Tumor DNA structure in plant cells transformed by A. tumefaciens

Crown gall tumors are induced in plants by infection with the soil bacterium Agrobacterium tumefaciens. Because the tumor induction involves transfer of a portion of the tumor-inducing (Ti) plasmid DNA from the bacterium to the plant cells, this system is of interest for the study of genetic exchange as well as tumor induction. The boundaries of the transferred DNA (T-DNA) have been cloned from transformed plant cells of tobacco. Detailed mapping with restriction enzymes and nucleotide sequence analysis of two independent clones were used to study the molecular structure of the ends of the T-DNA. One clone contains the two ends of the T-DNA joined together; the other contains one end of the T-DNA joined to repetitive plant DNA sequences. These studies provide direct evidence that the T-DNA can be integrated into the plant genome. In addition, the data suggest that in the plant, T-DNA can be tandemly repeated. Sequence analysis of the junction of crown gall clone 1 reveals several direct repeats as well as an inverted repeat; these structures may be involved in the transfer of the DNA from Agrobacterium to plant cells.     

Mossbauer investigation of shocked and unshocked iron meteorites and fayalite

M?ssbauer spectra of several iron meteorites have been measured by a resonant scattering technique rather than by the conventional transmission method, thereby eliminating the necessity for the preparation of thin samples. No significant differences were observed in the spectra of specimens of mechanically deformed, shocked, and unshocked iron meteorites, nor in the absorption spectra of artificially shocked and unshocked fayalite.     

Functional links between Aβ toxicity, endocytic trafficking, and Alzheimer's disease risk factors in yeast

Aβ (beta-amyloid peptide) is an important contributor to Alzheimer's disease (AD). We modeled Aβ toxicity in yeast by directing the peptide to the secretory pathway. A genome-wide screen for toxicity modifiers identified the yeast homolog of phosphatidylinositol binding clathrin assembly protein (PICALM) and other endocytic factors connected to AD whose relationship to Aβ was previously unknown. The factors identified in yeast modified Aβ toxicity in glutamatergic neurons of Caenorhabditis elegans and in primary rat cortical neurons. In yeast, Aβ impaired the endocytic trafficking of a plasma membrane receptor, which was ameliorated by endocytic pathway factors identified in the yeast screen. Thus, links between Aβ, endocytosis, and human AD risk factors can be ascertained with yeast as a model system.     

5-Lipoxygenase expression in benign and malignant canine prostate tissues

5-Lipoxygenase (5-LO) is overexpressed in human prostate carcinomas (PCs), and its inhibition decreases proliferation and induces apoptosis in prostate cancer cell lines. We hypothesized that 5-LO would be overexpressed in canine PC compared with benign prostate tissue and may be important in the pathogenesis of the disease. Immunoblot analysis of canine PC and benign prostatic hyperplasia (BPH) tissues demonstrated 5-LO expression in both. 5-LO immunohistochemical staining was not significantly different within the stromal or epithelial components of canine primary PC, BPH or suppurative prostatitis, suggesting that differential expression of this enzyme does not occur in these conditions. The percentage of tumour cells expressing 5-LO was significantly lower in metastatic PC lesions compared with primary PC (P < 0.0001). This decreased expression may indicate down-regulation or altered expression of the enzyme with progression of canine PC to a metastatic phenotype.     

Nine simultaneous primary tumors in a Boxer dog.

An aged Boxer dog had 9 distinct primary tumors: chemodectoma, osteosarcoma, bronchiolo-alveolar adenocarcinoma, interstitial cell tumor, seminoma, basal cell tumor, fibropapilloma, adrenal cortical adenoma, and pancreatic adenoma. From this report, as well as from other studies, it is clear that Boxers have special susceptibilities to a variety of tumors. Analysis of clinical data on canine tumors indicated that the risk of Boxers for multiple tumors is only slightly higher than all tumors, indicating little or no specific predisposition for multiple tumors. In Boxers, however, certain tumor types occurred more frequently as multiple primary tumors than would be expected by chance.
