More than 25 atomic percent of aluminum can susbtitute for other cations in the structure of apatite [A(10)(XO(4)) (6)Z(2)]. Such a synthetic product was obtained by expelling volatile constituents (H(2)O and F) from morinite during thermal treatment. Infrared absorption spectra, chemical analysis, and x-ray powder diffraction demonstrate that the aluminum has two coordination numbers, and more than twice as much aluminum substitutes for calcium (A position) as for phosphorus (X position). 相似文献

Strains of Phytophthora megasperma insensitive to the fungicide metalaxyl showed limited cross-resistance to the structurally related fungicide cyprofuram and to metolachlor, a herbicide with antifungal properties. The three compounds each inhibited nucleic acid synthesis, but the wide variation in resistance factors and in the degree of nucleic acid inhibition at the concentrations inhibiting mycelial growth by 50% indicated that cyprofuram and metolachlor possess alternative mechanisms of toxicity. Studies with protoplasts suggested that cyprofuram has a direct effect on the plasma membrane which may be important against those resistant strains where nucleic acid synthesis is least affected. Metolachlor also was extremely effective in causing protoplast lysis. Sporangia production by P. palmivora was stimulated by low concentrations of cyprofuram, and germination was delayed. As with other ‘acylalanine-type’ fungicides, inhibition of nucleic acid synthesis is considered to be the main mode of action. Some degree of cross-resistance between cyprofuram and other acylalanines is likely to occur in the field but the dual mode of action of cyprofuram might delay its onset. Horizontal resistance (HR) was assessed as a strategy to reduce potato defoliation by the Colorado potato beetle (CPB). Hybrids amongSolanum tuberosum varieties grown in Iowa were first evaluated with a detached leaflet feeding bioassay. Beginning in 1998, resistance was increased annually over three years by intermating and selecting the most resistant l%–2% of each generation of 300–400 plants using the bioassay. When C (= mg leaf tissue consumed* neonate larva-1 over 48 h) was measured for the 1% most-resistant genotypes of all generations grown side by side in 2000, it ranged from 6.1 mg* larva-1 in the parents to 2.3 for progeny of the F3 generation compared to 2.7 in ND2858-1, a resistant check. Field trials in Wisconsin showed less defoliation in the F1 and F2 selected genotypes than in the parents. A second breeding program using moreS. tuberosum parents with greater genetic diversity, and much larger breeding populations, resulted in significantly faster accumulation of resistance, further suggesting that recurrent mass selection is effective. Foliar glycoalkaloid analyses revealed low levels of solanine and chaconine in the F3 selections; no leptines or leptinines were detected. Because the parents wereS. tuberosum varieties in which vertical resistance to CPB is unknown, and because the resistance to feeding from parents to the F3 resistant plants was linear and gradual while ranging from initially very low to considerably higher in the third year, horizontal resistance is strongly implicated. The development of a significant degree of resistance after three rounds of breeding using relatively small breeding populations suggests that HR can be developed from varieties ofSolanum tuberosum lacking initial high levels of resistance to CPB. 相似文献

Accumulation of misfolded protein in the endoplasmic reticulum (ER) triggers an adaptive stress response-termed the unfolded protein response (UPR)-mediated by the ER transmembrane protein kinase and endoribonuclease inositol-requiring enzyme-1alpha (IRE1alpha). We investigated UPR signaling events in mice in the absence of the proapoptotic BCL-2 family members BAX and BAK [double knockout (DKO)]. DKO mice responded abnormally to tunicamycin-induced ER stress in the liver, with extensive tissue damage and decreased expression of the IRE1 substrate X-box-binding protein 1 and its target genes. ER-stressed DKO cells showed deficient IRE1alpha signaling. BAX and BAK formed a protein complex with the cytosolic domain of IRE1alpha that was essential for IRE1alpha activation. Thus, BAX and BAK function at the ER membrane to activate IRE1alpha signaling and to provide a physical link between members of the core apoptotic pathway and the UPR. 相似文献

Sixteen rams were used to quantify the effects of long days, imposed during late winter/early spring, with or without exogenous melatonin, on plasma testosterone concentrations and ram serving capacity. Rams were assigned to two groups: photoperiod‐treated rams (Artificial Photoperiod, AP; n = 8), exposed to 2 months of long days (16 hr of light/day) between 22 December and 22 February, and control rams (Natural Photoperiod, NP; n = 8). At the end of the long‐day period, AP rams were returned to the natural photoperiod, and each ram in the two groups either did (+M) or did not (‐M) receive three subcutaneous melatonin implants. Four groups were created as follows: AP+M (n = 4), AP‐M (n = 4), NP+M (n = 4) and NP‐M (n = 4). Thirty days after of the onset of photoperiodic treatment, AP rams (13.5 ± 2.8 ng/ml) had significantly (p < .05) lower testosterone levels than NP rams (36.7 ± 1.0), and similar differences were not apparent at the end of the photoperiod treatment. A month later, AP rams (24.3 ± 7.9) had higher (p < .10) testosterone levels than NP rams (13.1 ± 5.0), with no effect of melatonin treatment. Fifty days after melatonin implantations, rams were exposed for 20 min to three oestrous ewes. AP rams (2.50 ± 0.42) exhibited significantly (p < .05) more serves than did NP rams (1.11 ± 0.39), and melatonin treatment had no significant effect; however, the interaction between treatments was significant. Time to first serve was significantly (p < .05) shorter in AP (2.30 ± 1.20 min) than it was in NP rams (5.58 ± 0.68 min). In conclusion, exposure to 2 months of long days in late winter/early spring, with a return to natural day length of shorter duration, increased plasma testosterone concentrations and sexual performance in rams with or without exogenous melatonin. This particular management is an option if a non‐hormonal reproductive strategy is scheduled; yet, if the use of exogenous hormones is feasible, melatonin implants increase the mating efficiency of rams. 相似文献

