effectiveness (EMSE)

Studies were conducted in the laboratory and greenhouse to determine the distribution of Erwinia rhapontici in plants arising from naturally infected seeds of pea or artificially inoculated seeds of bean and wheat, and whether the
pathogen is transmitted to the subsequent generation of seeds. Infected seeds were planted in pots of Cornell mix in the greenhouse,
and sampled at specified intervals throughout the plant growth cycle (seedling stage, elongation stage, flowering stage, seed
formation stage, and maturity). Plating of surface sterilized lateral roots, tap roots, basal stems, mid-stems, apical stems,
petioles, pods, and seeds of pea and bean, and of lateral roots, sub-crown internodes, basal stems, mid-stems, apical stems,
peduncles, glumes, and seeds of wheat revealed that the bacterial pathogen spread from infected seeds to the lower parts of
the plant tissues, but failed to spread further to the seeds produced on these plants. The study concludes that E. rhapontici did not establish systemic infection throughout the plants. Possible mechanisms of infection of seeds are discussed. 相似文献
