The levels of diffusable and extractable gibberellin-like substances (GAs) of apple fruits were investigated in the alternate-bearing ‘King of the Pippins’ and the regularly fruiting ‘Golden Delicious’, using the barley endosperm bioassay. During 1977 and 1978 there were marked differences between the two cultivars in both the chronological appearance and the absolute amounts of diffusable GAs. The application of fruit-thinning chemicals (carbaryl, ethephon, NAAm) to ‘King of the Pippins’ suppressed the early increase of GAs in diffusates, which normally occurs during the second week after full bloom. The following year, the thinned trees exhibited heavy flower- and fruit-set, while the untreated control trees showed a very poor bloom. The later appearance of the GA peak in diffusates from fruits of ‘Golden Delicious’ (5 weeks after full bloom), was not accompanied by a similar inhibitory effect on subsequent flowering.In contrast to their effect on the levels of diffusable GAs, the thinning-treatments caused no real change in the amount of extractable GAs, which was very similar in fruits from both cultivars.  相似文献

Veränderungen in der Futterqualität von Phalaris arundinacea wurden in Abhängigkeit vom Schnittzeitpunkt im 1. Aufwuchs und variierter Nachwuchsdauer im 2. Aufwuchs untersucht. Daraus ließen sich die optimalen Nutzungszeitspannen von Phalaris arundinacea bestimmen. Überflutungen beeinflussen ebenfalls die Länge der optimalen Nutzungszeitspannen und die Futterqualität. Bei Phalaris arundinacea stieg der Rohfaser‐ und Ligningehalt sowie der K/N—Quotient mit zuneymendem physiologischen Alter an, wogegen der Rohprotein‐ und Kaliumgehalt sank.  相似文献

Citrus, especially K innow (Citrus deliciosa × Citrus nobilis), fruit yield and quality in Pakistan is not competitive with that of other countries which could be mainly attributed to the lack of good nutrient management for citrus orchards. The yield losses in this fruit crop occur mainly due to heavy fruit dropping. Experiments to overcome these problems were conducted at four different sites one each in Faisalabad, Toba Tek Singh, Jhang and Sargodha districts of Punjab, Pakistan. The soil and leaf chemical analysis showed severe deficiency of Zn and our pervious results have shown that soil amendment with potassium (K) at 75 K2O kg ha?1 improved the citrus fruit yield and quality at all selected sites. In the present experiments, effect of foliar application of Zn and K alone or in combination was studied on nutrient uptake, fruit yield, fruit dropping and juice quality. The fruit trees were pretreated with a selected K level of sulfate of potash (SOP) or muriate of potash (MOP), i.e., 75 kg K2O ha?1 along with recommended nitrogen (N) and phosphorus (P) doses. Zinc [Zn, 1% zinc sulfate (ZnSO4) solution], K [1% potassium sulfate (K2SO4) solution] and Zn + K (solution containing 0.5% each of ZnSO4 and K2SO4) were sprayed at the onset of spring and flush of leaves or flowers, fruit formation and at color initiation on fruit. Overall, application of Zn, K or Zn + K was effective in improving the nutrient uptake, yield and quality parameters of citrus fruit at all sites. Fruit dropping was also reduced by the foliar spray of Zn, K or Zn + K but the most promising results were recorded with foliar spray containing both Zn and K.  相似文献

Citrus, especially Kinnow, fruit yield and quality in Pakistan is not competitive with other countries, which could be mainly attributed to a lack of good nutrient management for citrus orchards. Many of the soils under these orchards have been reported as deficient in potassium (K). Therefore, work was initiated for improving citrus fruit yield, size, and quality through K nutrition management. Experiments were conducted in four districts of Punjab including Faisalabad, Toba Tek Singh, Jhang, and Sargodha and four rates of potash, i.e., 0, 50, 75 and 100 kg dipotassium oxide (K2O) ha?1 were applied using sulfate of potash (SOP) along with recommended rates of phosphorus (P) and nitrogen (N). Soils of all the selected sites were clay loam or sandy clay loam and deficient in K. The leaf analysis also showed that all the selected orchards were deficient in K. Application of K increased the fruit yield and quality but K at100 kg K2O ha?1 was more effective in increasing the fruit weight and size, and peel thickness than other K rates in all the selected orchards. Juice volume and percentage significantly increased when K was applied at 75 kg K2O ha?1 at all sites. Nutrient uptake like K, P, and N enhanced with the increase in K application rate, however, the highest calcium (Ca)+ magnesium (Mg) was recorded at 50 kg K2O ha?1. The results indicated that all rates of K application improved the fruit yield and quality and reduced fruit dropping, however, 75 kg K2O ha?1 rate was more effective as juice volume and percentage, total soluble solid (TSS)/acid ratio and nutrient uptake showed significant improvement.  相似文献