Single layer centrifugation (SLC) through a colloid is a tool for selecting viable mammalian spermatozoa but has not been used previously for fresh dromedary camel sperm. Semen from six camels (2 ejaculates/male) was diluted 1:5 (v:v) or 1:10 (v:v) in a Tris–citrate–fructose buffer for mechanical liquefaction by gentle pipetting. Following liquefaction, semen was processed either by SLC or by centrifugation without a colloid (control). Total and progressive motilities, CASA kinematics, vitality and acrosome integrity (eosin–nigrosin) and plasma membrane integrity (Hypo‐osmotic swelling test; HOST), and fertilizing ability in a heterologous assay (zona‐free goat oocytes) were evaluated. Both total (p = .003) and progressive motilities (p = .003) were higher in SLC‐processed than in control semen samples, irrespective of dilution. Positive HOST values increased when using colloid in 1:5 (p = .001) and 1:10 dilution (p = .010). Colloid‐selected sperm had higher penetration rates than controls (p < .001 and p = .02 for 1:5 and 1:10 dilutions, respectively). However, only the SLC sperm at 1:5 dilution showed higher percentages of pronuclear formation (p = .02) than controls. Dilution effect was only significant for total motility before in vitro fertilization, with higher values for the 1:5 dilution (p = .033). The recovery rates of motile sperm between dilutions were similar (26.1% vs 35.4%; p = .226). In conclusion, SLC is a promising tool for selecting functional dromedary camel sperm and warrants more research. 相似文献

【目的】对中国6个区域奶牛子宫内膜炎病原菌的种类和区域分布特征进行研究,为筛选出最佳治疗药物以减少子宫内膜炎对畜牧业造成经济损失提供数据支持。【方法】分别从赤峰区域、张家口区域、潍坊区域、重庆区域、宝鸡区域和北京区域选择不同规模的奶牛养殖场中患子宫内膜炎的奶牛,用采样器每个区域采集60份(6个区域共计360份)患病牛的子宫内容物,通过革兰氏染色和生化鉴定的方法鉴定分离纯化的奶牛子宫内膜炎病原菌,并分析病原菌的分布特征。【结果】潍坊、赤峰、北京区域引起奶牛子宫内膜炎以单一感染为主。宝鸡、张家口、重庆区域引起奶牛子宫内膜炎以混合感染为主。赤峰、重庆、张家口、潍坊、宝鸡、北京区域引起奶牛子宫内膜炎的病原菌以革兰氏阳性菌为主。赤峰奶牛子宫内膜炎的主要致病菌是葡萄球菌。重庆奶牛子宫内膜炎的主要致病菌是链球菌。张家口奶牛子宫内膜炎的主要致病菌是大肠杆菌。潍坊奶牛子宫内膜炎的主要致病菌是链球菌。宝鸡奶牛子宫内膜炎的主要致病菌是葡萄球菌。北京奶牛子宫内膜炎的主要致病菌是蜡样芽孢杆菌。 相似文献

Effective species management often requires understanding patterns of movement and habitat use. A common approach in identifying where individuals reside relies upon chemical tracers from the environment that are incorporated into an individual's tissues. For fish, isotopes in their otoliths, specifically the portion of their otolith formed during their larval stage, have been used to identify the natal origin. Complicating this work, however, is the fact that during this life stage, there is a shift in the source of isotopes deposited onto the growing otolith from maternally to environmentally derived. The objective of this study was to identify the portion of the otolith representing this transition to environmentally derived isotopes so as to accurately investigate questions of natal origin for a threatened population of fall Chinook salmon (Oncorhynchus tshawytscha). We exposed developing larvae to four treatments that differed in terms of their water strontium isotope ratio (87Sr/86Sr) and used change-point analysis of otolith 87Sr/86Sr and strontium to calcium ratio (Sr/Ca) to identify the otolith radius corresponding to the transition to environmentally derived isotopes. Our results indicated this transition occurred, on average, at 132 μm (87Sr/86Sr; ±50 μm standard deviation) and 127 μm (Sr/Ca; ±29 μm) from the otolith core, which corresponded to the developmental time between hatching and exogenous feeding. A substantial proportion of our otoliths (i.e., 61%) did not show convergence between otolith and water 87Sr/86Sr by the end of the 113-day experiment, which was likely due to the dietary contribution of marine-based feed. Therefore, we were unable to recommend an otolith radius to target for the purposes of reconstructing natal origin apart from being beyond approximately 130 μm. 相似文献
