Summary A RFLP assay which had been developed for distinguishing between a hybrid pepper and its parents, has been converted into a simple applicable PCR assay, suitable for large-scale quality-control assessment of the commercial hybrid. For this conversion the sequences of the ends of the probe used in the previous RFLP assay were determined. From these sequences suitable primers were devised for inverse PCR of heterogeneous DNA fragments derived from the male parent. The inverse-PCR product was cloned and partially sequenced. These sequences, in turn, made it possible to determine primers on both sides of the locus of mutation, and to develop the reported conventional PCR assay.  相似文献

Genistein, like other phytochemicals, has beneficial health effects, but its bioavailability is limited. This research studied the effect of complexation of genistein with starch on genistein bioavailability. Genistein release from these complexes was tested in vitro under simulated intestinal conditions and in vivo in rats fed high-amylose corn starch (HACS)-genistein complexes (experimental group) as compared to those fed a physical mixture of HACS and genistein (controls). In vitro results showed that genistein release is sustained and fits the normal transit time of food in the intestine. The genistein concentration in the plasma was twice as high in the experimental group versus controls; the genistein concentration in the urine was also higher in the experimental group but lower in the feces. These results indicate that starch-genistein complexes increase genistein bioavailability and suggest that starch can affect the bioavailability of additional food components.  相似文献

In Israel, the cotton mealybug Phenacoccus solenopsis Tinsley, an invasive scale insect, was reported for the first time in the Jordan Valley in 2008 on basil (Ocimum basilicum) and bell pepper (Capsicum annuum). This mealybug is highly polyphagous with economic and environmental impacts. Since then, Ph. solenopsis has spread to almost every region of Israel and developed high populations on several ornamental plants, mainly Hibiscus sp. (Malvaceae) and Lantana sp. (Verbenaceae). It has become a pest in greenhouses, mainly on bell pepper, tomato, and eggplant (Solanaceae) and a serious threat in cotton fields. Fourteen species of insect natural enemies have been found in association with Ph. solenopsis in Israel the common ones were: Aenasius arizonensis (Girault) (Hym. Encyrtidae), Cheilomenes propinqua (Mulsant), Hyperaspis vinciguerrae (Capra); H. polita Weise, Exochomus nigripennis (Erichson), Parascymnus varius Kirsch and Scymnus flagellisiphonatus (Fursch) (Col., Coccinellidae). To date, the population density of Ph. solenopsis in Israel is steeply decreasing in most regions of Israel due to the activity of A. arizonensis. An identification key to distinguish between adult females of the eight species belonging to the genus Phenacoccus in Israel is also provided.  相似文献

Pepper (Capsicum annuum) is an important crop worldwide. In Israel, approximately 2,500 ha are grown all year round for the local and export markets. Herein, we report the identification of a viral pathogen causing a new devastating disease in pepper crops. The disease syndrome includes shortening of stem internodes, interveinal yellowing, and upward rolling of the leaf blade, accompanied by fruit discoloration and size reduction. Virus purification from infected plants yielded isometric particles, 25 nm in diameter. The causal agent of the disease was tentatively named Pepper yellow leaf curl virus (PYLCV). The virus cross-reacted in DAS ELISA with antisera against Cucurbit aphid-borne yellows virus and Potato leafroll virus, members of the Polerovirus genus. The partial nucleotide sequence obtained from the cloned viral coat protein and movement protein genes indicated 92% identity at the amino acid level with Tobacco vein distortion virus (TVDV), another member of the Polerovirus genus. However, the host range of PYLCV is significantly different from the host range described for TVDV. Based on our findings, the taxonomic status of PYLCV is discussed.  相似文献

