The effect of porcine reproductive and respiratory syndrome virus (PRRSV) on semen quality was examined in a group of 11 spontaneously infected boars in a commercial boar stud. Semen samples were collected 4 weeks prior to 4 weeks post-infection (wpi). Infection with PRRSV of the European genotype subtype 1 (EU-1) was verified by specific quantitative real-time polymerase chain reaction (RT-PCR) in 36% of the serum samples. All boars seroconverted before 4 wpi and remained in normal condition throughout the study. Comparison of the percentage of morphologically intact spermatozoa revealed an increase of acrosome-defective spermatozoa (P = 0.012) between −4 and 4 wpi. Significant deleterious effects on semen quality were detected for membrane integrity when semen had been stored for 2 days after sampling. Analysis of sperm subpopulations in a thermoresistance test on day 7 after sampling revealed alterations in the percentage of circular, progressively motile spermatozoa (P = 0.013), in the percentage of non-linear, progressively motile spermatozoa (P = 0.01), and on the amplitude of lateral sperm head displacement (P = 0.047). There was no difference in the incidence of mitochondrially active spermatozoa (P = 0.075). Investigation of routine production data between pre- and post-infection status showed no differences on ejaculate volume (P = 0.417), sperm concentration (P = 0.788), and percentage of motile spermatozoa (P = 0.321). A short-chain 15N-peptide mixture characterized by an average chain length of 2.3 was obtained when 15N-labeled yeast protein has hydrolyzed enzymatically by thermitase from Thermoactinomyces vulgaris. Fifteen newborn Wistar-rats were given a single pulse of [15N]glycine. [15N]H4Cl and [15N]yeast protein-thermitasehydrolysate (YPTH) in a dosage of 50 mg 15N excess kg-1 by gastric tube. In comparison with [15N]glycine the 15N-incorporation rates of brain, muscle and liver were approximately 150% higher after [15N]YPTH-application. Uniform labeling, high 15N-enrichment, almost complete absorption, avoidance of imbalances and the low price make this tracer substance superior to other tracers conventionally used for organ labeling. 相似文献

Due to their marked antiinflammatory effect, synthetic corticosteroids are used to mask illness, especially lameness in horses. The detection of these drugs in equine body fluids requires accurate methods, particularly where misuse of corticosteroids is suspected. Gas chromatography/mass spectrometry (GC/MS) is well established as a reliable technique for the identification of drugs in biological fluids. Using GC/MS, we determined dexamethasone levels in horse urine and serum after intravenous application of a therapeutic dose. Dexamethasone was detectable, in serum for up to six hours, and in urine for up to 32 hours, after its administration. These findings indicate that serum measurements are unreliable for the detection of corticosteroid abuse, and demonstrate urine to be a more suitable body fluid for investigation. Nevertheless, it should be emphasized that, regardless of the technique employed, the clinical effects of dexamethasone last longer than 32 hours; thus, failure to detect dexamethasone does not disprove corticosteroid abuse. 相似文献

OBJECTIVE: To develop a radiographic stress technique to quantify hip joint laxity in dogs. STUDY DESIGN: Prospective study on client-owned dogs presented for hip dysplasia screening. ANIMAL POPULATION: 302 nonselected dogs (63 breeds). METHODS: Dogs were sedated and placed in dorsal recumbency. During pelvic radiography, the femoral heads were displaced manually in a craniodorsal direction. On these stress radiographs, the degree of lateral displacement of the femoral heads was assessed in terms of a subluxation index (SI) and compared with the degree of femoral head subluxation and the severity of hip dysplasia found on conventional extended hindlimb radiographs. RESULTS: The degree of subluxation on stress radiographs was significantly greater than on conventional radiographs. Correlation between the severity of canine hip dysplasia (CHD), graded according to conventional techniques, and the degree of subluxation, induced by radiographic stress technique, was positive (r = .57; P<.0001). Two critical SI values were noted. Of the dogs with an SI value of 0.3 or less, 99% were classified as CHD grade normal, borderline, or mildly dysplastic. In dogs with an SI value between 0.3 and 0.5, CHD grading ranged from normal to severely dysplastic. Of the dogs with an SI value greater than 0.5, 95% had dysplastic hip joints. CONCLUSIONS: Hip joint laxity cannot be quantified precisely on standard position radiographs. Carbon and hydrogen stable isotope ratios of carotenoids and beta-carotene-based dietary supplements

