Ozone (O3) toxicity is a potential yield‐limiting factor for soybean (Glycine max L. Merr.) in the United States and worldwide. The most economical solution to the problem is to use O3‐tolerant cultivars. Thirty‐four cultivars and 87 near‐isogenic lines (NILS) of soybean were screened for O3 tolerance in a fumigation chamber (250 ppb for three hrs). Most tolerant cultivars tested were ‘Cloud’, ‘T‐276’, ‘T263’, and ‘Kindu’. Moderately tolerant cultivars included ‘Davis’, ‘T‐210’, and ‘Elton’. Most sensitive cultivars were ‘Corsoy 79’, ‘Noir’, and ‘Midwest’. The original ‘Clark’ cultivar was not tested, but ‘Clark 63’ tended to be more tolerant than ‘Harosoy’. The aluminum (Al)‐tolerant ‘Perry’ cultivar also tended toward greater O3 tolerance than the Al‐sensitive ‘Chief’, as observed earlier. Our rankings of ‘Hark’ as moderately sensitive and ‘Davis’ as moderately tolerant are also in agreement with earlier reports. Among NILS, the order of O3 tolerance was generally Williams>Clark>Harosoy, but differences were also observed within these parental groups. For example, L68–560 was more tolerant than some other NILS of ‘Harosoy’. ‘L76–1988’ appeared more tolerant to O3 than other NILS of ‘Williams’, but all ‘Williams’ NILS were more tolerant than most NILS of ‘Harosoy’ and ‘Clark’. Ozone‐tolerant and ‐sensitive soybean cultivars or NILS identified in our study may be useful tools in studies on mechanisms of 03 tolerance and differential 03 tolerances in plants and in the development of ameliorative measures.  相似文献

Ozone toxicity can reduce soybean yields by an estimated 5 to 20%. The most economical and practical solution to the problem would be to use ozone‐tolerant cultivars. Because ozone toxicity is an oxidative stress, one would expect that plants having higher concentrations and proper kinds of antioxidants would be more tolerant. To test this hypothesis, 20 soybean lines or cultivars varying in contents of flavonol glycosides (naturally occurring antioxidants) were tested for ozone tolerance in a fumigation chamber and in phyto‐toxic concentrations of ozone in ambient, unfiltered, air in a greenhouse. In general, ozone tolerance was associated with the presence of kaempferol glycosides, particularly K3 through K6 and K9. Lines containing no kaempferol glycosides (OX942 & OX281) were among the most sensitive to ozone stress. The K9 compound has been associated with reduced numbers of stomata, drought tolerance, and slower growth rates, and this could reduce ozone absorption and toxicity, or K9 may be more effective in detoxifying ozone than other glycosides. Lee soybean was more tolerant to ozone and to manganese (Mn) toxicity (also an oxidative stress) than Forrest. Cross tolerances of plant genotypes to these two stresses deserve additional study. Among cultivars compared, Mukden was ozone tolerant, Columbia and Harosoy were sensitive, and Blackhawk was moderately sensitive.  相似文献

Murray cod, Maccullochella peeli, originally captured from the wild, underwent normal gonadal development in earthen ponds. Handling of broodfish in the 3 months before a breeding season caused atresia and resorption of oocytes in most females. Cod were removed from the ponds when the water temperature reached 20°C during spring, and final oocyte maturation and ovulation were induced in mature females by injecting 1000 or 2000 IU/kg human chorionic gonadotrophin (HCG) or 2–5 mg/kg of a preparation of the pituitary gland from common carp (CPG). Control treatments and dosages of 100–750 IU/kg HCG did not induce ovulation. Broodfish were held at 21 ± 1°C in 2000-l tanks after injection. The time of stripping and fertilization of Murray cod eggs was an important factor determining their hatchability. There was generally high post-fertilization mortality of eggs stripped within 1 h or between 4 and 6 h of ovulation, but high hatchability of eggs stripped 2–3 h after ovulation. The mean hatchability of eggs stripped 48.5–49.5 h after the injection of 1000 IU/kg HCG was 79.8%, but there were significantly lower mean hatchabilities of eggs stripped after 46–48 h and 50–52 h, as well as after the injection of 2000 IU/kg HCG. Results using CPG were variable. Possible reasons for the high post-fertilization mortality of Murray cod eggs are discussed, and techniques for broodfish handling, injection, stripping and the fertilization and incubation of eggs are presented.  相似文献