Plant residues from herb crops were examined as organic amendments for the control of soilborne pathogens with and without soil heating for a short exposure of 2 weeks. Residues of herbaceous plants of oregano, sage, rosemary, tarragon, bay, wild rocket, spearmint and thyme were incubated for 14 days in a controlled laboratory system which emulates soil solarization. Green cabbage residues were included for comparison. The tested pathogens included Fusarium oxysporum f. sp. radicis-lycopersici, Macrophomina phaseolina, Rhizoctonia solani and Meloidogyne javanica. Heating soil amended with herb residues, particularly tarragon, spearmint and wild rocket, resulted in high mortality of the tested pathogens. Volatile compounds which were generated in heated soils amended with wild rocket or tarragon showed significant toxicity to M. phaseolina and R. solani. Only oregano, thyme, bay, wild rocket and tarragon amendments were effective in pathogen control without heating. Incorporation of leaf and stem residues of thyme, sage, tarragon, rosemary or wild rocket in soil and exposure to solarization under field conditions resulted in 95-100% mortality of the tested pathogen. This study demonstrates that the residues from herb crop production can serve as organic amendments for the control of soilborne pathogens, especially when combined with heating or solarization.  相似文献

Visible, near-infrared and shortwave-infrared (VNIR-SWIR) spectroscopy is an efficient approach for predicting soil properties because it reduces the time and cost of analyses. However, its advantages are hampered by the presence of soil moisture, which masks the major spectral absorptions of the soil and distorts the overall spectral shape. Hence, developing a procedure that skips the drying process for soil properties assessment directly from wet soil samples could save invaluable time. The goal of this study was twofold:proposing two approaches, partial least squares (PLS) and nearest neighbor spectral correction (NNSC), for dry spectral prediction and utilizing those spectra to demonstrate the ability to predict soil clay content. For these purposes, we measured 830 samples taken from eight common soil types in Israel that were sampled at 66 different locations. The dry spectrum accuracy was measured using the spectral angle mapper (SAM) and the average sum of deviations squared (ASDS), which resulted in low prediction errors of less than 8% and 14%, respectively. Later, our hypothesis was tested using the predicted dry soil spectra to predict the clay content, which resulted in R2 of 0.69 and 0.58 in the PLS and NNSC methods, respectively. Finally, our results were compared to those obtained by external parameter orthogonalization (EPO) and direct standardization (DS). This study demonstrates the ability to evaluate the dry spectral fingerprint of a wet soil sample, which can be utilized in various pedological aspects such as soil monitoring, soil classification, and soil properties assessment.  相似文献

Electrical heating of food products provides rapid and uniform heating, resulting in less thermal damage to the product. The objective of this research was to examine the effects of ohmic heating on the stability of orange juice with comparison to conventional pasteurization. During storage at 4 degrees C, degradation curves of ascorbic acid followed a linear decrease pattern in both ohmic-heated and conventionally pasteurized orange juices. For five representative flavor compounds (decanal, octana, limonene, pinene, and myrcene), higher concentrations were measured during storage in the ohmic-heated orange juice than in conventionally pasteurized juice. Although residual pectin esterase activity remained negligible in both types of juices, particle size was lower in the ohmic-heated orange juice. The sensory shelf life was determined by using the Weibull-Hazard method. Although both thermal treatments prevented the growth of microorganisms for 105 days, the sensory shelf life of ohmic-treated orange juice was >100 days and was almost 2 times longer than that of conventionally pasteurized juice.  相似文献

Soy isoflavones are widely recognized for their potential health benefits. The increased use of traditional and new food products calls for the assessment of their stability during processing and storage. The present study examines the stability of genistein and daidzein derivatives in soy milk. Soy milk was stored at ambient and elevated temperatures, and the change in isoflavone concentration was monitored with time. Genistin loss in time showed typical first-order kinetics, with rate constants ranging from 0.437-3.871 to 61-109 days(-1) in the temperature ranges of 15-37 and 70-90 degrees C, respectively. The temperature dependence of genistin loss followed the Arrhenius relation with activation energies of 7.2 kcal/mol at ambient temperatures and 17.6 kcal/mol at elevated temperatures. At early stages of soy milk storage at 80 and 90 degrees C, the 6' '-O-acetyldaidzin concentration increased, followed by a slow decrease. The results obtained in this study can serve as a basis for estimating the shelf life of soy milk as related to its genistin content.  相似文献
