The specific binding of Staphylococcal protein A for mammalian immunoglobulin G was used to demonstrate IgG associated with Aleutian disease virus (ADV) when isolated from infected mink tissues. Protein A specifically bound to mink serum Ig with no reaction with other serum or tissue proteins. Protein A labeled with 131Iodine reacted with crude virus preparations but not with virus that had been purified by freon extraction to the point where it became reactive with antibody by counterimmunoelectrophoresis. Binding to purified ADV was restored when the purified virus was first reacted with antibody. Results of urea treatment indicated this as an alternative method for isolation of ADV free from antibody.  相似文献

Botrytis calthae is a necrotrophic plant pathogen, closely related to the ubiquitous broad host range fungus Botrytis cinerea, but highly host specific. Botrytis isolates from lesions of Caltha palustris grown at different locations were classified with genetic markers as either B. calthae or Botrytis pseudocinerea, or less frequently as B. cinerea. A PCR‐based identification of B. calthae was developed. Seven haplotypes of B. calthae could be distinguished. Compared to B. cinerea, mycelium growth of B. calthae was similar, but conidiation less abundant, and sclerotia formation was only partially repressed by light. Conidia of B. calthae germinated more slowly, and showed a highly acidic optimum (pH 2·5) compared to B. cinerea conidia (pH 5·3). All B. calthae isolates were sensitive to common anti‐Botrytis fungicides, but showed partial resistance to the succinate dehydrogenase inhibitors boscalid, fluopyram and carboxin. Infection experiments revealed a weak capability of B. calthae to induce necrotic lesions on plants that are hosts for B. cinerea. On C. palustris leaves, B. calthae induced similar lesions to B. cinerea. These data provide a basis for comparative molecular investigation of the physiology and host specificity of B. calthae and closely related Botrytis species.  相似文献

AIM: To identify fungi isolated from infections of the bovine mammary gland, and establish their possible sources.

METHODS: From a herd of 420 cows, milk samples were collected from all quarters at calving and cultured to detect causative organisms. Quarters identified as infected with fungi were further sampled during early lactation. Samples from feedstuffs, the feed pad and ends of teats were also collected and analysed for the presence of fungi.

RESULTS: Eleven of 420 cows were diagnosed with intramammary infections (IMI) caused by yeasts (nine cows, 10 quarters) and moulds (two cows, three quarters). Six of the yeast species had previously been reported as being responsible for mastitis. Elevated somatic cell counts (SCC) were observed in many quarters, but most infections were eliminated spontaneously. Two of the fungi isolated from milk samples were also isolated from feedstuffs and teat swabs, and seven other fungi isolated from milk samples were not isolated from feed, the feed pad or cows' teats.

CONCLUSIONS: Isolation of fungi from the udder is rarely reported in dairy cows in New Zealand. In this herd, contamination of the end of the teat originating from feedstuffs and possibly exacerbated by the use of a feed pad may have led to the establishment of IMI caused by fungi.

CLINICAL RELEVENCE: Fungi are infrequently if ever reported in mastitis trial data or surveys in New Zealand and are probably of little clinical significance.  相似文献

A live attenuated auxotrophic S. typhimurium (S. tm.)-mutant was used by orally administration via drinking water several times during rearing, combined with 1- or 2-times parenteral injection of an autogenous S. enteritidis (S. e.)/S. tm.-oil emulsion vaccine. In a 8-month period, more than 500,000 birds were vaccinated. The vaccine was safe. Challenge test showed protection in the vaccinates and their offspring. The number of isolates in the farms detected by regular monitoring decreased. The protection of the live-culture mutant lasted about only 8 to 10 weeks. A. S. tm.-mutant more potent for chickens will be tested now. We consider vaccinations as one important factor in a salmonella control program, especially for commercial layers and broiler breeders.  相似文献

Grain yield and its component trait thousand kernel weight are important traits in triticale breeding programmes. Here, we used a large mapping population of 647 doubled haploid lines derived from four families to dissect the genetic architecture underlying grain yield and thousand kernel weight by multiple‐line cross QTL mapping. We identified 3 QTL for grain yield and 13 for thousand kernel weight which cross‐validated explained 5.2% and 48.2% of the genotypic variance, respectively. Both traits showed a positive phenotypic correlation, and we found two QTL overlapping between them. Full two‐dimensional epistasis scans revealed epistatic QTL for both traits, suggesting that epistatic interactions contribute to their genetic architecture. Based on QTL identified in our results, we conclude that the potential for marker‐assisted selection is limited for grain yield but more promising for thousand kernel weight.  相似文献

European tofu manufacturers are becoming more and more interested in locally grown soybeans. A prerequisite for the development of European tofu cultivars is to improve our understanding of how the environment and genetics affect soymilk and tofu quality, as well as how quality and agronomic traits are correlated. This study was based on 215 recombinant inbred lines derived from two populations and grown at three locations that were evaluated for tofu traits in a bench‐scale tofu laboratory. The heritabilities of most of the evaluated tofu traits were moderately high with h2 > 0.6. We observed significant genotypic variance components, but an even stronger contribution of the location. The network analysis of the evaluated traits was population‐dependent; however, the tofu traits were not associated with any of the agronomic traits. Collectively, our results indicate the potential to improve tofu‐related traits in European soybean. The bench‐scale tofu production method provides a valuable tool to test soybean lines in breeding programmes; however, the method needs to be further improved and automated to minimize errors due to the laboratory staff.  相似文献

Five red shikonin pigments, deoxyshikonin, shikonin, acetylshikonin, isobutylshikonin, and beta-hydroxyisovalerylshikonin, were isolated from the roots of Lithospermum erythrorhizon cultivated in Korea. The purified pigments were red, purple, and blue at acidic, neutral, and alkaline pH values, respectively. Physical stability of the purified pigments against heat and light in an aqueous solution was examined for possible value-added food colorants. The thermal degradation reactions were carried out at pH 3.0 (50 mM glycine buffer) in 50% EtOH/H(2)O. Deoxyshikonin (t(1/2) = 14.6 h, 60 degrees C) and isobutylshikinin (t(1/2) = 19.3 h, 60 degrees C) are relatively less stable than other shikonin derivatives (t(1/2) = 40-50 h, 60 degrees C). Activation energies of thermal degradation of the isolated pigments were calculated. The activation energy of deoxyshikonin was the highest (12.5 kcal mol(-)(1)) and that of beta-hydroxyisovalerylshikonin was the lowest (1.71 kcal mol(-)(1)) value. Light stabilities of the pigments were similar to each other in that the half-life values of photodegradation for 20000 lx light intensity were 4.2-5.1 h.  相似文献
