This study was conducted to evaluate the potential of graded levels of GroBiotic®‐A to improve performance of Nile tilapia, Oreochromis niloticus, fed a 29% crude protein (CP) diet. A 29% CP diet was formulated and supplemented with 0, 0.4, 0.8, and 1.2% GroBiotic®‐A and compared to performance of fish fed a 33% CP diet. Enhanced weight gain and feed efficiency were generally observed in fish fed the diets supplemented with GroBiotic®‐A compared to the 29% CP diet. No significant differences in these responses were observed between fish fed diets supplemented with GroBiotic®‐A compared to those fed the 33% CP diet. Supplementation of 0.8 and 1.2% GroBiotic®‐A induced significantly lower condition factor and hepatosomatic index compared to fish fed the 29% CP diet, but those values were similar to that of fish fed the 33% CP diet. GroBiotic®‐A supplementation and protein reduction had no effect on the viscerosomatic index of fish or moisture, lipid, and protein content of muscle samples. However, muscle ash increased significantly with protein reduction (29% CP diet), but GroBiotic®‐A supplementation (0.8 and 1.2%) reduced muscle ash content. Activities of catalase and superoxide dismutase were markedly reduced in fish fed GroBiotic®‐A (0.8 and 1.2%) compared to those fed the control diet. GroBiotic®‐A supplementation also induced significantly higher neutrophil oxidative radical production compared to fish fed the 29% CP diet, but no significant difference was observed in comparison with the 33% CP diet. After 8 wk of feeding, exposure to Aeromonas hydrophila for 3 wk resulted in 40% (0.4, 0.8% GroBiotic®‐A) and 27% (1.2% GroBiotic®‐A) mortality and reduced signs of disease, while 47% mortality was observed in fish fed the 29% CP diet. Based on the result of this study, it is concluded that 0.8 and 1.2% GroBiotic®‐A positively influenced growth performance and feed efficiency of tilapia fed diets containing 29% crude protein to levels comparable to fish fed the 33% CP diet. GroBiotic®‐A supplementation also significantly increased neutrophil oxidative radical production as well as resistance to Ae. hydrophila infection. 相似文献

Intestinal microorganisms play important roles in maintaining host health, but their functions in aquatic animal hosts have yet to be fully elucidated. The Chinese mitten crab, Eriocheir sinensis, is one such example. We attempted to identify the shift of gut microbiota that occurred in response to infection of white spot syndrome virus (WSSV), an emerging viral pathogen in the crab aquaculture industry. The microbiota may exert some control over aspects of the viral pathogenesis. We investigated the changes in composition and structure of the crab gut microbiome during various WSSV infection stages of 6 h post‐infection (hpi) and 48 hpi, using a 16S rRNA approach on the MiSeq Illumina sequencing platform. Four phyla (Firmicutes, Proteobacteria, Tenericutes and Bacteroidetes) were most dominant in the gut of E. sinensis regardless of the WSSV infection stages. However, further analysis revealed that over 12 bacterial phyla, 44 orders and 68 families were significantly different in abundance at various states of WSSV infection. Several intriguing aspects of E. sinensis gut bacteria that had not been previously reported were also uncovered, such as class Mollicutes was dominant here, but absent in crabs from Yangtze River estuary and Chongming Islands. Overall, this study provided the first evidence that changes in gut microbiome were closely associated with the severity of WSSV infection and that indicator taxa could be used to evaluate the crab health status. 相似文献