Infestations of parasitic monogenean trematodes (Lepidotrema bidyana and Gyrodactylus sp.) on freshwater silver perch (Bidyanus bidyanus Mitchell) in earthen ponds were treated with formalin (37% formaldehyde). Concentrations of 30 and 40 mg L?1 formalin were effective, but fish in ponds treated with 20 or 25 mg L?1 remained infested. At temperatures of 24.1–26.9°C, concentrations of 30 or 40 mg L?1 formalin caused dissolved oxygen (DO) to decline from 10.1–11.9 to 3.0–3.3 and 1.2–1.7 mg L?1, respectively, within 36–42 h of treatment. In addition, pH declined from 7.2–8.4 to 6.3–6.7, within 36 h and turbidity decreased over 48 h. In the ponds where DO was 1.2–1.7 mg L?1, silver perch showed signs of severe stress, but continuous aeration (10 hp ha?1) for 3 days and inflow of well‐oxygenated water for 6–8 h prevented mortalities. At temperatures of 13.2–15.7°C, concentrations of 30 or 40 mg L?1 formalin caused DO to decline from 9.0–10.0 to 6.0–8.1 mg L?1 and pH from 7.0–7.3 to 5.9–6.6 within 72 h. Total ammonia‐nitrogen increased over 72 h in ponds treated with 30 or 40 mg L?1 formalin. Fish became re‐infested with L. bidyana in all ponds within 30 days of treatment. A concentration of 30 mg L?1 formalin is recommended as a treatment for monogeneans on silver perch in ponds, but aeration is necessary to maintain adequate water quality at higher temperatures.  相似文献

Increased amounts of reactive oxygen species (ROS) during in vitro fertilization (IVF) may cause cytotoxic damage to gametes, whereas small amounts of ROS favour sperm capacitation. The aim of this study was to investigate the effect of antioxidants [50 μ m β-mercaptoethanol (β-ME) and 50 μ m cysteamine (Cyst)] or a pro-oxidant (5 m m buthionine sulfoximine) on the quality and penetrability of spermatozoa into bovine oocytes and on the subsequent embryo development and quality when added during IVF. Sperm quality, evaluated by the integrity of plasma and acrosomal membranes, and mitochondrial function, was diminished (p < 0.05) after 4-h culture in the presence of antioxidants. Oocyte penetration rates were similar between treatments (p > 0.05), but antioxidants adversely affected the normal pronuclear formation rates (p < 0.05). The incidence of polyspermy was high for β-ME (p < 0.05). No differences were observed in cleavage rates between treatments (p > 0.05). However, the developmental rate to the blastocyst stage was adversely affected by Cyst treatment (p < 0.05). The quality of embryos that reached the blastocyst stage, evaluated by total, inner cell mass (ICM) and trophectoderm cell numbers and ICM/total cell ratio was unaffected (p > 0.05) by treatments. The results indicate that ROS play a role in the fertilizing capacity in bovine spermatozoa, as well as in the interaction between the spermatozoa and the oocytes. It can be concluded that supplementation with antioxidants during IVF procedures impairs sperm quality, normal pronuclear formation and embryo development to the blastocyst stage.  相似文献

