FtsZ is a tubulin homolog and the major cytoskeletal protein in bacterial cell division. It assembles into the Z ring, which contains FtsZ and a dozen other division proteins, and constricts to divide the cell. We have constructed a membrane-targeted FtsZ (FtsZ-mts) by splicing an amphipathic helix to its C terminus. When mixed with lipid vesicles, FtsZ-mts was incorporated into the interior of some tubular vesicles. There it formed multiple Z rings that could move laterally in both directions along the length of the liposome and coalesce into brighter Z rings. Brighter Z rings produced visible constrictions in the liposome, suggesting that FtsZ itself can assemble the Z ring and generate a force. No other proteins were needed for assembly and force generation.  相似文献

A small RNA of Bacillus subtilis bacteriophage phi 29 is shown to have a novel and essential role in viral DNA packaging in vitro. This requirement for RNA in the encapsidation of viral DNA provides a new dimension of complexity to the attendant protein-DNA interactions. The RNA is a constituent of the viral precursor shell of the DNA-packaging machine but is not a component of the mature virion. Studies of the sequential interactions involving this RNA molecule are likely to provide new insight into the structural and possible catalytic roles of small RNA molecules. The phi 29 assembly in extracts and phi 29 DNA packaging in the defined in vitro system were strongly inhibited by treatment with the ribonucleases A or T1. However, phage assembly occurred normally in the presence of ribonuclease A that had been treated with a ribonuclease inhibitor. An RNA of approximately 120 nucleotides co-purified with the phi 29 precursor protein shell (prohead), and this particle was the target of ribonuclease action. Removal of RNA from the prohead by ribonuclease rendered it inactive for DNA packaging. By RNA-DNA hybridization analysis, the RNA was shown to originate from a viral DNA segment very near the left end of the genome, the end packaged first during in vitro assembly.  相似文献

通过对兰州市两次强度上有较大差异的尘暴样的采集、粒度分析及其与自然降尘样的对比研究,揭示出兰州市尘暴降尘粒度特性的一些细微特征。不论是自然降尘样还是尘暴样,皆以4~63μm的粉砂为主,说明粉砂粒级的颗粒是兰州地区自然降尘与沙尘暴所携带的主要组分;虽然含量甚微,但是尘暴样中存在近源低空气流所搬运的粒径(181.97~724.44μm)较大的粗颗粒物质。在粒径-体积百分含量曲线上,尘暴样基本上为三峰分布模式,区别于自然降尘样的双峰模式。同时,尘暴样具有向粗、细颗粒区间(尤其是细颗粒区间)延伸并富集的特点,尘暴强度越大,其延伸富集的程度也越明显,而且,随着尘暴强度的增大,其主峰的众数粒径呈减小趋势。  相似文献

Ascospores of Sclerotinia sclerotiorum produced from apothecia are the primary source of inoculum for causing diseases such as white mold of common bean, pod rot of pea, stem blight of canola and head rot of sunflower and safflower in the Canadian prairies. A field study was conducted for 4 years to determine efficacy of control of production of apothecia from carpogenically germinated sclerotia of S. sclerotiorum by soil amendment with Perlka® (calcium cyanamide) and S-H mixture (a formulated compound). Results of the 4-year experiments showed that amendment of soil with Perlka® at low (30 g/m2) or high (60 g/m2) rate was effective in reducing carpogenic germination of sclerotia and production of apothecia under the canopy of host crops (common bean and canola) and a non-host crop (wheat). In the experiments of 1988, for example, the numbers of apothecia produced in the treatments of Perlka®-low rate (30 g/m2), Perlka®-high rate (60 g/m2) and untreated control were 42, 46, and 182 apothecia/plot (m2), respectively, for bean; 89, 42, and 318 apothecia/plot (m2), respectively, for canola; and 146, 143, and 412 apothecia/plot (m2), respectively, for wheat. However, soil amendment of S-H mixture at low (30 g/m2) or high (60 g/m2) rate was ineffective in reducing carpogenic germination of sclerotia and production of apothecia for all the 4 years of testing in all three crops. The ineffectiveness of S-H mixture and the practicality of Perlka® for control of Sclerotinia diseases of crops grown under Canadian prairie conditions are discussed.  相似文献

Chlortetracycline is an antibiotic that is used to increase weight gain, efficiency of gain, carcass grade, and conception rates. The objective of this experiment was to evaluate the effects of supplementation of 350 mg/d of chlortetracycline on ADG, G:F, BCS, thyroxine, and systemic glucose concentrations in yearling dairy heifers. Forty 12-mo-old Holstein heifers (initial BW = 363 +/- 21 kg) were housed in a free-stall barn with ad libitum access to feed and water for 104 d. A transition period was begun 14 d before the age of 12 mo to acclimate the heifers to the diet. The chlortetracycline-fed group (n = 20) consumed 328 +/- 8.2 mg of chlortetracycline/heifer daily. Measurements for BW, withers and hip heights, BCS, and health score were recorded weekly. Dry matter intake was measured daily. Blood was sampled every 4 d to determine plasma thyroxine and glucose concentrations and every 2 d to determine progesterone concentrations. Heifers were artificially inseminated on the first observed standing heat after 13 mo of age. There were no effects of chlortetracycline on ADG, G:F, withers and hip heights, BCS, blood glucose concentrations, peak progesterone concentrations, health, or conception rate. There was an interaction between treatment and time for chlortetracycline on serum thyroxine concentration. In the beginning of the experiment, serum thyroxine concentration was lower in heifers supplemented with chlortetracycline. There was no difference between treatments in thyroxine concentration at the end of the experiment. Chlortetracycline supplementation was not beneficial for yearling dairy heifers.  相似文献