The Lower Mekong Basin is facing unprecedented threats to fish diversity from hydropower development. There is increasing pressure on developers and construction authorities to design solutions to improve fish survival through turbines, thus protecting the resources in regions being developed for hydropower. A hydraulic characteristic of hydropower turbines with known fisheries impacts is fluid shear stress. Elevated shear stress occurs where rapidly flowing water passes near spillways, screens and within turbine draft tubes. Shear stress can have adverse impacts on fish, but no work has assessed whether this holds true for Mekong River species. A flume was used to determine critical tolerances of silver shark, Balantiocheilos melanopterus (Bleeker), to shear stress rates at a high‐velocity jet which simulated a hydropower turbine. Fish were assessed for injury or mortality following exposure. Results were compared against a no‐shear control. Injury and mortality were greater at higher shear stress exposures. Injuries occurred at all shear exposure levels with mortality at shear levels higher than 600/s. This approach should help design future hydropower turbines if data on other species demonstrate similar results. If the likelihood of adverse impact is high, then shear stress will need to be considered in the design of future hydropower facilities. 相似文献

Granular cold water gelling (GCWG) starch was obtained by treatment of native chickpea starch with liquid ammonia at low temperature and atmospheric pressure. A free‐flowing powder of the gelatinized starch granules was produced by adding ethanol to the mixture of starch and liquid ammonia. Four ratios of liquid ammonia to starch (A/S) (2:1, 4:1, 6:1 and 8:1) and four ratios of starch ethanol (S/E) (1:1, 1:2, 1:3 and 1:4) were tested for production of GCWG starch. Homogenous mixtures of liquid ammonia and starch granules appeared when the A/S ratio was >4:1. Treating starch with a 4:1:3 ammonia‐starch‐ethanol (A/S/E) ratio on a w/w/v basis resulted in a free‐flowing powder of gelatinized starch, that formed a gel on addition of water at 23°C. Differential scanning calorimetry (DSC) thermograms of the GCWG starch, regardless of A/S/E ratio, displayed enthalpy values of 2.2–3.1 J/g compared with 15.5 J/g for native starch, indicating disappearance of crystallinity due to modification. 相似文献

The highly variable environmental conditions across the Pacific Northwest (PNW) influence the milling and baking quality of wheat grain produced in this region. This study was conducted to compare the flour composition, dough rheology, and baking quality of soft and hard spring wheat grain produced in diverse environments. Thirteen soft and five hard spring wheat cultivars were grown at Lind, WA (semiarid) and Fairfield, WA (high precipitation) for three years. Grain was evaluated for flour composition, rheology, and experimental baked product quality. Flour composition, rheological properties, and baking qualities were primarily influenced by the environment. Protein contents, microSDS values, and water absorption levels were significantly (P < 0.0001) higher for all cultivars grown at Lind compared with those from Fairfield. Cookie diameters were larger (P < 0.0001) for soft flours from Fairfield, whereas loaf volumes were higher (P < 0.0001) for hard wheat flours from Lind. Results indicate that producing soft or hard wheat outside of its optimal climatic zone reduces experimental baked product quality. 相似文献

