Background
Mg chelatase is a multi-subunit enzyme that catalyses the first committed step of chlorophyll biosynthesis. Studies in higher plants and algae indicate that the Mg chelatase reaction product, Mg-protoporphyrin IX plays an essential role in nuclear-plastid interactions. A number of Mg chelatase mutants have been isolated from higher plants, including semi-dominant alleles of ChlI, the gene encoding the I subunit of the enzyme. To investigate the function of higher plant CHLI, bacterial orthologues have been engineered to carry analogous amino acid substitutions to the higher plant mutations and the phenotypes examined through in vitro characterization of heterologously produced proteins. Here, we demonstrate the utility of a transient expression system in Nicotiana benthamiana for rapidly assaying mutant variants of the maize CHLI protein in vivo. 相似文献172.
Two experiments were conducted to evaluate the impacts on forage use and beef cattle performance of incorporating divergent wheat milling by-products in a 30% CP supplement. The by-products were wheat bran (high fiber) and second clears (high starch). The by-products were added as 1) 100% wheat bran; 2) 67% wheat bran, 33% second clears; or 3) 33% wheat bran, 67% second clears to constitute approximately 47 to 49% of the supplement. In Exp. 1, 90 Hereford x Angus cows (BW = 554 kg) grazing winter, tallgrass-prairie range were fed the supplement treatments (2.27 kg/cow daily) from early December 1997 until calving (average calving date = 3/11/98). Cumulative BW and condition changes from trial initiation through calving were not significantly different among treatments. Similarly, significant treatment effects on cow pregnancy rates as well as calf birth weights, ADG, and ending weights were not evident. In Exp. 2, 16 ruminally fistulated Hereford x Angus steers (BW = 484 kg) were blocked by weight and assigned to one of the same three supplement treatments or to a negative control (forage only). Steers had ad libitum access to tallgrass-prairie hay (76.4% NDF, 3.1% CP) and were fed supplement at the same rate (relative to BW) as the cows in Exp. 1. Forage OM, NDF, and digestible OM intakes were lower (P < 0.01) for the negative control than for supplemented steers but were not significantly different among the supplemented steers. Digestion of OM was lower (P = 0.03) for the negative control than for supplemented steers, although significant treatment differences were not evident among the supplemented groups. Digestion of NDF was not affected (P = 0.49) by treatment. Within the context of the amount of supplemental protein offered, changes in the combination of wheat milling by-products in the supplement did not affect cow performance or intake and digestion of low-quality forage. 相似文献

Surgical removal is the treatment of choice for subcutaneous (SC), intermuscular (InterM), and intramuscular (IntraM) mast cell tumors (MCTs). Advanced imaging (CT or MRI) is frequently used for presurgical planning, but InterM and IntraM MCTs can be difficult to identify and delineate on CT. Aims of the current retrospective, diagnostic accuracy, observer agreement study were to describe the imaging features of SC, InterM, and IntraM MCTs on CT and to assess the limitation of CT to identify the full local extent of the MCT. Inclusion criteria for the study were dogs with a cytologically or histologically diagnosed MCTs determined to be SC, InterM, or IntraM MCT based on histology and/or a CT scan performed in the gross disease setting. Two board-certified veterinary radiologists reviewed the CT images and recorded location, contrast enhancement pattern, and delineation between the normal and abnormal tissue. Sensitivity and specificity of CT for determining location (SC/InterM versus IntraM) was 85.71% and 55.56%, respectively, when compared to consensus location based on surgical pathology report/CT/MRI review. There was a low inter-rater agreement for delineation (kappa: 0.150 (−0.070 to 0.370) and measurement had a low/moderate correlation (rho: 0.4667 to 0.5792). Upon review by a surgical oncologist, CT findings were deemed insufficient for curative surgical planning in 13 of 16 due to inadequate definition of tumor depth, compartment boundary (fascial plane) or MCT margins. The use of CT for presurgical planning of SC/InterM/IntraM MCT dogs has limitations, especially when differentiating MCT from the adjacent muscle. 相似文献

Observations made from the Viking I orbiter show very little water vapor in the Mars atmosphere in the southern hemisphere (0 to 3 precipitable micrometers) with a gradual increase across the equator to northern latitudes. Maximum amounts between 20 and 30 micrometers have been observed in the short period covered by the observations to date. The season, northern midsummer, corresponds to the beginning of the water vapor cycle in that hemisphere. A strong repetitive diurnal cycling between the solid and vapor phases is observed at a site to the east of the Tharsis Ridge at 10 degrees north latitude; the vapor lies close to the martian surface and is most probably in saturation equilibrium with a surface haze or fog throughout much of the day. 相似文献

A wafer-scale graphene circuit was demonstrated in which all circuit components, including graphene field-effect transistor and inductors, were monolithically integrated on a single silicon carbide wafer. The integrated circuit operates as a broadband radio-frequency mixer at frequencies up to 10 gigahertz. These graphene circuits exhibit outstanding thermal stability with little reduction in performance (less than 1 decibel) between 300 and 400 kelvin. Cellular checkpoints prevent mitosis in the presence of stalled replication forks. Whether checkpoints also ensure the completion of DNA replication before mitosis is unknown. Here, we show that in yeast smc5-smc6 mutants, which are related to cohesin and condensin, replication is delayed, most significantly at natural replication-impeding loci like the ribosomal DNA gene cluster. In the absence of Smc5-Smc6, chromosome nondisjunction occurs as a consequence of mitotic entry with unfinished replication despite intact checkpoint responses. Eliminating processes that obstruct replication fork progression restores the temporal uncoupling between replication and segregation in smc5-smc6 mutants. We propose that the completion of replication is not under the surveillance of known checkpoints. 相似文献