Considering the increasing nutritional and commercial importance of carotenoids, there is an interest in developing a reliable method for authenticity assessment of these compounds. Applying isotope ratio mass spectrometry using elemental analysis in the "combustion" (C) and "pyrolysis" (P) modes (EA-C/P-IRMS), the delta (13)C V-PDB and delta (2)H V-SMOW values of selected carotenoids and alpha/beta-carotene-based commercial dietary supplements were determined in comparison to those of synthetic and "natural" references. The delta (13)C V-PDB and delta (2)H V-SMOW values of synthetic beta-carotene samples ( n = 4), ranging from -25.3 per thousand to -26.4 per thousand and from -144 per thousand to -155 per thousand, respectively, differed clearly from the data determined for carotenoids from various natural sources, including C 3 plant material ( n = 9; delta (13)C V-PDB ranging from -28.5 per thousand to -32.8 per thousand and delta (2)H V-SMOW from -180 per thousand to -275 per thousand) and microalgae Dunaliella salina ( n = 1; delta (13)C V-PDB value of -15.6 per thousand and delta (2)H V-SMOW value of -191 per thousand). From five commercial dietary supplements under study, two revealed delta (13)C V-PDB and delta (2)H V-SMOW values in areas as found for synthetic references, and the other three had values near those of biotechnological beta-carotene produced by D. salina. The delta (13)C V-PDB and delta (2)H V-SMOW values recorded for natural lycopene ( n = 4) and lutein ( n = 5) ranged from -31.1 per thousand to -31.8 per thousand and from -180 to -201 per thousand, as well as from -28.8 per thousand to -32.2 per thousand and from -186 per thousand to -245 per thousand, respectively. Synthetic canthaxanthin ( n = 3) exhibited delta (13)C V-PDB and delta (2)H V-SMOW values ranging from -25.0 per thousand to -28.6 per thousand and from -133 per thousand to -153 per thousand, respectively. The EA-C/P-IRMS application of this study showed that the natural stable isotopic composition of carotenoids is a powerful tool for determining their origin. 相似文献

The objective of this investigation was to provide a comprehensive analysis of the total lipid composition of present-day retail beef meat available at the consumer level and to evaluate the total lipid composition with special emphasis on the nutritional value. For this purpose, 40 beef cuts were obtained from four cattle farms based on either a natural grazing system (NGS) or an intensive production system (IPS). The total lipid composition was analyzed using complementary chemical and chromatographic procedures. The content of n-3 LC-PUFA, CLA, total trans-18:1, and branched-chain fatty acids was significantly higher in NGS beef than in IPS beef. The trans-18:1 and CLA profiles were affected by the different production systems, whereby they can be utilized empirically to differentiate between feeding regimen and production management. Fatty acid ratios that have health implications ( n-6/ n-3, LA/alphaLNA, and AA/EPA) were remarkably beneficial for NGS beef compared with IPS beef. In conclusion, from the human health perspective, beef raised on NGS is clearly superior with regard to a more favorable fatty acid profile in comparison to IPS beef. 相似文献

Two fatty acids, (2E,4Z)-decadienoic acid and (2E,4Z,7Z)-decatrienoic acid, the latter being described for the first time as a natural product, were detected in the culture filtrate of Streptomyces viridochromogenes Tü 6105 by HPLC-diode array screening, purified by chromatographic methods and structurally elucidated by NMR techniques. Both metabolites show strong herbicidal activity against Lemna minor and Lepidium sativum.