Milled, long-grain rice was exposed to air at temperatures (T) of 20, 30, and 40°C, and relative humidities (RH) ranging from 25 to 85%. The kernels then were subjected to a breakage test to determine the extent of damage that occurred during each exposure condition. Increasing air T levels produced higher amounts of broken kernels across the RH range. Milled rice at higher moisture content (MC) levels sustained more extensive stress crack damage at low RH conditions and less stress crack damage at high RH conditions relative to milled rice at lower MC levels. Amylose contents of prime starches from nonwaxy and high-amylose barley, determined by colorimetric method, were 24.6 and 48.7%, respectively, whereas waxy starch contained only a trace (0.04%) of amylose. There was little difference in isoamylase-debranched amylopectin between nonwaxy and high-amylose barley, whereas amylopectin from waxy barley had a significantly higher percentage of fraction with degree of polymerization < 15 (45%). The X-ray diffraction pattern of waxy starch differed from nonwaxy and high-amylose starches. Waxy starch had sharper peaks at 0.58, 0.51, 0.49, and 0.38 nm than nonwaxy and high-amylose starches. The d-spacing at 0.44 nm, characterizing the amylose-lipids complex, was most evident for high-amylose starch and was not observed in waxy starch. Differential scanning calorimetry (DSC) thermograms of prime starch from nonwaxy and high-amylose barley exhibited two prominent transition peaks: the first was >60°C and corresponded to starch gelatinization; the second was >100°C and corresponded to the amylose-lipid complex. Starch from waxy barley had only one endothermic gelatinization peak of amylopectin with an enthalpy value of 16.0 J/g. The retrogradation of gelatinized starch of three types of barley stored at 4°C showed that amylopectin recrystallization rates of nonwaxy and high-amylose barley were comparable when recrystallization enthalpy was calculated based on the percentage of amylopectin. No amylopectin recrystallization peak was observed in waxy barley. Storage time had a strong influence on recrystallization of amylopectin. The enthalpy value for nonwaxy barley increased from 1.93 J/g after 24 hr of storage to 3.74 J/g after 120 hr. When gel was rescanned every 24 hr, a significant decrease in enthalpy was recorded. A highly statistically significant correlation (r = 0.991) between DSC values of retrograded starch of nonwaxy barley and gel hardness was obtained. The correlation between starch enthalpy value and gel hardness of starch concentrate indicates that gel texture is due mainly to its starch structure and functionality. The relationship between the properties of starch and starch concentrate may favor the application of barley starch concentrate without the necessity of using the wet fractionation process. 相似文献

The sex ratio of the feral Southern catfish was reported to be about 1:1, while the fish obtained by artificial fertilization
were always female. Hence, we examined the possible influence of the micro-environment during artificial insemination (pH
of the ovarian fluid and concentration of the semen) and early development (feed, hatching temperature, and water) on the
sex ratio of Southern catfish fry. In order to examine the possibility of the occurrence of gynogenesis during artificial
propagation, cytological observations on the insemination processes and the artificial induction of gynogenesis were also
performed. However, no male fish were obtained even in these experiments, excluding the possibilities of these micro-environmental
changes on catfish sex ratio and the occurrence of gynogenesis during artificial propagation. Female-to-male sex reversal
was achieved by treatment with fadrozole (an aromatase inhibitor) and tamoxifen (an estrogen receptor antagonist). Histological
analyses on the gonadal development of both female and induced male fish were subsequently performed. Moreover, several genes
involved in sex differentiation, such as dmrt1, foxl2, and cyp19, and three subunits of gonadotropin (gth), i.e., gthα, lhβ, and fshβ, were isolated. Their expression patterns were studied under normal gonadal development and sex reversal conditions. The
results revealed that dmrt1, foxl2, and cyp19a were closely related to catfish sex differentiation, and the gth subunits were possibly related to ovarian differentiation and oocyte development. Taken together, we hypothesized that estrogen
was highly responsible for the ovarian differentiation and feminization of catfish fry under artificial propagation, although

A new marine fish cell line, TK, derived from turbot (Scophthalmus maximus) kidney, was established by the method of trypsin digestion and subcultured for more than 50 passages over a period of 300 days.
The TK cells were maintained in Minimum Essential Medium Eagle (MEM) supplemented with HEPES, antibiotics, fetal bovine serum
(FBS), 2-Mercaptoethanol (2-Me), and basic fibroblast growth factor (bFGF). The suitable growth temperature for TK cells was
24°C, and microscopically, TK cells were composed of fibroblast-like cells. Chromosome analysis revealed that the TK cell
line has a normal diploid karyotype with 2n = 44. Two fish viruses LCDV-C (lymphocystis disease virus from China) and TRBIV (turbot reddish body iridovirus) were used
to determine the virus susceptibility of TK cell line. The TK cell line was found to be susceptible to TRBIV, and the infection
was confirmed by cytopathic effect (CPE) and transmission electron microscopy, which detected the viral particles in the cytoplasm
of virus-infected cells. Finally, significant green fluorescent signals were observed when the TK cells were transfected with
pEGFP-N3 vector, indicating its potential utility for fish virus study and genetic manipulation. 相似文献
