The number of drought and low-N tolerant hybrids with elevated levels of provitamin A (PVA) in sub-Saharan Africa could increase when PVA genes are optimized and validated for developed drought and low-N tolerant inbred lines. This study aimed to (a) determine the levels of drought and low-N tolerance, and PVA concentrations in early maturing PVA-quality protein maize (QPM) inbred lines, and (b) identify lines harbouring the crtRB1 and LcyE genes as sources of favourable alleles of PVA. Seventy early maturing PVA-QPM inbreds were evaluated under drought, low-N and optimal environments in Nigeria for two years. The inbreds were assayed for PVA levels and the presence of PVA genes using allele-specific PCR markers. Moderate range of PVA contents was observed for the inbreds. Nonetheless, TZEIORQ 55 combined high PVA concentration with drought and low-N tolerance. The crtRB1-3′TE primer and the KASP SNP (snpZM0015) consistently identified nine inbreds including TZEIORQ 55 harbouring the favourable alleles of the crtRB1 gene. These inbreds could serve as donor parents of the favourable crtRB1-3′TE allele for PVA breeding in maize.  相似文献

Surveys on land use dynamics were conducted over a 3-year period along various toposequences in benchmark inland valley watersheds of the Ashanti region, Ghana. Six typical land use systems were selected for more detailed studies, Le., primary forest (PF), cacao farm (CP), fallow (Fallow), mixed cropping (MC), and lowland rice-based systems including both traditional rice (TR) and sawah{ft1} (paddy)-based rice farming (Sawah). Topsoil samples (0-20 cm) and samples of leaves and bark of various plants were collected from these land use systems for nutrient characterization using laboratory analyses. The remaining primary and secondary forests cover only a patchy area, accounting for less than 20%, in forest reserves and surrounding areas. Cacao farms were developed both in the upland areas, middle slopes, and lowland areas, which occupied about 20% of the watershed. Lowland rice farming is estimated to account for about 10% of the watershed. Oil palm cultivation is also common in the lowland areas. The remaining land use systems were mostly fallow and mixed cropping of maize, cocoyam, cassava, and plantain. Plant leaves of various species in the lowland traditional rice area showed higher concentrations of phosphorus and potassium but lower calcium and strontium concentrations than those of the leaves in the upland surveyed plots, especially, the forest area. Nitrogen, sulfur, magnesium, and the other mineral elements, however, did not show any clear differences among the six land use systems. Bark samples tended to exhibit lower concentrations of nitrogen, phosphorus, sulfur, magnesium, sodium, silicon, aluminum, iron, manganese, and copper but higher concentrations of calcium and strontium. Cacao leaves showed silicon-accumulator characteristics and the magnesium concentration was high too. Cacao bark contained potassium and zinc in appreciable amounts in compared to the leaves. Topsoil samples of the primary forest and cacao farms in the upland areas showed higher levels of TC, TN, exchangeable Ca and Mg, and eCEC while the levels of available P showed a rather reverse trend compared to the lowland traditional and sawah rice plots. Topsoil samples of the fallow and mixed cropping plots at upland and fringe sites showed intermediate levels. The pH at the mixed cropping site was the lowest. These general fertility trends along upland and lowland topo-sequences were different from those of monsoon Asia and Japan. With the implementation of the sawah rice-based farming system, the inland valleys could contribute to the enhancement of sustainable food production in the country.  相似文献

Volatiles from crushed and intact guava leaves (Psidium guajava L.) were collected using static headspace SPME and determined using GC-PFPD, pulsed flame photometric detection, and GC-MS. Leaf volatiles from four common citrus culitvars were examined similarly to determine the potential component(s) responsible for guava's protective effect against the Asian citrus psyllid (Diaphorina citri Kuwayama), which is the insect vector of Huanglongbing (HLB) or citrus greening disease. Seven sulfur volatiles were detected: hydrogen sulfide, sulfur dioxide, methanethiol, dimethyl sulfide (DMS), dimethyl disulfide (DMDS), methional, and dimethyl trisulfide (DMTS). Identifications were based on matching linear retention index values on ZB-5, DB-Wax, and PLOT columns and MS spectra in the case of DMDS and DMS. DMDS is an insect toxic, defensive volatile produced only by wounded guava but not citrus leaves and, thus, may be the component responsible for the protective effect of guava against the HLB vector. DMDS is formed immediately after crushing, becoming the major headspace volatile within 10 min. Forty-seven additional leaf volatiles were identified from LRI and MS data in the crushed guava leaf headspace.  相似文献

A study was carried out to assess the growth, flowering and fruiting of stecklings (rooted cuttings), grafts and seedlings of Allanblackia floribunda in Cameroun. Thirty-one individuals of each plantlet type were planted in May 2006. Height, collar and crown diameters were recorded in September and October, while flowering and fruiting were recorded each month from January to December. In 2014, the trees with the greatest height were those from seeds (6.98 ± 0.25 m), followed by those from grafts (1.65 ± 0.23 m) and cuttings (1.09 ± 0.21 m). Grafts started flowering and fruiting 3 years after planting, while seedlings started flowering and fruiting after four and 6 years, respectively. Stecklings still had not flowered 8 years after planting. Fruit weight and length from seed-origin trees are about seven times higher than grafts. Correlations between height and crown diameter were significant and positive irrespective of the nature of the planting material. In conclusion, grafts fruited earlier, while seedlings grew faster and had greater yields than grafts and stecklings of A. floribunda.  相似文献

An improved high performance liquid chromatographic (HPLC) method was developed for the detection and quantitation of host-specific AA-toxin produced byAlternaria alternata, a water hyacinth pathogen. Precolumn derivatization of the toxin witho-phthalaldehyde (OPA) plus mercaptoethanol yielded highly fluorescent products showing well resolved peaks on reverse-phase HPLC. The minimum amounts of AA-toxins detectable by this method were approximately 1 ng. This method seems to offer advantages over conventional techniques, because prechromatographic derivatization of the toxin with OPA is rapid and easy, and fluorescent derivatives permit excellent detection sensitivity.
