The causes and control of type 2 diabetes mellitus are not clear, but there is strong evidence that dietary factors are involved in its regulation and prevention. We have shown that extracts from cinnamon enhance the activity of insulin. The objective of this study was to isolate and characterize insulin-enhancing complexes from cinnamon that may be involved in the alleviation or possible prevention and control of glucose intolerance and diabetes. Water-soluble polyphenol polymers from cinnamon that increase insulin-dependent in vitro glucose metabolism roughly 20-fold and display antioxidant activity were isolated and characterized by nuclear magnetic resonance and mass spectroscopy. The polymers were composed of monomeric units with a molecular mass of 288. Two trimers with a molecular mass of 864 and a tetramer with a mass of 1152 were isolated. Their protonated molecular masses indicated that they are A type doubly linked procyanidin oligomers of the catechins and/or epicatechins. These polyphenolic polymers found in cinnamon may function as antioxidants, potentiate insulin action, and may be beneficial in the control of glucose intolerance and diabetes.  相似文献

Chamber measurements of total ecosystem respiration (TER) in a native Canadian grassland ecosystem were made during two study years with different precipitation. The growing season (April–September) precipitation during 2001 was less than one-half of the 30-year mean (1971–2000), while 2002 received almost double the normal growing season precipitation. As a consequence soil moisture remained higher in 2002 than 2001 during most of the growing season and peak aboveground biomass production (253.9 g m−2) in 2002 was 60% higher than in 2001. Maximum respiration rates were approximately 9 μmol m−2 s−1 in 2002 while only approximately 5 μmol m−2 s−1 in 2001. Large diurnal variation in TER, which occurred during times of peak biomass and adequate soil moisture, was primarily controlled by changes in temperature. The temperature sensitivity coefficient (Q10) for ecosystem respiration was on average 1.83 ± 0.08, and it declined in association with reductions in soil moisture. Approximately 94% of the seasonal and interannual variation in R10 (standardized rate of respiration at 10 °C) data was explained by the interaction of changes in soil moisture and aboveground biomass, which suggested that plant aboveground biomass was good proxy for accounting for variations in both autotrophic and heterotrophic capacity for respiration. Soil moisture was the dominant environmental factor that controlled seasonal and interannual variation in TER in this grassland, when variation in temperature was held constant. We compared respiration rates measured with chambers and that determined from nighttime eddy covariance (EC) measurements. Respiration rates measured by both techniques showed very similar seasonal patterns of variation in both years. When TER was integrated over the entire growing season period, the chamber method produced slightly higher values than the EC method by approximately 4.5% and 13.6% during 2001 and 2002, respectively, much less than the estimated uncertainty for both measurement techniques. The two methods for calculating respiration had only minor effects on the seasonal-integrated estimates of net ecosystem CO2 exchange and ecosystem gross photosynthesis.  相似文献

We have identified a small-molecule inhibitor of tumor necrosis factor alpha (TNF-alpha) that promotes subunit disassembly of this trimeric cytokine family member. The compound inhibits TNF-alpha activity in biochemical and cell-based assays with median inhibitory concentrations of 22 and 4.6 micromolar, respectively. Formation of an intermediate complex between the compound and the intact trimer results in a 600-fold accelerated subunit dissociation rate that leads to trimer dissociation. A structure solved by x-ray crystallography reveals that a single compound molecule displaces a subunit of the trimer to form a complex with a dimer of TNF-alpha subunits.  相似文献

An endoscopic sterilisation technique for use in Galapagos tortoises (Geochelone nigra) was developed as part of a conservation and ecosystem restoration project. Fifteen female giant Galapagos tortoises were anaesthetised, intubated and positioned in dorsal recumbency. A bilateral prefemoral approach was made and the ovaries were identified using a 5 mm × 33 cm rigid telescope. In the case of endoscope-assisted oophorectomy, the ovaries were exteriorised through the same incision, the vasculature was ligated and the mesovarium was transected. Two tortoises had immature ovaries that could not be exteriorised. In these animals, endoscopic oophorectomy was performed using radiosurgery. Closure of the incisions was routine. All tortoises except one recovered well from surgery. There were no reported complications six weeks and six months postoperatively, and all were successfully released on to Pinta Island in May 2010.  相似文献

Stallion spermatozoa recovered and examined immediately after colloidal centrifugation resulted in a higher straight‐line velocity (VSL) than sperm processed using direct conventional centrifugation (p = 0.000), but there was no differences in the progressive motility or sperm DNA fragmentation (SDF) as determined by the sperm chromatin dispersion assay. However, when centrifuged spermatozoa were incubated at 37°C for 24 h to determine the rate of SDF (r‐SDF), a lower r‐SDF (p = 0.0011) was observed in those sperm recovered after colloidal separation (0.5 ± 0.1%/h) compared to direct (1.2 ± 0.4%/h) or no centrifugation (r‐SDF = 1.2 ± 0.3%/h). These results confirm that colloidal separation of stallion spermatozoa results in prolonged sperm DNA longevity, but these differences were only apparent following a period of incubation and dynamic assessment. Consequently, we strongly recommend the use of the dynamic form of the SDF assay for evaluating centrifugation and/or other ex vivo procedures, as a single basal assessment of SDF may inadvertently result in a false‐positive evaluation of DNA quality.  相似文献

Of 1,235 individual birds from 130 species tested for haemagglutinating virus and/or NDV antibody in far northern Queensland, none gave a positive response. On available evidence pittas and rainforest pigeons are considered the species most likely to bring virulent NDV into Australia followed by gulls and night herons which move between dense seabird breeding colonies and other avian communities. Both can easily be monitored by strategic sampling along migratory pathways or at breeding islands. Wild parrots, waterfowl and migratory waders appear to present a minimal threat.  相似文献

We compared vertical gradients in leaf gas exchange, CO(2) concentrations, and refixation of respired CO(2) in stands of Populus tremuloides Michx., Pinus banksiana Lamb. and Picea mariana (Mill.) B.S.P. at the northern and southern boundaries of the central Canadian boreal forest. Midsummer gas exchange rates in Populus tremuloides were over twice those of the two conifer species, and Pinus banksiana rates were greater than Picea mariana rates. Gas exchange differences among the species were attributed to variation in leaf nitrogen concentration. Despite these differences, ratios of intercellular CO(2) to ambient CO(2) (c(i)/c(a)) were similar among species, indicating a common balance between photosynthesis and stomatal conductance in boreal trees. At night, CO(2) concentrations were high and vertically stratified within the canopy, with maximum concentrations near the soil surface. Daytime CO(2) gradients were reduced and concentrations throughout the canopy were similar to the CO(2) concentration in the well-mixed atmosphere above the canopy space. Photosynthesis had a diurnal pattern opposite to the CO(2) profile, with the highest rates of photosynthesis occurring when CO(2) concentrations and gradients were lowest. After accounting for this diurnal interaction, we determined that photosynthesizing leaves in the understory experienced greater daily CO(2) concentrations than leaves at the top of the canopy. These elevated CO(2) concentrations were the result of plant and soil respiration. We estimated that understory leaves in the Picea mariana and Pinus banksiana stands gained approximately 5 to 6% of their carbon from respired CO(2).  相似文献