Disturbance patterns are strongly coupled with forest composition and structure, and patterns change through time in response to shifts in climate, anthropogenic impacts and other factors. Knowledge of the natural disturbance patterns for establishing baseline conditions for a forest type or ecosystem facilitates change detection for other elements of the biophysical system important to management and conservation. Dendrochronological reconstructions from old-growth forest remnants throughout northeastern North America document average decadal rates of disturbance of 5%–<10% over the last 150–300 years. Relatively frequent, low severity disturbance characterized by small gaps representing canopy openings made by 1–3 trees prevail in these forests dominated by varying mixtures of late-successional tree species. Few studies, though, have explicitly characterized differences related to composition or topographic setting in old-growth landscapes. We addressed this by comparing the temporal and spatial disturbance patterns reconstructed from tree rings at two spatial resolutions (0.5 ha and 200 m2). Sites were selected to assess the influence of topography (slope) and cover type in stands where red spruce (Picea rubens Sarg) and balsam fir (Abies balsamea L. Mill) were key components. Low rates of disturbance (average <10% per decade) and small gap sizes (≤30 m2) prevailed in all stands during the decades from 1850–1980. Episodic pulses of disturbance, of nearly moderate intensity in some stands, opened ca. 20–30% of the canopy area and were associated with wind events and/or insect outbreaks that differentially affected stands. We found no significant difference in the average temporal rates of disturbance related to cover type or topography in 0.5-ha plots. However, the influence of these factors was evident in comparisons of gap areas estimated for 200-m2 plot sections. At this resolution, the largest canopy openings (≥100 m2) occurred most frequently in slope sites, enabling pulses of canopy accession for Betula alleghaniensis (Britton). Whereas the smallest canopy openings (≤30 m2) dominated softwood stands, favoring red spruce, balsam fir and other shade-tolerant species throughout the forest. The variable effects of common disturbance agents, regardless of topographic position and/or cover types, points to the important role of biological legacies determining stand structure and composition on subsequent disturbance events and long-term patterns.  相似文献

Disease ecology involves a systematic approach to understanding the interactions and evolution of host-pathogen systems at the population level, and is essential for developing a comprehensive understanding of the reasons for disease persistence and the most likely means of control. This systems or ecological approach is being increasingly recognised as a progressive method in disease control and is exploited in diverse fields ranging from obesity management in humans to the prevention of infectious disease in animal populations. In this review we discuss bovine tuberculosis (bTB) in Great Britain (GB) within a disease ecology context, and suggest how a comparative ecological perspective helps to reconcile apparent conflicts with the evidence on the effectiveness of badger culling to assist in the control of bTB in GB and the Republic of Ireland (ROI). Our examination shows that failure of past measures to control bTB and the disparity in outcomes of badger culling experiments are the result of a complex relationship amongst the agent, host and environment, i.e. the episystem, of bTB. Here, we stress the role of distinctive bTB episystems and badger culling trial design in the ambiguity and resulting controversy associated with badger culling in GB and ROI. We argue this episystem perspective on bTB control measures in cattle and badger populations provides a useful and informative perspective on the design and implementation of future bTB management in GB, particularly at a time when both scientific and lay communities are concerned about the ongoing epidemic, the cost of current control measures and the execution of future control procedures.  相似文献

