Molecular cloning of a gene sequence regulated by nerve growth factor   总被引:22,自引:0,他引:22  
Nerve growth factor (NGF) is essential for the development and differentiation of sympathetic or sensory neurons. A complementary DNA was cloned that corresponds to a gene sequence induced more than 50-fold in a cultured target cell line of pheochromocytoma cells (PC12 cells) 5 hours after the addition of NGF. The induced messenger RNA encodes a 90,000-dalton polypeptide that may represent one of the primary events in NGF-induced differentiation of neurons.  相似文献

Many animals create disturbances on the soil surface while constructing habitat and resting sites, or foraging for food. This soil disturbance, which is sometimes known as biopedturbation, is a major contributor to landscape patchiness in arid and semi-arid environments. In the semi-arid woodlands of eastern Australia, the Short-Beaked Echidna (Tachyglossus aculeatus) creates a mosaic of foraging pits close to the canopies of large trees. The effects of pits on physical, chemical and biological properties of soils were compared at seven sites, each with two levels of disturbance (foraging pit vs. surface) and two canopy locations (under the canopy, in the open) associated with two tree species (Eucalyptus intertexta, Alectryon oleifolius). Foraging pits trapped twice the mass of litter compared with adjacent non-pit surfaces, and there was more litter under the tree canopies than in the open. Pits contained more bark and leaf material, and larger pits tended to trap more litter. Soil electrical conductivity levels were lower in the pits, and although there were greater concentrations of soil nutrients under the canopy, the concentrations of total carbon, nitrogen and sulphur were lower in the pits compared with the surface. Changes in litter mass did not explain differences in soil carbon, nitrogen or phosphorus. Soil in the pit was moister and more porous, and surface temperatures below the litter in the pits about 2 °C lower than at the surface. Respiration was about 30% greater in the pits, and both the early (sorptivity) and late (steady-state infiltration) stages of infiltration were significantly greater in the pits. Soil micro-arthropods were more abundant in the pits, which supported a different complement of taxa, but a similar diversity, to non-pit surfaces. Our results indicate that echidna foraging pits act as substantial resource traps. Given their extensive distribution in semi-arid woodlands, and their marked influence on soil biogeochemistry, echidnas should be seen as important ecosystem engineers in woodland critical for the maintenance of small-scale patchiness and, therefore, the efficient functioning of arid and semi-arid ecosystems.  相似文献

An extensive field comparison of the gamma interferon (IFN-gamma) assay and the single intradermal tuberculin test for the diagnosis of bovine tuberculosis was conducted in Australia. The specificity of the IFN-gamma assay was determined by testing more than 6000 cattle from tuberculosis-free herds and varied from 96.2% to 98.1%, depending on the cut-off point chosen to define a positive reactor. For the sensitivity trial, cattle from herds being de-populated because of bovine tuberculosis were examined with both assays. The sensitivity of the IFN-gamma assay was shown to be significantly higher than the single intradermal tuberculin test and varied from 76.8% to 93.6% depending on the method of interpretation. A maximum overall sensitivity of 95.2% was obtained by testing with the IFN-gamma and the tuberculin test in parallel. The superior sensitivity of the IFN-gamma assay and the ability to adjust the sensitivity of the system depending on the task involved, will provide the Australian Tuberculosis Eradication Campaign with a valuable additional test to enable it to accomplish its goals.  相似文献

Brachygnathia superior and generalized diarthrodial degenerative joint disease were seen in 17 related, purebred Angus calves ranging in age from 2 days to 4 months. Craniometrical studies revealed decreased maxillary and palatine bone lengths and increased cranial, skull, and facial indices. Radiological evaluation of major appendicular joints demonstrated lipping of the joint margins with osteophyte formation, sclerosis of subchondral bone, and narrowing of joint spaces. Synovial fluid evaluation indicated joint degeneration but no etiologic agent. Rheumatoid factor analysis of plasma was negative. Grossly, all major appendicular joints were defective including the atlanto-occipital articulation. Lesions ranged from loss of surface luster to erosions and deep ulcers with eburnation of the subchondral bone and secondary proliferative synovitis. Histological changes were degeneration of the articular cartilage matrix, chondrocyte necrosis, flaking and fibrillation, chondrone formation, erosions and ulcers of the articular cartilage with subchondral bone sclerosis, vascular invasion with fibrosis, and chronic, nonsuppurative, proliferative synovitis. Growth plates had defective chondrocyte proliferation and hypertrophy with aberrant ossification of calcified cartilaginous matrix. Histochemical analysis of cartilage and bone failed to incriminate which component was defective, glycosaminoglycan or collagen, but indicated different distribution or absence of one or the other. Genealogic studies revealed a genetic basis for the new defect.  相似文献

Background

Multiple sclerosis (MS), Hashimoto's disease and Graves' disease are autoimmune diseases that may share similar pathogenic mechanisms. The co-occurrence rates and demographic characteristics of Graves' disease and Hashimoto's disease (HT) in our MS population are compared with the general population.

Methods

The prevalence of thyroid disease in our MS patients was determined by chart review and survey. Previous diagnosis of thyroid disease, age at diagnosis, treatment used, and about the use of disease modifying medications used to treat their MS were asked. Chart reviews were used to estimate the population prevalence of Graves' disease and Hashimoto's disease and to estimate the demographics of patients with thyroid disease.

Results

A significant co-occurrence of Graves' disease with MS (p = 0.002), and a non-significant co-occurrence of Hashimoto's disease were noted (p = 0.097). No difference in the age of onset or gender of thyroid disease in MS patients compared to the general population was found.

Conclusion

There is a significant co-occurrence in patients with MS and Graves' disease, and a trend to co-occurrence in patients with MS and Hashimoto's disease. There are no differences in the demographics of patients with thyroid disease in our MS patients compared to the general population.

Parthenote embryos are being considered as an alternative source of embryonic stem cells. However, as there is still a dearth of knowledge of this kind of embryos, a better understanding of their biology is needed for their application. In this work, we studied the differences and similarities between parthenotes and normal embryos at the blastocyst stage in vivo developed. We analysed the expression of factor OCT‐4, vascular endothelial growth factor (VEGF), insulin‐like growth factor I (IGF‐I) and uteroglobin (UG) by real‐time PCR. To do so, oocytes were recovered and after activation procedure were transferred by ventral middle laparoscopy to receptive does to undergo completely in vivo development. Does were slaughtered 6 days post‐ovulation induction, and parthenote and normal embryos were recovered for mRNA expression analysis. Our results reported that parthenotes and normal embryos showed similar mRNA expression for OCT‐4 and VEGF. However, IGF‐I and UG showed to be over‐expressed in parthenote embryos. Thus, our study highlights that despite the in vivo development of parthenotes, they still seem to have an altered expression and, therefore, to be different to normal embryos. The altered expression pattern of parthenote embryos suggests that these embryos should be studied carefully before future application.  相似文献
