Theileria equi (T. equi) is an obligate intra- and extra-erythrocytic parasite that causes equine theileriosis (ET) in equids. Equine theileriosis is considered a notifiable disease of global significance, a major constraint to the international movement of horses, and endemic in many countries. This disease may be difficult to diagnose, as it can produce variable and nonspecific clinical signs. A cross-sectional study was designed for the molecular characterisation of T. equi and to investigate the associated risk factors of ET accompanied by its consequences on haematological and sero-biochemical parameters. A convenience sampling of 500 blood samples were collected from ET suspect horses from January to December 2017. PCR was performed on all blood samples targeting the 18S rRNA gene of T. equi followed by sequencing; 9% animals tested positive with confirmed sequences. The isolates of this study showed high homology with Cuban, Russian and Brazilian isolates of T. equi (accession numbers KY111762.2 , MG551915.1 and KY952237.1 , respectively). Based on multivariate analysis, the principal risk factors consisted of absence of dogs on the premises and presence of tick infestation. The haemato-biochemical parameters showed a decrease in granulocytes and erythrocytes, and an increase in lymphocytes, monocytes, mean corpuscular volume, mean corpuscular haemoglobin, mean platelet volume, glucose, phosphorus and aspartate aminotransferase in positive horses. This is the first study which identified ET in Punjab (Pakistan) using molecular techniques and risk factors together with the haemato-biochemical variations in horses. 相似文献

Summary The competitive ability of inoculated and indigenous Rhizobium/Bradyrhizobium spp. to nodulate and fix N2 in grain legumes (Glycine max, Vigna unguiculata, Phaseolus vulgaris) and fodder legumes (Vicia sativa, Medicago sativa, and Trifolium subterraneum) was studied in pots with two local soils collected from two different fields on the basis of cropping history. The native population was estimated by a most-probable-number plant infectivity test in growth pouches and culture tubes. The indigenous rhizobial/bradyrhizobial population ranged from 3 to 2×104 and 0 to 4.4×103 cells g-1 in the two soils (the first with, the second without a history of legume cropping). Inoculated G. max, P. vulgaris, and T. subterraneum plants had significantly more nodules with a greater nodule mass than uninoculated plants, but N2 fixation was increased only in G. max and P. vulgaris. A significant response to inoculation was observed in the grain legume P. vulgaris in the soil not previously used to grow legumes, even in the presence of higher indigenous population (>103 cells g-1 soil of Rhizobium leguminosarum bv phaseoli). No difference in yield was observed with the fodder legumes in response to inoculation, even with the indigenous Rhizobium sp. as low as <14 cells g-1 soil and although the number and weight of nodules were significantly increased by the inoculation in T. subterraneum. Overall recovery of the inoculated strains was 38–100%, as determined by a fluorescent antibody technique. In general, the inoculation increased N2 fixation only in 3 out of 12 legume species-soil combinations in the presence of an indigenous population of rhizobial/bradyrhizobial strains. 相似文献

MALDI‐TOF MS was tested for the identification of Photobacterium damselae subsp. piscicida on isolates grown on two media, cultured at three incubation times and applied on the target plate by the direct sample spotting (DS), by the on‐target extraction (OTE) and by the full extraction (FE) method, in triplicates. The identification of samples grown on blood agar (BA) outperformed identification on tryptic soya agar (TSA) by 0.64% for DS and OTE. The OTE gave the highest scores in both culture media, all incubation times and replicates. Reliable 24‐hr species identification was 61.54%, 84.61% and 53.85% for samples grown on TSA and identified by DS, OTE and FE, respectively. For isolates grown on BA, they were 76.92%, 96.15% and 30.77%, respectively. When identified by OTE, the 48‐hr identification was 93.58%, but for 72 hr declined to 71.79%. The reliable identification with the highest score from the first measurement was 100% only for OTE from BA (24 hr), whereas OTE from TSA gave 84.61% (24 hr), 76.92% (48 hr) and 84.61% (72 hr). The present study is focused on studying the swelling kinetics, thermal and aqueous stabilities, and determination of various forms of water in the chitosan (CS) and polyacrylonitrile (PAN) blend and semi-interpenetrating polymer network (sIPN). CS/PAN blend hydrogel films were prepared by solution casting technique. The blend film with optimum swelling properties was selected for the synthesis of sIPN. CS in the blend was crosslinked with the vapors of Glutaraldehyde (GTA) to prepare sIPN. The fabricated CS/PAN blend and sIPN hydrogels films were characterized with Fourier transform infrared (FTIR), thermal gravimetric analysis (TGA) and field emission scanning electron microscope (FESEM). The kinetics of swelling, bound and unbound waters and aqueous stability were determined experimentally. FESEM showed good miscibility between CS and PAN, FTIR showed no chemical interaction between CS and PAN; however, it did show a doublet for the sIPN, TGA showed improved thermal stability and swelling kinetic followed second order kinetics. The degree of swelling of the sIPN hydrogels samples at room temperature varied from ~2200 % (with a fair degree of stability (~30 %)) to ~1000 % (with high degree of aqueous stability (43 %)) with increase in the crosslinking time. The calculated unbound water (WUB) max., for the blend was 52.3 % whereas for the bound (WB) the max., was 41.9 %. However, for sIPN hydrogel films, the WUB water decreased (max. 21.0 %) where as the WB increased (max. 52.0 %). The decrease in WUB and increase in the WB is attributed to the formation of a compact structure and increase in the contact area between the water and polymers in sIPN hydrogels due to the induction of new water contacting point in these hydrogel films, respectively. 相似文献