The aim of this work was to obtain a series of genetically related lines of Eragrostis curvula with different ploidy levels and reproductive modes, which can be used for the discovery of genes associated to diplospory. E. curvula is a widely cultivated forage grass native to Southern Africa that reproduces naturally by obligate diplosporous apomixis (asexual reproduction by seeds) of the Antennaria type. In vitro culture of immature inflorescences of the apomictic cultivar Tanganyika led to the regeneration of a diploid plant with a particular morphology (plant UNST1122). Embryo sac analysis and progeny tests determined that plant UNST1122 reproduces by sexuality. UNST1122 was cloned by dividing its tillers and a R 1 generation was obtained from seeds after open pollination among the diploid clones. A set of 500 seeds from a diploid R 1 plant was treated with colchicine and two plants showing a duplicated chromosome number (2n = 4x = 40) were obtained (plants UNST1112 and UNST1131). Progeny tests using RAPDs indicated that these plants reproduce also by sexuality. The availability of sexual tetraploid genotypes of E. curvula will allow the production of hybrids with new interesting combinations of agronomic traits and the developing of mapping populations segregating for diplospory. Besides, these genetically related plants with different reproduction modes constitutes an excellent model for the identification of gen(es) involved in diplosporous apomixis by mRNA profiling as well as to study the genomic rearrangements and gene expression alterations associated to changes at ploidy levels.  相似文献

A new silver stain for electrophoretically separated polypeptides can be rapidly and easily used and can detect as little as 0.01 nanogram of protein per square millimeter. When employed with two-dimensional electrophoresis, it should permit qualitative and quantitative characterization of protein distributions in body fluids and tissues. Twenty‐four ejaculates from six (four ejaculates each) Surti buffalo bulls aged 4–8 years were used to assess various attributes of spermatozoa influencing the zona‐binding and zona‐penetration tests. Ejaculates from each bulls were subjected to in vitro sperm‐‐zona binding and sperm‐‐zona penetration tests (four replicates per bull) using immature buffalo oocytes. The average number of spermatozoa bound per oocyte was 27.79 ± 5.90. The average number of spermatozoa penetrated per oocyte was 3.35 ± 0.64. The average number of zona‐bound and ‐penetrated spermatozoa differed significantly between animals. Significant difference (p < 0.05) was observed between the plasmalemma integrity as assessed by eosin‐‐nigrosin stain and hypo‐osmotic swelling (HOS) test. Furthermore, the percentage of cells positive for the HOS test, i.e. functional membrane integrity (51.25 ± 2.32) was significantly (p < 0.05) higher than hypo‐osmotic swelling‐Giemsa (HOS‐G) test, i.e. the subpopulation of spermatozoa positive for functional membrane and acrosomal integrities (42.87 ± 4.56). The HOS test had significant correlations with plasmalemma integrity as measured by the vital stain, eosin‐‐nigrosin (r = 0.85, p < 0.05). The HOS‐G test also had significant correlation with plasmalemma integrity measured by vital stains such as eosin‐‐nigrosin (r = 0.90, p < 0.05) and fluorogenic stains [carboxyfluorescein diacetate (CFDA) and propidium iodide (PI); r = 0.92, p < 0.01] and HOS test (r = 0.93), acrosomal integrity (r = 0.86, p < 0.05) and mitochondrial membrane potential (r = 0.99, p < 0.01). The plasmalemma integrity (fluorogenic stain), functional membrane integrity (HOS test), subpopulation of spermatozoa positive for functional membrane and acrosomal integrities (HOS‐G test) and mitochondrial membrane potential had significant (p < 0.05) correlation with sperm zona binding and penetration. The present study indicates that these parameters could represent important determinants of sperm quality influencing zona binding and penetration.  相似文献

Objective To measure acute pain in sheep, based on a human pain model, and examined changes in both electroencephalogram frequency spectrum and behavioural responses to increased electrical stimulation in sheep. Design Analysis of variance (treatment and animal effects) for stimulus intensity where each animal received each electric shock treatment given in the order 0, 5,10 and 20 mA. Procedure Eight sheep with electrodes implanted over the surface of the brain were examined for escape-avoidance and electroencephalogram responses to four levels of electrical stimulation from 0–20 mA. Results With increasing stimulus intensity at the time of feeding, the sheep were more hesitant to return to the feeder or remain near the feeder following stimulation. There was little difference between the 0 and 5 mA stimuli for any of the behaviour variables (P > 0.05). However, there were marked increases in the time taken to re-approach the feeder after receiving an electric shock of 5 mA and of 20 mA (P < 0.05; mean values 3 and 119 s, respectively) and remaining near the feeder for 5 s (P < 0.001; mean values 10 and 167 s, respectively). Following the stimulus, there was an overall increase in the electroencephalogram power spectrum in the first four seconds, which then rapidly returned to normal. In particular, the 20 mA stimulus resulted in higher absolute power values than in the control (0 mA) treatment for delta 2 (P < 0.001), theta 1 (P < 0.05), theta 2 (P < 0.05), alpha 1 (P < 0.001), alpha 2 (P < 0.001) and beta 1 (P < 0.01) band-widths. Similarly, for the 10 mA stimulus, the absolute power values were greater than the control treatment for delta 2 (P < 0.05), alpha 1 (P < 0.01), alpha 2 (P < 0.001) and beta 1 (P < 0.01) bandwidths. Conclusion The experiment suggests that a human acute pain model is applicable to sheep and that these electroencephalogram changes may provide a good measure of acute pain in sheep.  相似文献
