Habitat loss and associated fragmentation effects are well-recognised threats to biodiversity. Loss of functional connectivity (mobility, gene flow and demographic continuity) could result in population decline in altered habitat, because smaller, isolated populations are more vulnerable to extinction. We tested whether substantial habitat reduction plus fragmentation is associated with reduced gene flow in three ??decliner?? woodland-dependent bird species (eastern yellow robin, weebill and spotted pardalote) identified in earlier work to have declined disproportionately in heavily fragmented landscapes in the Box-Ironbark forest region in north-central Victoria, Australia. For these three decliners, and one ??tolerant?? species (striated pardalote), we compared patterns of genetic diversity, relatedness, effective population size, sex-ratios and genic (allele frequency) differentiation among landscapes of different total tree cover, identified population subdivision at the regional scale, and explored fine-scale genotypic (individual-based genetic signature) structure. Unexpectedly high genetic connectivity across the study region was detected for ??decliner?? and ??tolerant?? species. Power analysis simulations suggest that moderate reductions in gene flow should have been detectable. However, there was evidence of local negative effects of reduced habitat extent and structural connectivity: slightly lower effective population sizes, lower genetic diversity, higher within-site relatedness and altered sex-ratios (for weebill and eastern yellow robin) in 10 × 10?km ??landscapes?? with low vegetation cover. We conclude that reduced structural connectivity in the Box-Ironbark ecosystem may still allow sufficient gene flow to avoid the harmful effects of inbreeding in our study species. Although there may still be negative consequences of fragmentation for demographic connectivity, the high genetic connectivity of mobile bird species in this system suggests that reconnecting isolated habitat patches may be less important than increasing habitat extent and/or quality if these need to be traded off. 相似文献

A 56‐day feeding trial was conducted to elucidate the effects and mechanism action of dietary α‐linolenic acid (ALA, 18:3n‐3) on lipid accumulation and fatty acid profile of muscle, hepatopancreas and intraperitoneal fat (IPF) in juvenile grass carp using three isonitrogenous and isoenergetic semi‐purified diets containing 0.0% (control group), 1.0% and 2.0% ALA, respectively. The lowest intraperitoneal fat (IPF) ratio was found in 2.0% group. In the muscle, hepatopancreas and IPF, docosahexaenoic acid (DHA, 22:6n‐3) and eicosapentaenoic acid (EPA, 20:5n‐3) contents increased with the increase in dietary ALA. In the IPF, caspase 3, caspase 8 and caspase 9 showed the highest activities in 2.0% group, while the value of Bcl‐2/Bax (B‐cell leukaemia 2/Bcl‐2‐associated X protein) reached the lowest. Meanwhile, swelling of the IPF mitochondria was observed in 2.0% group. The gene expressions of fatty acid desaturase (FAD) and fatty acid elongase (ELO) in the hepatopancreas and muscle showed significantly higher levels in the treatment groups, whereas an opposite trend was existed in the IPF. Fatty acid synthase (FAS), sterol regulatory element binding protein‐1c (SREBP‐1c) in the IPF and hepatopancreas reached the lowest in 2.0% group. Overall, dietary ALA could promote n‐3 highly unsaturated fatty acids (HUFAs) synthesis and suppress the accumulation of lipid by decreasing the expression of related genes and promoting the apoptosis in IPF. 相似文献

Recent outbreaks of the H5N1 strain of avian influenza in Europe have highlighted the need for continuous surveillance and early detection to reduce the likelihood of a major outbreak in the commercial poultry industry. In Great Britain (gb), one possible route by which H5N1 could be introduced into domestic poultry is through migratory wild birds from Europe and Asia. Extensive monitoring data on the 24 wild bird species considered most likely to introduce the virus into GB, and analyses of local poultry populations, were used to develop a risk profile to identify the areas where H5N1 is most likely to enter and spread to commercial poultry. The results indicate that surveillance would be best focused on areas of Norfolk, Suffolk, Lancashire, Lincolnshire, south-west England and the Welsh borders, with areas of lower priority in Anglesey, south-west Wales, north-east Aberdeenshire and the Firth of Forth area of Scotland. Summary The heads of 1,489 sheep and 320 goats were examined for larvae ofOestrus ovis at 17 abattoirs in northern Libya in July to November 1988. The prevalence ofO. ovis in sheep was 22·6% and in goats it was 18·4 per cent. Up to 14 and 11 larvae were collected from individual sheep and goats respectively. All larvae were recovered from the nasal passages and frontal sinuses, but only second and third instars were seen.

