Using extracts from suspension-cultured cells of soybean (Glycine max cv. Mandarin) as a source of active enzymes, the activities of glutathione transferases (GSTs) catalysing the conjugation of 1-chloro-2,4-dinitrobenzene (CDNB) and selective herbicides were determined to be in the order CDNB≫ fomesafen>metolachlor=acifluorfen>chlorimuron-ethyl. GST activities showed a thiol dependence in a substrate-specific manner. Thus, GST activities toward acifluorfen and fomesafen were greater when homoglutathione (hGSH), the endogenously occurring thiol in soybean, was used as the co-substrate rather than glutathione (GSH). Compared with GSH, hGSH addition either reduced or had no effect on GST activities toward other substrates. In the absence of enzyme, the rates of hGSH conjugation with acifluorfen, chlorimuron-ethyl and fomesafen were negligible, suggesting that rapid hGSH conjugation in soybean must be catalysed by GSTs. GST activities were subsequently determined in 14-day-old plants of soybean and a number of annual grass and broadleaf weeds. GST activities of the plants were then related to observed sensitivities to post-emergence applications of the four herbicides. When enzyme activity was expressed on a mg-1 protein basis, all grass weeds and Abutilon theophrasticontained considerably higher GST activity toward CDNB than soybean. With fomesafen as the substrate, GST activities were determined to be in the order soybean≫Echinochloa crus-galli>Digitaria sanguinalis>Sorghum halepense=Setaria faberi with none of the broadleaf weeds showing any activity. This order related well to the observed selectivity of fomesafen, with the exception of A. theophrasti, which was partially tolerant to the herbicide. Using metolachlor as the substrate the order of the GST activities was soybean>A. theophrasti≫S. halepense>Amaranthus retroflexus>Ipomoea hederacea, with the remaining species showing no activity. GST activities toward metolachlor correlated well with the selectivity of the herbicide toward the broadleaf weeds but not toward the grass weeds. Acifluorfen and chlorimuron-ethyl were selectively active on these species, but GST activities toward these herbicides could not be detected in crude extracts from whole plants. © 1997 SCI 相似文献

Brans from rice, oats, corn, and wheat were cooked in a twin-screw extruder at either high or low energy input, and their cholesterol-lowering effects were compared with those of unprocessed brans when fed to four-week-old male golden Syrian hamsters (n = 10 per treatment) for three weeks. Peanut oil was added to oat, corn, and wheat bran during the extrusion process to match the oil content of rice bran. Diets contained 10% total dietary fiber, 10.3% fat, 3% nitrogen, and 0.3% cholesterol. Plasma and liver cholesterol and total liver lipids were significantly lower with low-energy extruded wheat bran compared with unprocessed wheat bran. Extrusion did not alter the hypocholesterolemic effects of rice, oat, or corn brans. Plasma and liver cholesterol levels with corn bran were similar to those with oat bran. Relative cholesterol-lowering effects of the brans, determined with pooled (extruded and unextruded) bran data, were rice bran > oat bran > corn bran > wheat bran. Rice bran diets resulted in significantly lower levels of total plasma cholesterol and very low density lipoprotein cholesterol compared with all other brans. Total liver cholesterol and liver cholesterol concentrations (mg/g) were significantly lower with high-energy extruded rice bran compared with the cellulose control group. Plasma cholesterol and total liver cholesterol values with low-energy extruded wheat bran were similar to those with rice bran (unextruded or extruded) diets. Lowered cholesterol with rice bran diets may result in part from greater lipid and sterol excretion with these diets. Results with low-energy extruded wheat bran suggest that this type of processing may improve the potential for lowering cholesterol with wheat bran products. 相似文献

High concentrations of the mycotoxins HT-2 and T-2 (HT2?+?T2), primarily produced by Fusarium langsethiae, have occasionally been detected in Norwegian oat grains. In this study, we identified weather variables influencing accumulation of HT2?+?T2 in Norwegian oat grains. Oat grain samples from farmers’ fields were collected together with weather data (2004–2013). Spearman rank correlation coefficients were calculated between the HT2?+?T2 contamination in oats at harvest and a range of weather summarisations within estimated phenological windows of growth stages in oats (tillering, flowering etc.). Furthermore, we developed a mathematical model to predict the risk of HT2?+?T2 in oat grains. Our data show that adequate predictions of the risk of HT2?+?T2 in oat grains at harvest can be achieved, based upon weather data observed during the growing season. Humid and cool conditions, in addition to moderate temperatures during booting, were associated with increased HT2?+?T2 accumulation in harvested oat grains, whereas warm and humid weather during stem elongation and inflorescence emergence, or cool weather and absence of rain during booting reduced the risk of HT2?+?T2 accumulation. Warm and humid weather immediately after flowering increased the risk, while moderate to warm temperatures and absence of rain during dough development, reduced the risk of HT2?+?T2 accumulation in oat grains. Our data indicated that HT2?+?T2 contamination in oats is influenced by weather conditions both pre- and post-flowering. These findings are in contrast with a previous study examining the risk of deoxynivalenol contamination in oat reporting that toxin accumulation was mostly influenced by weather conditions from flowering onwards. 相似文献

