The coccidian parasite Neospora caninum is an intracellular protozoan, causing abortion in cattle in many countries around the world. In this study, the protective potential of the major N. caninum surface antigen NcSRS2, expressed in Escherichia coli and formulated into immunostimulating complexes (iscoms), was investigated in an experimental mouse model. The recombinant protein was specially designed for binding to iscoms via biotin-streptavidin interaction. Two groups of 10 BALB/c mice were immunised twice, on days 0 and 28 with iscoms containing either the recombinant NcSRS2 (NcSRS2 iscoms) or similar iscoms with NcSRS2 substituted by an unrelated recombinant malaria peptide (M5) as a control (M5 iscoms). A third group of 10 age-matched BALB/c mice served as an uninfected control group. Immunisation with recombinant NcSRS2 iscoms resulted in production of substantial antibody titres against N. caninum antigen, while the mice immunised with M5 iscoms produced only very low levels of antibodies reacting with N. caninum antigen. After challenge infection with N. caninum tachyzoites on day 69, mice immunised with NcSRS2 iscoms showed only mild and transient symptoms, whereas the group immunised with M5 iscoms showed clinical symptoms until the end of the experiment at 31 days post inoculation. A competitive PCR assay detecting Nc5-repeats was applied to evaluate the level of parasite DNA in the brain. The amount of Nc5-repeats in the group vaccinated with NcSRS2 iscoms was significantly lower than in the control group given M5 iscoms. In conclusion, it was found that the recombinant NcSRS2 iscoms induced specific antibodies to native NcSRS2 and immunity sufficient to reduce the proliferation of N. caninum in the brains of immunised mice.  相似文献

This study evaluated the site-specific applicability and efficacy of the GPFARM decision support system (DSS) based on underlying simulation model performance for dry mass grain yield, crop residue, total soil profile water content, and total soil profile residual NO3-N across a landscape catena for dryland no-till experimental locations in eastern Colorado. Relative error of simulated mean, normalized objective function (root mean square error divided by the observed mean), and index of agreement evaluation statistics were calculated to compare modeled results to observed data. A one-way, fixed-effect ANOVA was also performed to determine differences among experimental locations and summit, sideslope, and toeslope landscape positions. GPFARM simulations matched observed data trends, with the model correctly distinguishing variations between the summit and toeslope landscape positions. In addition, experimental observations and GPFARM simulations both indicated that the toeslope landscape position was the most productive for grain yield and also exhibited higher amounts of crop residue, total soil profile water content, and total soil residual NO3-N. The GPFARM crop model performed adequately but was inconsistent in simulating winter wheat, corn, and sorghum dry mass grain yield. GPFARM performance in simulating crop residue was poorer than for crop grain yield. GPFARM predicted mean total soil profile water content was generally within ±20% of the observed mean across locations and landscape positions, with the model somewhat biased towards overpredicting total soil profile water content at the summit and sideslope landscape positions. Total soil profile residual NO3-N was underpredicted by GPFARM across all locations and landscape positions by an average of 30%. Although GPFARM appears to have reasonably simulated long-term output responses across a landscape catena for the eastern Colorado experimental locations (especially given the simplifying assumptions in many of the GPFARM simulation components and the inherent variability present at the experimental plot level), different interpretations of GPFARM performance can be made depending on the evaluation statistic of interest. Furthermore, the model cannot fully account for water and chemical movement across the landscape catena; simulation results suggest that addition of a spatially-distributed routing component should offer improvements in GPFARM prediction accuracy across a catena where surface runoff or lateral subsurface flow is occurring.  相似文献

Steroidal glycoalkaloids occur in potatoes and are reported to impart resistance to phytopathogens including bacteria, fungi, and insects. Because glycoalkaloids can be passed to progenies during breeding programs designed to develop improved potatoes, it is of importance to determine the quality of desired characteristics and the composition of glycoalkaloids of new somatic hybrids. The objective of this study was to determine the appearance, size, and shape (morphological characters) as well as the glycoalkaloid content of potato tubers of somatic hybrids between tetraploid Solanum tuberosum cv. Dejima (2n = 4x = 48 chromosomes) and the dihaploid clone ATDH-1 (2n = 2x = 24 chromosomes) induced by anther culture from Solanum acuale-T (acl-T, 2n = 4x = 48 chromosomes). Tuber size and shape in somatic hybrids were in accord with those of cv. Dejima, whereas the tuber skin color resembled that of ATDH-1. Thin-layer chromatography, high-performance liquid chromatography, and gas-liquid chromatography/mass spectrometry studies showed that the two steroidal glycoalkaloids (alpha-chaconine and alpha-solanine) were present in the tubers of S. tuberosum, whereas acl-T and ATDH-1 tubers were found to contain alpha-tomatine and demissine. The concentrations of total glycoalkaloids in both acl-T and ATDH-1 was >100 mg/100 g of fresh weight tuber cortex, much higher than in S. tuberosum. All somatic hybrids, except one clone, contained four glycoalkaloids (alpha-chaconine, alpha-solanine, alpha-tomatine, and demissine) derived from the fusion parents. The lack of alpha-tomatine in the remaining clone may be due to somaclonal variation. The results show that character expression is influenced by ploidy level and that total glycoalkaloid levels in most somatic hybrids were intermediate between those of the fusion parents. The possible significance of these findings for plant breeding and food safety is discussed.  相似文献

The bioactivity-guided purification of the culture broth of the shipworm endosymbiont Teredinibacter turnerae strain 991H.S.0a.06 yielded a new fatty acid, turneroic acid (1), and two previously described oxylipins (2–3). Turneroic acid (1) is an 18-carbon fatty acid decorated by a hydroxy group and an epoxide ring. Compounds 1–3 inhibited bacterial biofilm formation in Staphylococcus epidermidis, while only 3 showed antimicrobial activity against planktonic S. epidermidis. Comparison of the bioactivity of 1–3 with structurally related compounds indicated the importance of the epoxide moiety for selective and potent biofilm inhibition.  相似文献

A 15-year-old, pregnant Dutch Warmblood mare was exhibiting intermittent hemorrhage secondary to a large vestibulovaginal varicosity. The vessel was reduced with transendoscopic Nd:YAG laser photocoagulation. This technique is minimally invasive compared with traditional surgical ligation, and significantly decreases the risk of severe hemorrhage, especially with advancing pregnancy. One laser treatment resulted in complete reduction in vessel size and allowed the pregnancy to continue to term without incident or further vaginal hemorrhage.  相似文献

1. Chick embryo cells and halved embryos were successfully implanted into unfertilised eggs. Yolks containing implants were placed in recipient eggshells, covered by transparent vacuum-formed plastic cones and incubated for 72 h. 2. Dispersed cells were obtained from eggs expelled from the uterus or from eggs that had been laid. Implantation of these cells often resulted in aggregation and epithelial growth, in several cases with axial development. 3. Growth of implanted halved embryos of different ages was often observed, including one 10-somite embryo. Non-axial epithelia, sometimes with a central hole, a central fluid-filled cellular vesicle or a vesicle only, were also observed. 4. In another culture system, whole and halved embryos obtained from laid eggs were cultured on a vitelline membrane stretched across semi-solid egg albumen. During the 72 h incubation, axial development was observed only in whole embryos, while halved embryos grew either into epithelia containing fluid-filled cellular vesicles or into vesicles only. 5. It was found that daily drainage of the accumulating fluid from the embryo compartment encouraged axial development in halved embryos, and almost abolished vesicle formation. Holes were formed in half the embryos cultured on a vitelline membrane. 6. It appeared that physical and biological conditions could inflict serious malformations on the implants.  相似文献