Bread-making quality of wheat flour is largely determined by the accumulation, concentration and composition of the proteins in the grain, which are influenced by genetic (G) and environment (E) variation and their interactions. We have therefore evaluated the importance of G and E factors and their interactions in determining the accumulation and composition of the proteins in the wheat grain. The cultivar determined development time (CDDT), together with the amount and timing of N application, played a significant role in determining the accumulation and final composition of the wheat grain proteins, explaining 21–59% of the variation. At low temperature, N application both at spike formation and at anthesis explained the highest proportion of variation (36%) in the percentage of sodium dodecyl sulphate (SDS) unextractable polymers in the total amount of polymers (% UPP), while at high temperature CDDT contributed most to the variation in % UPP (20%). The largest contributor to variation in the amount of total SDS extractable proteins (TOTE) was N application at anthesis, both at low and high temperatures (12% and 36%, respectively). Thus, the climate should be considered in recommendations for improving the protein quality and thereby the bread-making quality of wheat. 相似文献

Conservation tillage may improve yield of cotton in addition to improvement in soil quality if practiced for longer period. However, the practice may not be productive in short-term particularly when severe weeds are infesting the crops such as Cynodon dactylon, Conyza canadensis, Tribulus terrestris, and Cyperus rotundus, etc. Recent studies indicate that conventional tillage (CT) is more productive than zero tillage (ZT)/reduced tillage (RT). Performance of cotton under three tillage systems, viz., ZT, RT and CT; and five herbicides, i.e., haloxyfop-R-methyl 10.8 EC (108 g a.i. ha-1), lactofen 24 EC (168 g a.i. ha-1), haloxyfop 10.8 EC + lactofen 24 EC, hand weeding, and weedy check were evaluated during 2010-2011 at Gomal University, D.I.Khan, Pakistan, to explore the best management option for effective weed control, enhanced yield and quality of cotton grown after wheat. The results revealed that hand weeding and Haloxyfop as post emergence alone or in combination with Lactofen reduced weed density to the minimum irrespective of the tillage systems. Excessive rainfall and cooler temperature limited cotton growth and yield in 2010. The adverse weather conditions had more adverse effect on boll weight under ZT and RT than CT. Haloxyfop + lactofen produced higher seed cotton yield in RT than ZT, however, it could not exceed CT. Broad-spectrum herbicides×CT produced the highest number of bolls/plant, boll weight and seed cotton yield. Fiber quality and net returns were also the highest in broad-spectrum herbicides×CT. In conclusion, broad-spectrum herbicides under CT were more productive in wheat based cropping system on silty clay soil of D.I.Khan. 相似文献

The green lacewing Chrysopa pallens (Rambur) (Neuroptera: Chrysopidae) is a common and abundant predator in many cropping systems in palearctic realm and it’s conservation is helpful in sustainable pest management in agro-ecosystem. Prior to commercialization of Bt crops in any agro- ecosystem, it is necessary to evaluate the impact of Cry proteins upon non-target organisms especially biological control agents (BCA). In present study an artificial diet consisting of shrimp, beef, beef liver and egg yolk was developed to mass-rear C. pallens for its use as biological control agents in sustainable pest management. Moreover, an artificial diet based risk assessment protocol was developed to investigate the impact of Cry1Ac, Cry1Fa and Cry2Ab on the survival and reproductive performance of C. pallens adults. C. pallens was fed on diets incorporated with Cry proteins and without addition of Cry proteins (control). The same diet containing boric acid was served as a positive control. Temporal stability, bioactivity and intake of Cry proteins by C. pallens were confirmed using double-antibody sandwich, enzyme-linked immunosorbent assay and bioactivity verification bioassays. Survival and reproductive performance of C. pallens, e.g., pre-oviposition period, daily fecundity, total fecundity and 30-day old adults dry weights, exhibited non-significant differences (p?>?0.05) for the diets containing Cry1Ac, Cry1Fa and Cry2Ab (50 μg/g) against Control. However, significant reduction in survival and reproductive performance (p?<?0.05) was observed in positive control. Our findings reveal that artificial diet is a good source of nutritional requirement with enhanced survival and reproductive performance of C. pallens and can be used for mass rearing of predator in case of natural diet scarcity and Cry proteins are safe for adult C. pallens and Bt crops cultivation help in predators conservation in sustainable agriculture. 相似文献

BACKGROUND: Rhizobacteria have a good potential to suppress soilborne diseases, but their efficacy against sugarcane pests is rarely reported. Bacterial strains isolated from the rhizosphere of sugarcane were evaluated for their potential to suppress red rot disease on two susceptible varieties, Co‐1148 and SPF‐234, under field conditions. The strains were also characterised for the production of secondary metabolites associated with their antagonistic activity. RESULTS: One out of four strains, the Pseudomonas putida strain NH‐50 (EU627168), reduced disease severity by 44–60% in different field trials. This potent antagonistic strain produced pyoluteorin antibiotic, as confirmed by high‐performance liquid chromatography (HPLC). The PltB gene involved in pyoluteorin synthesis was amplified from the P. putida strain NH‐50 and sequenced. The extracellular metabolites and volatile and diffusible antibiotics secreted by the tested strains inhibited mycelial growth of Glomerella tucumensis (Speg.) Arx & E Mull in vitro by 7–55%. CONCLUSION: The pyoluteorin‐producing bacteria P. putida strain NH‐50 significantly reduced disease severity on both sugarcane varieties, irrespective of fungal inoculation, i.e. either inoculated through stem or through soil. This strain also possesses other plant growth characteristics and can be used as a biopesticide for sugarcane crop. Copyright © 2011 Society of Chemical Industry 相似文献