We have developed an antibody detection enzyme-linked immunosorbent assay (ELISA) for the identification of animals infected by feline immunodeficiency virus (FIV). The ELISA solid-phase antigen consists of recombinant FIV gag proteins expressed in bacteria. The proteins are purified from bacterial lysates as insoluble inclusion bodies. In the case of bacterially expressed p24gag, it is shown that all of the linear, sequential epitopes presented by viral p24 during infection are retained. Purified preparations can be substituted for solid-phase whole virus in the IDEXX PetChektm immunoassay. Germinal vesicle migration (GVM) and/or dissolution (GVD) were measured in goldfish oocytes, treated with 17α, 20β dihydroxyprogesterone (DHP) and other compounds considered to effect the cytoskeleton and oxidative phosphorylation,in vitro. Administration of DHP reinitiated meiotic maturation, increasing GVM and GVD in goldfish oocytes. Addition of 2,4-dinitrophenol

An 8‐week feeding trial was conducted to determine lysine requirement of juvenile yellow catfish (Pelteobagrus fulvidraco) by feeding formulated diets containing crystalline l ‐lysine. Six isonitrogenous and isoenergetic diets (405 g kg?1 protein, 18 kJ g?1 gloss energy) containing fish meal together with soybean protein concentrate as protein sources and fish oil together with soybean oil as lipid sources were formulated. Crystalline l ‐lysine was added into the six diets to acquire lysine concentrations of 17.3, 21.8, 26.0, 31.3, 35.5 and 41.9 g kg?1 dry diets, respectively. Mixture of crystalline amino acid was supplemented to simulate the amino acid profile in muscle of yellow catfish. The results indicated that final body weight (FBW), weight gain (WG), specific growth rate (SGR), feed efficiency (FE) and protein efficiency (PE) increased with the increase in dietary lysine level from 17.3 to 31.3 g kg?1 of diet and then decreased as the dietary lysine levels further increased. No significant difference in survival rate was found among all the dietary treatments. One‐slope, quadratic broken‐line analysis on the basis of SGR showed that the dietary l ‐lysine requirement of juvenile yellow catfish was 33.1 g kg?1 of dry diet (83.2 g kg?1 of dietary protein). 相似文献

This study was conducted to assess regional patterns of diversity of Ethiopian tetraploid wheat accessions and to identify areas of diversity that can be used as source of new germplasm for developing high yielding and stable varieties. A collection of 133 Ethiopian tetraploid wheat accessions and eight introduced cultivars was analysed using 29 wheat microsatellite markers. A total of 383 alleles were detected with an average value of 13.14 alleles per locus. Relatively more alleles were observed in the B genome than in the A genome. Gene diversity indices ranged from 0.08 to 0.95, with a mean value of 0.72. Accessions collected from the same region were pooled and the number of alleles and gene diversity were calculated over the 29 simple sequence repeats for each region. Higher numbers of alleles were detected in the Shewa region (8.72), followed by Tigray (5.86) and Hararghe (5.76). The highest average gene diversity value was found in Shewa (0.65), followed by Gondar (0.64). No significant correlation was observed between geographic distance and genetic distance. Out of 383 different alleles detected, 93 (24.4%) were observed to be region‐specific. Region‐specific alleles were found across all chromosomes except for Xgwm752, Xgwm155 and Xgwm148. Genetic similarity coefficients were estimated for all the possible 55 pairs of regional comparisons and they ranged from 0.16 to 0.52, with a mean value of 0.50. All provinces were differentiated in the UPGMA cluster diagram. 相似文献
