A comparative study was carried out to determine the susceptibility of five different cell lines to pseudorabies virus (PRV), a herpes virus of pigs. The cell systems tested were swine testicle (ST), mink lung (ML), equine dermal (ED), porcine kidney (PK15), and bovine turbinate (BT) cells. Virus titers obtained were 104.88, 104.38, 103.75, 102.63, and 100.25 for ML, ST, PK15, BT and ED cells, respectively indicating that ML, ST, and PK15 are optimal cell lines for the growth of PRV whereas BT and ED are not very sensitive. 相似文献

Time course changes in plasma amino acid concentrations and the hepatic metabolism of serine and alanine were measured in six mature wethers during endotoxin-induced fever. In separate trials, the animals' responses to injections of saline and endotoxin were measured. The endotoxin was from Escherichia coli serotype 055:B5 and was injected intravenously (4 micrograms/kg body weight). Liver biopsies were obtained from the sheep at 6 h postinjection during both endotoxin and saline injection trials. Rectal temperature in the endotoxin treated animals was increased (P less than 0.05, above that in control animals from 4.25 h to 9 h postinjection, with a maximum rise of 2.43 degrees C at 5.5 h postinjection. Glucose concentration in jugular plasma decreased (P less than 0.05) by 3 h postinjection and remained depressed throughout the 24 h postinjection sampling period. Plasma serine concentration was decreased (P less than 0.05) by 3 h postinjection. Plasma alanine concentration was decreased significantly (P less than 0.05) only at 24 h postinjection. Endotoxin injection increased (P less than 0.05) hepatic oxidation of 14C-serine (162%) and the net incorporation of 14C-serine carbon into hepatic protein (173%) and glycogen (275%). The net incorporation of 14C-alanine carbon into hepatic protein (172%) and glycogen (323%) were increased (P less than 0.05) by endotoxin injection, while alanine oxidation was not affected by endotoxin treatment (P greater than 0.05). The increased hepatic use of serine may explain, in part, the dramatic decrease in plasma concentrations of this amino acid following endotoxin injection into sheep. 相似文献

The purpose of this study was to investigate experimental infection of the piglet as a model of Rhodococcus equi pneumonia in the foal. Three litters of eight piglets each were exposed to an aerosol of 3.4 X 10(7) R. equi per piglet per day for seven consecutive days. Over the next 23 days the piglets were observed for clinical signs of disease. Periodically after infection one piglet from each litter was killed, the lungs were cultured quantitatively for R. equi and the gross and microscopic pulmonary lesions were assessed. The only clinical evidence of disease was the occurrence of elevated temperatures in the infected piglets. Rhodococcus equi was slowly cleared from the piglets' lungs during the 23 days following aerosolization. Piglets sacrificed seven to ten days after aerosolization had the most extensive pulmonary lesions, consisting of severe consolidation of the cranioventral lobes. Microscopic examination revealed thickened interalveolar septa and alveoli containing many neutrophils and macrophages with intracytoplasmic Gram-positive coccobacilli. The pulmonary lesions in these piglets differed from those of naturally infected foals in that they were not characterized by macrophage-rich abscesses and the infection gradually resolved. 相似文献

A purified protein derivative-live preparation of Brucella abortus strain 45/20 was tested for immunogenic properties either alone, after lipid conjugation, or in association with defined adjuvants. The adjuvants were trehalose dimycolate (cord factor), isolated from wax D of mycobacteria and murmyl dipeptide (N-acetyl-muramyl-L-alamyl-D-isoglutamine), a synthetic glycopeptide analog of peptidoglycan subunits found in many bacterial cell walls and wax D of mycobacteria. Guinea pigs were intradermally inoculated with a single injection of the vaccine preparations eight weeks before intramuscular challenge with B. abortus, strain 2308. None of the purified protein derivative-like preparations were as effective in the prevention of splenic infections with B. abortus as were killed whole cells of strain 45/20 in Freund's complete adjuvant. Whole cells in Freund's complete adjuvant were able to reduce mean colony counts by 97% (P = 0.02), while purified protein derivative-like vaccines were able to reduce mean colony counts by only 32 to 61% as compared to control animals. Results suggest that purified protein derivative-like preparations have limited immunogenic properties under present test conditions. 相似文献

An eight month survey of bovine eyes from an abattoir in southern Ontario revealed the presence of the eyeworms Thelazia gulosa and T. lacrymalis in 32% of the cattle. To evaluate the efficacy of a Haemophilus somnus bacterin a total of 1114 beef calves or yearling bulls were used in the province of Saskatchewan. The six herds were included in a vaccination trial in which the vaccine was administered subcutaneously in one or two doses. The prevalence of H. somnus in nasal swabs of these animals at the time of the initial vaccination was 0.35% Postvaccination information on morbidity and mortality for a four month period was requested from the five ranches where there were nonvaccinated control calves. Postvaccination outbreaks of infectious thromboembolic meningoencephalitis occurred in two of these herds and, although the numbers were limited, there was a trend to reduced morbidity and mortality in the vaccinated animals compared to controls. Seroconversion rates, as determined by the complement fixation test, in the six herds were 28.3% for control calves, 57% for animals vaccinated once and 80.3% for animals vaccinated twice. On the basis of these results the bacterin was considered to be sufficiently efficacious to warrant its further evaluation under field conditions. 相似文献

