Erbovirus is a genus of the family Picornaviridae and equine rhinitis B virus (ERBV) is the sole species. Erboviruses infect horses causing acute respiratory disease and sub-clinical and persistent infections. Despite the high seroprevalence and worldwide distribution of these viruses, the pathogenesis and antigenic structure of the three ERBV serotypes (ERBV1, 2 and 3) is poorly understood. To characterise linear epitopes on ERBV structural proteins, a set of fusion proteins were expressed in Escherichia coli. These proteins were tested in Western blot and ELISA and reactive proteins were also used to identify neutralisation epitopes. VP1 contained serotype specific epitopes whereas VP2 was highly cross-reactive across the serotypes. The C-terminus of VP1 accounted for most of the reactivity of full-length VP1 and was also the location of a neutralising site in each serotype.  相似文献

Determination of the atmospheric concentrations and lifetime of trichloroethane (CH(3)CCI(3)) is very important in the context of global change. This halocarbon is involved in depletion of ozone, and the hydroxyl radical (OH) concentrations determined from its lifetime provide estimates of the lifetimes of most other hydrogen-containing gases involved in the ozone layer and climate. Global measurements of trichloroethane indicate rising concentrations before and declining concentrations after late 1991. The lifetime of CH(3)CCI(3) in the total atmosphere is 4.8 +/- 0.3 years, which is substantially lower than previously estimated. The deduced hydroxyl radical concentration, which measures the atmosphere's oxidizing capability, shows little change from 1978 to 1994.  相似文献

The factors that influence changes in the upland plant communities of Scotland, and hence result in changes in the landscape, are examined with particular reference to the decline in the cover of heather moorland. These factors include natural disturbances, such as climate change and fire, but the influence of man-made disturbances on the landscape, notably due to sheep grazing and deforestation, has been much greater. Despite this, it is argued that plant communities differ in their fragility and some current upland landscapes are not particularly sensitive to some types of disturbances, at least under moderate herbivore loads. However, the impact of potential reductions in herbivore loads, in combination with other factors (e.g., climate change and increased atmospheric nitrogen deposition), on the rate of vegetation change cannot be predicted with confidence at present. Understanding the mechanisms underlying spatial and temporal variation in the sensitivity of upland plant communities to increasing disturbances in the future is the challenge facing both ecologists and those interested in landscape management.  相似文献

Determining the relative temperature sensitivities of the decomposition of the different soil organic matter (SOM) pools is critical for predicting the long-term impacts of climate change on soil carbon (C) storage. Although kinetic theory suggests that the temperature sensitivity of SOM decomposition should increase with substrate recalcitrance, there remains little empirical evidence to support this hypothesis. In the study presented here, sub-samples from a single bulk soil sample were frozen and sequentially defrosted to produce samples of the same soil that had been incubated for different lengths of time, up to a maximum of 124 days. These samples were then placed into an incubation system which allowed CO2 production to be monitored constantly and the response of soil respiration to short-term temperature manipulations to be investigated. The temperature sensitivity of soil CO2 production increased significantly with incubation time suggesting that, as the most labile SOM pool was depleted the temperature sensitivity of SOM decomposition increased. This study is therefore one of the first to provide empirical support for kinetic theory. Further, using a modelling approach, we demonstrate that it is the temperature sensitivity of the decomposition of the more recalcitrant SOM pools that will determine long-term soil-C losses. Therefore, the magnitude of the positive feedback to global warming may have been underestimated in previous modelling studies.  相似文献

Protein interaction mapping using large-scale two-hybrid analysis has been proposed as a way to functionally annotate large numbers of uncharacterized proteins predicted by complete genome sequences. This approach was examined in Caenorhabditis elegans, starting with 27 proteins involved in vulval development. The resulting map reveals both known and new potential interactions and provides a functional annotation for approximately 100 uncharacterized gene products. A protein interaction mapping project is now feasible for C. elegans on a genome-wide scale and should contribute to the understanding of molecular mechanisms in this organism and in human diseases.  相似文献

Three closely related genes, GluR1, GluR2, and GluR3, encode receptor subunits for the excitatory neurotransmitter glutamate. The proteins encoded by the individual genes form homomeric ion channels in Xenopus oocytes that are sensitive to glutamatergic agonists such as kainate and quisqualate but not to N-methyl-D-aspartate, indicating that binding sites for kainate and quisqualate exist on single receptor polypeptides. In addition, kainate-evoked conductances are potentiated in oocytes expressing two or more of the cloned receptor subunits. Electrophysiological responses obtained with certain subunit combinations show agonist profiles and current-voltage relations that are similar to those obtained in vivo. Finally, in situ hybridization histochemistry reveals that these genes are transcribed in shared neuroanatomical loci. Thus, as with gamma-aminobutyric acid, glycine, and nicotinic acetylcholine receptors, native kainate-quisqualate-sensitive glutamate receptors form a family of heteromeric proteins.  相似文献
