OBJECTIVE: To determine whether healthy dogs given high doses of methylprednisolone sodium succinate (MPSS) develop gastrointestinal tract ulcers and hemorrhage. ANIMALS: 19 healthy male hound-type dogs. PROCEDURE: Dogs were assigned randomly to intravenously receive high doses of MPSS (30 mg/kg of body weight, initially, then 15 mg/kg 2 and 6 hours later, and, subsequently, every 6 hours for a total of 48 hours; n = 10) or an equal volume of saline (0.9% NaCl) solution (9). Gastroduodenoscopy was performed before and after treatment. Endoscopic evidence of gross hemorrhage in the cardia, fundus, antrum, and duodenum of each dog was graded from none (0) to severe (3), and a total stomach score was calculated as the sum of the regional gastric scores. Number of ulcers were recorded. The pH of gastric fluid and evidence of occult gastric and fecal blood were measured. Food retention was recorded. RESULTS: Gastric hemorrhage was evident in all dogs after MPSS administration and was severe in 9 of 10 dogs but not visible in any dog after saline treatment. Occult gastric blood was detected more commonly (9/10 vs 2/9), median gastric acidity was greater (pH 1 vs pH 3), and food was retained more commonly (7/10 vs 1/9) in the stomach of MPSS-treated dogs. CONCLUSIONS AND CLINICAL RELEVANCE: High doses of MPSS cause gastric hemorrhage in dogs. All dogs treated with high doses of MPSS should be treated with mucosal protectants or antacids to prevent gastric hemorrhage.  相似文献

The hemolytic activity of 22 Trichomonas gallinae isolates was investigated using an 18 h erythrocyte hemolysis assay which has been shown to correlate with the clinical virulence of T. vaginalis. Absorbance of the assay supernatants was measured at 540 nm and expressed as percentage of complete hemolysis. Mean hemolytic activity of the T. gallinae isolates ranged from 3.5% to 53.4% and did not correspond with clinical virulence. The results of this investigation suggest hemolytic activity is not a useful in vitro virulence assay for T. gallinae.  相似文献

Low‐molecular‐weight organic substances (LMWOS) such as amino acids, sugars and carboxylates, are rapidly turned over in soil. Despite their importance, it remains unknown how the competition between microbial uptake and sorption to the soil matrix affects the LMWOS turnover in soil solution. This study describes the dynamics of LMWOS fluxes (10 µm ) in various pools (dissolved, sorbed, decomposed to CO2 and incorporated into microbial biomass) and also assesses the LMWOS distribution in these pools over a very wide concentration range (0.01–1000 µm ). Representatives of each LMWOS group (glucose for sugars, alanine for amino acids, acetate for carboxylates), uniformly 14C‐labelled, were added to sterilized or non‐sterilized soil and analysed in different pools between 1 minute and 5.6 hours after addition. LMWOS were almost completely taken up by microorganisms within the first 30 minutes. Surprisingly, microbial uptake was much faster than the physicochemical sorption (estimated in sterilized soil), which needed 60 minutes to reach quasi‐equilibrium for alanine and about 400 minutes for glucose. Only acetate sorption was instantaneous. At a concentration of 100 µm , microbial decomposition after 4.5 hours was greater for alanine (76.7 ± 1.1%) than for acetate (55.2 ± 0.9%) or glucose (28.5 ± 1.5%). In contrast, incorporation into microbial biomass was greater for glucose (59.8 ± 1.2%) than for acetate (23.4 ± 5.9%) or alanine (5.2 ± 2.8%). Between 10 and 500 µm , the pathways of the three LMWOS changed: at 500 µm , alanine and acetate were less mineralized and more was incorporated into microbial biomass than at 10 µm , while glucose incorporation decreased. Despite the fact that the LMWOS concentrations in soil solution were important for competition between sorption and microbial uptake, their fate in soil is mainly determined by microbial uptake and further microbial transformations. For these substances, which represent the three main groups of LMWOS in soil, the microbial uptake out‐competes sorption.  相似文献