Phytosterols (PS) lower low density lipoprotein (LDL) levels by as much as 10–14% in normal and hypercholesteromic people. High LDL levels in the blood are associated with an increased risk of coronary heart disease. Wheat germ and bran contain significant amounts of PS. The main objective of this study was to evaluate the effect of genotype and environment on PS content and composition in whole wheat grain. Protein and ash contents and mineral composition of the samples were also determined. Grain samples were collected from three cultivars, Jagger, Trego, and Intrada, grown at three locations, Alva, Balko, and Goodwell, OK in 2005. Irrigated and dryland samples were obtained from Goodwell. Total PS content and composition in the grain samples were determined using a gas chromatography system. Whole wheat grain samples varied in PS content from 202 mg/kg to 355 mg/kg. β‐Sitosterol, campesterol, and stigmasterol were the major PS compounds found in all the samples. PS exhibited a significant location‐by‐cultivar random effect (P < 0.048), indicating the presence of genotype‐by‐location interaction. Within each location, a significant cultivar effect was also observed. Protein and ash contents and mineral composition of the samples, except those collected from Goodwell‐dryland, were within the range published in the literature for other wheat cultivars. A fundamental understanding of compositional variation in wheat grain requires multi‐environment testing of genotypes, perhaps over several years. ObjectiveTo measure the extradural pressures in goats before and after extradural injection, and to investigate the occurrence of extradural pressure waves.Study designProspective experimental trial.AnimalsNine healthy adult goats weighing 59.4 ± 6.4 kg, scheduled for stifle arthroscopy.MethodsThe goats were pre–medicated with midazolam and anaesthesia was induced with propofol and maintained with sevoflurane. The goats were placed in lateral recumbency and extradural puncture was performed via the lumbosacral space. Correct placement of the needle was assessed by lack of resistance to the injection of saline. The needle was connected to an electronic pressure transducer to record extradural pressure. Measurements were taken before and after extradural injection of methadone (0.1 mg kg?1, diluted to a total volume of 0.2 mL kg?1) and 10 minutes later. Contrast medium was injected and correct extradural needle placement confirmed by radiography.ResultsLack of resistance to injection of saline occurred in all goats, but there were no pressure waves observed before injection in any animal. Radiography indicated incorrect needle placement in four animals and one had pressure waves synchronous with the arterial pulse after methadone injection. Correct needle placement was confirmed in the remaining five animals which exhibited pressure waves after extradural methadone injection. In the five goats with successful needle placement the baseline extradural pressure ranged from 0.4 to 2.5 kPa (3–19 mmHg), increasing to 4.4–39.9 kPa (33–300 mmHg) after injection. Ten minutes after injection, extradural pressure remained elevated and ranged from 2.5 to 17.3 kPa (19–130 mmHg).Conclusions and clinical relevanceExtradural pressure waves were not useful to confirm correct extradural needle placement in laterally recumbent goats. The presence of such waves after injection of 0.2 mL kg?1 may be indicative of correct placement but even here we saw one of nine animals with extradural pressure waves where we failed to confirm correct needle placement. Genetic parameters of piglet survival traits and birth weight were estimated on the first generation data of a selection experiment aimed at improving piglet survival using a multiple trait linear and threshold model. Data on 5293 piglets for survival at birth, at day one after birth and during the entire nursing period, as well as individual birth weight and litter size, were recorded in an outdoor production system. Genetic effects of piglet survival traits and birth weight were estimated based on threshold and Gaussian models, respectively, using a Bayesian approach. The statistical model included as fixed effects selection group, parity, gender, fostering, gestation length and month of farrowing and, alternatively, an adjustment for litter size. Direct genetic effects (i.e. the piglet's genetic potential) for piglet survival and birth weight were estimated separately, whereas maternal genetic and environmental effects could only be estimated for the given data structure in a combined litter effect. Posterior means of heritabilities for direct genetic effects of survival at birth, at first day after birth and the entire nursing period, as well as birth weight, were 0.08, 0.07, 0.08 and 0.20, respectively. Genetic correlations among survival traits were in the range of 0.29 to 0.40 and indicate that these traits were mainly attributable to different genetic effects. Genetic correlations between direct effects of survival traits and birth weight ranged between 0.18 and 0.23 and were reduced when weights of stillborn piglets were omitted in the analysis or the traits were adjusted for litter size. The magnitudes of direct genetic effects of survival traits are substantially higher than estimates in the literature, which may indicate that these traits have a higher genetic influence under outdoor conditions. The use of birth weight in the multiple trait estimation provided important information for the estimation of survival traits due to its favourable genetic correlations with survival, its high heritability and its high information content as a continuously measured trait. 相似文献

Within-field variability in the Fusarium head blight (FHB) and its associated mycotoxins was studied in four European countries.
At each of 14 sites, each FHB pathogen and associated mycotoxins were quantified in 16 quadrat samples at harvest. Overall,
the incidence of quadrat samples with detectable and quantifiable pathogen DNA was significantly lower in the grain than in
the corresponding chaff. Deoxynivalenol (DON) was the most frequently detected toxin in the samples and its accumulation was
most strongly associated with the presence of Fusarium graminearum. Nivalenol (NIV) accumulation was significantly associated only with the presence of F. culmorum. Zearalenone (ZON) accumulation was strongly associated with the presence of all three pathogens (F. graminearum, F. culmorum and F. poae). The levels of both DON and ZON concentrations were positively related to the amount of F. graminearum DNA in the grain or in the chaff. The presence/absence of FHB pathogens within a single quadrat appeared to be independent
of each other. The presence of a particular FHB pathogen and the amount of its DNA, as well as the associated mycotoxin(s),
varied greatly among samples at each site. This study demonstrated the large extent of within-field variability of FHB and

Vermicomposts are produced through interactions between earthworms and microorganisms in the breakdown of organic wastes. Aqueous extracts were prepared in commercial brewing equipment (Growing Solutions Inc.) from vermicomposts processed from pre-consumer food waste. The ratio of vermicompost to water was 1 to 5 v:v to produce a 20% aqueous solution. The effects of soil drenches at dilutions of 20%, 10%, and 5% vermicompost extracts, were compared with those of deionized water, in the suppression of green peach aphids, mealybugs, and two spotted spider mites attacking tomatoes and cucumbers, in greenhouse cage experiments. 相似文献