为寻求减少草坪修剪次数以节约人力、物力的可行措施,本研究采用乙烯利和多效唑两种生长延缓剂对盆播早熟禾进行分别处理,并同时设修剪试验进行对照研究。结果是:在本试验所采取的剂量范围内,乙烯利使盆播早熟禾叶片枯死脱落,再生叶片窄小挺立,节间伸长和整株早衰;多效唑具有延缓盆播早熟禾生长的功能,且随着用量加大,延缓生长作用增强,多效唑还有使叶片变窄变小、绿色加深和更加鲜柔的作用。在本试验所选择的留茬高度和修剪频率条件下,修剪促进盆播早熟禾的生长,并抑制其分蘖和草皮层发育。与修剪试验对比,多效唑处理确可减少修剪次数。 相似文献

This paper describes a simple methodology for evaluating the bacterial binding of ciprofloxacin labelled with technetium Tc 99m. Using this methodology, the binding of 99mTc-ciprofloxacin by live Escherichia coli was compared with the binding of 99mTc-ciprofloxacin by killed E. coli and the binding of 99mTc-pertechnetate (99mTcO4-) by live E. coli. The antimicrobial effect of 99mTc-ciprofloxacin on E. coli was evaluated. Four groups were defined: live E. coli with 99mTc-ciprofloxacin, live E. coli with 99mTcO4 , killed E. coli with 99mTc-ciprofloxacin, and killed E. coli with 99mTcO4-. After 0, 2, and 4 h of incubation of 1 x 10(8) colony-forming units of E. coli suspended in 5 mL of sterile distilled water with 1.85 MBq of 99mTc-ciprofloxacin or 99mTcO4, 1 mL from each sample was centrifuged. The radioactivity of the bacterial pellet and that of the supernatant were measured separately, and the percentage of sample radioactivity attributable to bacterial binding was calculated. Of the 99mTc-ciprofloxacin, 3.6% to 5.9% was bound to live or killed E. coli; only 0.1% to 0.2% of the 99mTcO4- was bound to live E. coli (P < 0.0001). No significant difference in 99mTc-ciprofloxacin binding was found between live and killed E. coli (P = 0.887). An antimicrobial effect on E. coli was seen with 99mTc-ciprofloxacin: colony counts were reduced after 4 h. The small amount of 99mTc-ciprofloxacin binding and the lack of difference in binding between live and killed E. coli may limit the utility of this methodology in evaluating the presence of E. coli infection. 相似文献

Leishmaniases are a globally widespread group of parasitic diseases from the group of anthropozoonoses. They are caused by an intracellular protozoan parasite that causes a wide spectrum of diseases in humans and dogs worldwide. In fact, the dog is considered one of the most important reservoir. The spleen is one of the several haematopoietic and immunocompetent organs involved. As this organ is a blood filter, the authors looked for the expression of this infection over the microvascular environment and modifications of the spleen cell population related to immunological responses to this parasitic condition.

Results: Our results show three important modifications concerning the microvascular architecture of the spleen when compared to the normal pattern. Remarks: According to our results it seems that independent of the virulence of the parasite involved and the kind of immunity prevalent in a particular host, the spleen develops blood dynamic conditions that allows a sluggish blood flow so that cells involved in immunological processes can proliferate and differentiate and it also contributes to the trapping of lymphocytes to the area through the differentiation of typical characteristics of HEV endothelial cells. 相似文献

Perineuronal nets (PNs) of condensed extracellular matrix (ECM) have been shown to characterize the microenvironment of individual neurons and the chemoarchitecture of some brain regions. In the present study, PNs in the hippocampus were visualized with a cationic iron colloid method for sulphated proteoglycan content and a plant lectin from Vicia villosa agglutinin (VVA) for N-acetylgalactosamine containing glycoconjugates. The ECM molecules were organized in reticular coats (PNs) around non-pyramidal cells in the Ammons horn (Corneu Ammons, CA) and subicular region, in addition to pyramidal neurons located in CA2 and CA3 regions. CA2 stratum pyramidale exhibited the most intense staining of its PNs and a diffuse intervening neutrophil labelling, while CA3 region showed a graded fashion of staining intensity. The subiculum displayed intensely stained perineuronal coats. Notably, the hippocampal perineuronal nets revealed overlapped staining characteristics with both staining methods. However, cell coats of the subicular neurons showed various degrees of labelling characteristics with both markers. It is suggested that the PNs in the hippocampus are correlated with the fast spiking inhibitory GABAergic neurons and their target pyramidal cells, and this is important to keep the excitatory elements under control and therefore control the information processes within the hippocampus. 相似文献
