In model terrestrial ecosystems maintained for three plant generations at elevated concentrations of atmospheric carbon dioxide, increases in photosynthetically fixed carbon were allocated below ground, raising concentrations of dissolved organic carbon in soil. These effects were then transmitted up the decomposer food chain. Soil microbial biomass was unaffected, but the composition of soil fungal species changed, with increases in rates of cellulose decomposition. There were also changes in the abundance and species composition of Collembola, fungal-feeding arthropods. These results have implications for long-term feedback processes in soil ecosystems that are subject to rising global atmospheric carbon dioxide concentrations.  相似文献

Immunotherapies targeting checkpoint molecule programmed cell death 1 (PD‐1) protein were shown to be effective for treatment of non‐Hodgkin lymphoma in people, but little is known about the expression of PD‐1 or its ligand PD‐L1 by canine lymphoma. Therefore, flow cytometry was used to analyse expression of PD‐1 and PD‐L1 in canine lymphoma, using fine‐needle aspirates of lymph nodes from 34 dogs with B cell lymphoma (BCL), 6 dogs with T cell lymphoma (TCL) and 11 dogs that had relapsed. Furthermore, fine‐needle aspirates were obtained from 17 healthy dogs for comparison. Lastly, the impact of chemotherapy resistance on expression of PD‐1 and PD‐L1 was assessed in vitro. These studies revealed increased expression of PD‐L1 by malignant B cells compared to normal B cells. In the case of TCL, tumour cells and normal T cells both showed low to negative expression of PD‐1 and PD‐L1. In addition, tumour infiltrating lymphocytes from both BCL and TCL had increased expression of both PD‐1 and PD‐L1 expression compared to B and T cells from lymph nodes of healthy animals. In vitro, chemotherapy‐resistant BCL and TCL cell lines exhibited increases in both PD‐1 and PD‐L1 expression, compared to non‐chemotherapy selected tumour cells. These findings indicate that canine lymphomas exhibit upregulated checkpoint molecule expression, though the impact of checkpoint molecule expression on tumour biological behaviour remains unclear.  相似文献

Two outbreaks of an encephalitis apparently induced by an attenuated live distemper vaccine occurred in a large ferret breeding establishment in New Zealand. Approximately 350 of 6,000 young ferrets 16-22 weeks old died. Many were found dead with no premonitory signs, others showed severe neurological signs. Some with central nervous system (C.N.S.) signs recovered. Pathological examination showed no gross abnormalities except for a few with mild conjunctivitis, rhinitis and lung emphysema. Microscopically there was a moderate to massive non-inflammatory necrosis of hippocampal nerve cell bodies. In those animals which survived for several days with CNS signs there was also a mild to moderately severe non-supportive encephalitis, and in some of these distinct neuronal intranuclear and intracytoplasmic eosinophilic inclusion bodies were seen. Some ferrets also had a bronchiolitis with intracytoplasmic eosinophilic inclusion bodies in bronchiolar epithelium. All these lesions suggest that a distemper like condition was involved. About half of the ferrets also had a mild to severe inflammatory myocardial necrosis.  相似文献

Summary The purpose of this study was to examine the influence of the moisture level on the cell-wall material in wood using pulsed proton nuclear magnetic resonance. The wood species used were western hemlock (Tsuga heterophylla (Raf.) Sarg.) and sitka spruce (Picea sitchensis (Bong.) Carr.), distinguishing between heartwood and sapwood regions. The moisture contents of the specimens were below the fibre saturation point and they were conditioned to equilibrium moisture contents based on initial desorption, adsorption and secondary desorption processes. From the FID experiments, the NMR-based moisture contents and the solid-wood lineshape second moments were determined. Average relative proton-spin densities, which were needed to calculate the NMR-based moisture contents, were determined from known moisture contents and they were: hemlock sapwood: 0.616; hemlock heartwood: 0.537; spruce sapwood: 0.679; and, spruce heartwood: 0.446. The average RSD value, considering both heartwood and sapwood, for western hemlock species was 0.577 and for sitka spruce was 0.563; these are close to published RSD values for other species. The condition as to how the equilibrium moisture content was attained did not influence the second moment for hemlock; however, for spruce sapwood, the second moments were sorption dependent. The hemlock M2 decreased from about 5.1 × 109 s-2 at low MNMR to 4.5 × 109 s-2 (heartwood) and 4.3 × 109 s-2 (sapwood) at higher mnmr. The adsorption and secondary desorption M2 for the spruce sapwood region decreased from about 5.0 × 109 s-2 at low mnmr to about 4.1 × 109 s-2 near the MF, whereas M2 for the spruce heartwood decreased from about 4.3 × 109 s-2 at low MNMR to about 3.5 × 109 s-2 near MF. Epidemiological studies suggest that consumption of flavonol-rich diets decreases the risk of developing heart disease and certain cancers. Recent studies have detected flavonol conjugates in blood and urine following various dietary interventions. To assess to what extent flavonols also accumulate in tissues, where they might be expected to exert anti-carcinogenic and anti-atherogenic effects, [2-(14)C]quercetin-4'-glucoside was synthesized and fed to rats. After 60 min, 93.6% of the ingested radioactivity was recovered from the intestine, incorporated into 18 metabolites that had undergone deglycosylation followed by varying degrees of glucuronidation, methylation, and/or sulfation. [(14)C]Quercetin, the aglycon of the radiolabeled substrate, was present in the intestine and in trace amounts in the liver but was not detected in the plasma and kidneys. The original [2-(14)C]quercetin-4'-glucoside was detected exclusively in the intestine, where it accounted for only 26.2% of the radioactivity. The remainder of the recovered radioactivity was located mainly in the plasma, liver, and kidneys as (14)C-labeled metabolites. However, compared to the quantities in the gastrointestinal tract, the levels of metabolites in plasma and body tissues were very low, indicating only limited absorption into the blood stream. The data demonstrate that quercetin-4'-glucoside, which is a major flavonol in onions, undergoes rapid and extensive metabolism in the intestine, and this appears not to be associated to any extent with transport across the gut wall into the blood stream.  相似文献

The influence of pH was examined, over the range from 6 to 14, on the amounts of p-hydroxybenzoic, vanillic, p-coumaric, ferulic and syringic acids, p-hydroxybenzaldehyde and vanillin, extracted from four soils and associated roots or leaf litter. Adjustment of pH was obtained by the addition of graded amounts of Ca(OH)2 to water or by 2 m NaOH. The roots associated with three of the soils were from permanent pasture, perennial ryegrass and red clover, while the leaf litter associated with the fourth soil was from beech.The amounts of each phenolic compound extracted increased continuously with increasing pH, from a “threshold” value which varied between pH 7.5 and 10.5. The amounts extracted by water alone from the soil under permanent pasture, at pH 5.8, were equivalent to concentrations in the soil solution ranging from 1.4 μm for p-hydroxybenzoic acid to < 10 nm for ferulie acid. Amounts of up to 2000 times greater than these were extracted by 2 m NaOH. Similar effects of extractant pH were found with the other soils.Comparisons of the amounts of the phenolic compounds extracted from the soils, with the amounts extracted from the associated roots or leaf litter, suggested that substantial proportions of the soil phenolic compounds were either derived from organic residues more than 4 yr old or were the result of microbial synthesis.  相似文献