BACKGROUND: Late watergrass [Echinochloa phyllopogon (Stapf.) Koss.] is a major weed of Californian rice that has evolved P450-mediated metabolic resistance to multiple herbicides. Resistant (R) populations are also poorly controlled by the recently introduced herbicide clomazone. The authors assessed whether this cross-resistance was also P450 mediated, and whether R plants also had reduced sensitivity to photooxidation. Understanding mechanism(s) of resistance facilitates the design of herbicide management strategies to delay resistance evolution.RESULTS Ratios (R/S) of R to susceptible (S) GR(50) were near 2.0. [(14)C]Clomazone uptake was similar in R and S plants. Clomazone and its metabolite 5-ketoclomazone reduced chlorophyll and carotenoids in S more than in R plants. The P450 inhibitors disulfoton and 1-aminobenzo-triazole (ABT) safened clomazone in R and S plants. Disulfoton safened 5-ketoclomazone only in S plants, while ABT synergized 5-ketoclomazone mostly against S plants. Paraquat was more toxic in S than in R plants.CONCLUSION: Cross-resistance to clomazone explains failures to control R plants in rice fields, and safening by P450 inhibitors suggests that oxidative activation of clomazone is needed for toxicity to E. phyllopogon. Clomazone resistance requires mitigation of 5-ketoclomazone toxicity, but P450 detoxification may not significantly confer resistance, as P450 inhibitors poorly synergized 5-ketoclopmazone in R plants. Camelina (Camelina sativa L.) was introduced for the first time in Chile in 2008 as a potential feedstock for biodiesel and also as high omega-3-containing seed oil for the salmon feed industry. The objective of this study was to determine the optimum seeding date to maximize camelina seed yield in South Central Chile. The experiment was conducted under dryland conditions in Chillán, El Carmen, Los Angeles, Gorbea, and Osorno in 2008 and in Chillán and Los Angeles in 2009. The experimental design was a RCB with a split-plot arrangement with four replicates, where the main plot was the seeding date (five dates) and the sub-plot the spring type cultivars (Gold of Pleasure, Suneson, and Blaine Creek). Seeding dates at each environment were targeted to 30 April, 15 May, 30 May, 30 June, and 30 July of 2008 and 2009. The combined analysis of variance indicated no cultivar main effect or interactions with cultivar. The date by environment interaction was significant for seed yield. There were no seed yield differences among seeding dates in Chillán and El Carmen. The first three seeding dates were the highest yielding in Los Angeles and Osorno, and only the first seeding date was significantly higher in seed yield in Gorbea. Highest seed yield at Los Angeles, Gorbea, and Osorno were 1995, 1310, and 2314 kg ha−1. Seed oil content was not different among seeding dates in Chillán and El Carmen. Highest seed oil content occurred at Osorno and Gorbea on the first three seeding dates and ranged from 420 to 457 g kg−1. Spring camelina cultivars are well adapted to South Central Chile as a winter annual crop when seeded before 15 May except in Osorno when seeded before 30 May. No disease or insectpests were observed except for Aster Yellow infestation at Chillán and Los Angeles. Camelina has great potential as an oilseed for biodiesel and as a source of omega-3 for the pharmaceutical industry but markets for it need to be developed locally for camelina to become an option for growers.  相似文献

In conjunction with urbanization and its importance as a major driver of land-use change, increased efforts have been placed on understanding urban forests and the provisioning of ecosystem services. However, very little research has been conducted on private property and little is known about the structure and function of privately owned urban forests. This research examines the structure of and carbon storage services provided by private residential urban forests in a moderate-sized Midwestern city. The primary research questions are as follows: What is the structure of private urban forests, and how does it vary across parcels? How much carbon is stored in tree and soil pools of private urban forests, and how does carbon vary across parcels? Ecological inventories were conducted on 100 residential parcels within 14 Neighborhood and Homeowners Associations of varying size and development age. Tree species richness, diversity, density, and diameter distribution were determined on a per parcel basis and for the entire tree population sampled. Further, tree and soil carbon storage were determined for each parcel. Results of this research demonstrated large variability in per-parcel tree metrics. Twelve of the parcels sampled had two or fewer trees, while eleven had greater than 50 trees. Further, tree carbon storage ranged from no carbon to 11.22 kg C m?2. Alternatively, soil carbon storage was less variable and averaged 4.7 kg C m?2, approximately 1.9 times higher than the average carbon stored in trees (2.5 kg C m?2). Management efforts aimed at maintaining or enhancing carbon storage and other ecosystem services should focus on both soil protection and maximizing services in living biomass. Our results demonstrate that sustaining tree-produced ecosystem services requires maintenance of large old trees and species diversity, not only in terms of relative abundance, but also relative dominance, and in combination, species–specific size distributions.  相似文献

Partial sequencing of the γ-gliadin gene of 62 spelt and 14 soft wheat cultivars was performed. Fifty-six of the 62 spelt cultivars and 13 of the 14 soft wheat cultivars were shown to exhibit the typical spelt or soft wheat γ-gliadin sequence, respectively. Exceptions were ascribed to crossbreeding of soft wheat and spelt. Using the typical soft wheat γ-gliadin sequence, two alternative DNA-based analytical methods were developed for the detection and quantification of spelt flour "adulteration" with soft wheat. A simple and fast detection of soft wheat in spelt flours could be achieved by restriction fragment length (RFLP) analysis. In combination with lab-on-a-chip capillary gel electrophoresis (LOC-CE) the soft wheat proportion could be estimated. Heteroduplex formation served as additional confirmation for the presence of spelt besides soft wheat. Hence, RFLP-LOC-CE constitutes a perfect analysis tool for the quality control of cereal seeds and pure cultivars. A precise quantification of soft wheat "adulterations" in spelt flour down to 1% could be achieved by the developed real-time PCR method. The calibration parameters of the real-time PCR assay fulfilled the minimum performance requirements of the European Network of GMO (genetically modified organisms) Laboratories (ENGL).  相似文献