Background, aim and scope  Unresolved complex mixtures (UCMs) of aromatic hydrocarbons are widespread, but often overlooked, environmental contaminants. Since UCMs are generally rather resistant to bacterial degradation, bioremediation of UCM-contaminated sites by bacteria is a challenging goal. Branched chain alkyltetralins are amongst the individual classes of components of aromatic UCMs which have been identified in hydrocarbon-contaminated sediments and a number of synthetic alkyltetralins have proved toxic in laboratory studies. Thus, alkyltetralins should perhaps be amongst the targets for UCM bioremediation strategies. The slow degradation of several alkyltetralins by a microbial consortium has been reported previously; however, the bacteria involved remain unidentified and no single strain capable of alkyltetralin biodegradation has been isolated. The present project therefore aimed to enrich and identify bacterial consortia and single strains of bacteria from a naturally hydrocarbon-contaminated site (Whitley Bay, UK), which were capable of the degradation of two synthetic alkyltetralins (6-cyclohexyltetralin (CHT) and 1-(3’-methylbutyl)-7-cyclohexyltetralin (MBCHT)). Materials and methods  Bacteria were enriched from sediment collected from Whitley Bay, UK by culturing with CHT and MBCHT for a period of 4 months. Biodegradation experiments were then established and degradation of model compounds monitored by gas chromatography–mass spectrometry. Internal standards allowed the generation of quantitative data. 16S rRNA gene clone libraries were constructed from individual enrichments to allow assessment of microbial community structure. Selective media containing MBCHT were used to isolate single bacterial strains. These strains were then tested in liquid culture for their ability to degrade MBCHT. Results  The consortia obtained through enrichment culture were able to degrade 87% of CHT and 76% of MBCHT after only 46 days compared with abiotic controls. The 16S ribosomal RNA gene clone libraries of these bacteria were dominated by sequences of Rhodococcus spp. Using selective media, a strain of Rhodococcus was then isolated that was also able to biodegrade 63% of MBCHT in only 21 days. Discussion  The present report describes the isolation of a single bacterial strain able to degrade the resistant MBCHT. Although significant losses of MBCHT were observed, putative metabolites were not detectable. Rhodococcus sp. have been reported previously to be able to biodegrade a range of hydrocarbon compounds. Recommendations and perspectives  Due to their environmental persistence and toxicity, aromatic UCMs require bioremediation. The culturing and identification of such bacteria capable of rapid degradation of alkyltetralins may be an important step toward the development of bioremediation strategies for sites contaminated with toxic UCMs.  相似文献

The indeterminate nature and underground fruiting habit of peanut (Arachis hypogaea L.) makes it a challenge to determine the optimal harvest time. The objectives of this study were (1) to gain additional information about how maturity and canopy health influence peg strength, yield, and grade and (2) to address anecdotal claims of differences in peg strength and digging losses among cultivars. Field trials were established in Citra, FL, and Tifton, GA. Two peanut cultivars (“Georgia-06G” and “Tifguard”) were grown under two fungicide programs (full program: every 14-day spraying interval; and reduced program: every 28-day spraying interval), and harvested at three different maturity levels (early, optimum, and late based on an accumulated growing degree day method). The fungicide program had a significant impact on yield only in 2013 at Citra, likely because the canopy structure was decreased because of fungal disease known as leaf spots (causal agents: Cercospora arachidicola and Cercosporidium personatum). The timing of the harvest had a significant impact on yield, grade, peg strength, and digging loss. In addition, yield, grade, peg strength, and digging losses differed between the peanut cultivars. These results show that reduced fungicide program might affect the yield because of the disease severity and that the optimal harvest time should consider the level of the defoliation. Although there was no correlation between peg strength and digging loss, over-maturity can cause more digging losses compared to optimal harvest time. Tifguard had higher digging loss than Georiga-06G. This study provides additional useful information on cultivars to growers.  相似文献

Integrating bahiagrass (Paspalum notatum Flueggé) into peanut (Arachis hypogaea L.) production systems can improve soil structure, and increase peanut yield. A study was conducted in 2012 and 2013 in north Florida to evaluate practices for planting peanut into bahiagrass with three tillage methods i.e.; strip till (ST), ST and high residue cultivation (ST/HRC), and conventional (CT) tillage. Tillage times were 30 d before planting (DATE1) and at planting (DATE2). Peanut grown under CT outyielded ST/HRC (6940, 6580, and 6370 kg ha?1 for CT, ST, and ST/HRC, respectively), but there was no difference in economic return across treatments (adjusted revenue avg. = US $2478 ha?1). Tillage DATE2 increased total root length compared with DATE1, particularly for CT and ST. The ST/HRC had increased total root length below 30 cm for DATE1. When peanut was planted into bahiagrass, conservation tillage practices maintained adjusted revenue comparable with CT while providing environmental benefits.  相似文献
