The effects of various synthetic medium components and their interactions with each other ultimately impact laccase production in fungi. This was studied using a laccase-hyper-producing marine-derived basidiomycete, Cerrena unicolor MTCC 5159. Inducible laccases were produced in the idiophase only after addition of an inducer such as CuSO4. Concentration of carbon and nitrogen acted antagonistically with respect to laccase production. A combination of low nitrogen and high carbon concentration favored both biomass and laccase production. The most favorable combination resulted in 917 U L−1 of laccase. After sufficient growth had occurred, addition of a surfactant such as Tween 80 positively impacted biomass and increased the laccase activity to around 1,300 U L−1. Increasing the surface to volume ratio of the culture vessel further increased its activity to almost 2,000 U L−1. 相似文献

BACKGROUND: Clomazone is a herbicide used to control broadleaf weeds and grasses. Clomazone use in agriculturally important crops and forests for weed control has increased and is a potential water contaminant given its high water solubility (1100 µg mL?1). Soil sorption is an environmental fate parameter that may limit its movement to water systems. The authors used model rice and forest soils of California to test clomazone sorption affinity, capacity, desorption, interaction with soil organic matter and behavior with black carbon. RESULTS: Sorption of clomazone to the major organic matter fraction of soil, humic acid (HA) (Kd = 29–87 L kg?1), was greater than to whole soils (Kd = 2.3–11 L kg?1). Increased isotherm non‐linearity was observed for the whole soils (N = 0.831–0.893) when compared with the humic acids (N = 0.954–0.999). Desorption isotherm results showed hysteresis, which was greatest at the lowest solution concentration of 0.067 µg mL?1 for all whole soils and HA extracts. Aliphatic carbon content appeared to contribute to increased isotherm linearity. CONCLUSION: The results indicate that clomazone does not sorb appreciably to sandy or clay soils. Its sorption affinity and capacity is greater in humic acid, and consequently clomazone has difficulty desorbing from soil organic matter. Sorption appears to follow processes explained by the dual‐mode model, the presence of fire residues (black carbon) and a recently proposed sorption mechanism. NG-391 (1) and NG-393 (2), previously reported from undescribed Fusarium species as nerve-cell growth stimulants, were identified from fermentation extracts of the entomopathogenic fungus Metarhizium anisopliae. These compounds are 7-desmethyl analogues of fusarin C and (8Z)-fusarin C, mutagenic toxins from Fusarium species that contaminate corn. A mutant strain of M. anisopliae (KOB1-3) overproduces 1 and 2 by ca. 10-fold relative to the wild-type strain, ARSEF 2575, from which it was derived. Overproduction of these compounds in KOB1-3 imparts a yellow pigmentation to the culture medium of the fungus. These compounds were inactive at 100 mug/disk in antimicrobial disk diffusion assays. Compound 1 was inactive at 100 ppm in a mosquitocidal assay. However, like their fusarin analogues, 1 and 2 exhibited potent S9-dependent mutagenic activity in the Salmonella mutagenicity test. Discovery of these highly mutagenic mycotoxins in M. anisopliae suggests that screening for production of NG-391 and NG-393 in strains that are used as biocontrol agents would be a prudent course of action. The impact of these findings on the use of M. anisopliae as a biocontrol agent is currently unknown and requires further investigation. 相似文献

Domoic acid produced by marine algae has been shown to cause acute and chronic neurologic sequelae in Californian sea lions following acute or low-dose exposure. Histological findings in affected animals included a degenerative cardiomyopathy that was hypothesized to be caused by over-excitation of the glutamate receptors (GluRs) speculated to be present in the sea lion heart. Thus tissues from five sea lions without lesions associated with domoic acid toxicity and one animal with domoic acid-induced chronic neurologic sequelae and degenerative cardiomyopathy were examined for the presence of GluRs. Immunohistochemistry localized mGluR 2/3, mGluR 5, GluR 2/3 and NMDAR 1 in structures of the conducting system and blood vessels. NMDAR 1 and GluR 2/3 were the most widespread as immunoreactivity was observed within sea lion conducting system structures. PCR analysis, cloning and subsequent sequencing of the seal lion GluRs showed only 80% homology to those from rats, but more than 95% homologous to those from dogs. The cellular distribution and expression of subtypes of GluRs in the sea lion hearts suggests that exposure to domoic acid may induce cardiac damage and functional disturbances. 相似文献

Broadcast seeding is one of the most widely used post-wildfire emergency response treatments intended to reduce soil erosion, increase vegetative ground cover, and minimize establishment and spread of non-native plant species. We conducted an evidence-based review to examine the effectiveness and effects of post-wildfire seeding treatments on soil stabilization, non-native species invasion, and plant community recovery in the western U.S. We reviewed 94 scientific papers and agency monitoring reports identified using a systematic search protocol. As sampling designs have become more rigorous in recent years, evidence that seeding is effective in reducing erosion has decreased. Of highest and high quality studies evaluating soil erosion, 92% (11 of 12) were published since 2000, none of which showed an effective result. Before 2000, the majority of the studies (71%) fell into the lowest quality categories, of which 72% showed seeding to be effective. The majority of studies (20 of 27, 74%) evaluating soil erosion in seeded versus unseeded controls showed that seeding did not reduce erosion relative to unseeded controls. Even when seeding significantly increased vegetative cover, seeded sites rarely supported plant cover levels considered sufficient to stabilize soils within the first and second year post-wildfire. Of the 11 studies evaluating seeding effectiveness for curtailing invasions of non-native plant species, an almost equal percentage found seeding treatments to be either effective (54%, 6 studies) or ineffective (45%, 5 studies). However, the majority of effective and ineffective treatments (83% and 80%, respectively) used non-native species. Sixteen of 26 studies (62%) evaluating seeding effects on plant communities reported that seeding suppressed recovery of native plants, although data on long-term impacts of this reduction are limited. The literature suggests that post-wildfire seeding does little to protect soil in the short-term, has equivocal effect on invasion of non-native species, and can have negative effects on native vegetation recovery, although long-term studies are needed to assess lasting impacts of seeded species. 相似文献

Landscape influences on stream biotic integrity assessed at multiple spatial scales 总被引:31,自引:0,他引:31
The biological integrity of stream ecosystems depends critically on human activities that affect land use/cover along stream
margins and possibly throughout the catchment. We evaluated stream condition using an Index of Biotic Integrity (IBI) and
a habitat index (HI), and compared these measures to landscape and riparian conditions assessed at different spatial scales
in a largely agricultural Midwestern watershed. Our goal was to determine whether land use/cover was an effective predictor
of stream integrity, and if so, at what spatial scale. Twenty-three sites in first-through third-order headwater streams were
surveyed by electrofishing and site IBIs were calculated based on ten metrics of the fish collection. Habitat features were
characterized through field observation, and site HIs calculated from nine instream and bank metrics. Field surveys, aerial
photograph interpretation, and geographic information system (GIS) analyses provided assessments of forested land and other
vegetation covers at the local, reach, and regional (catchment) scales.
The range of conditions among the 23 sites varied from poor to very good based on IBI and HI scores, and habitat and fish
assemblage measures were highly correlated. Stream biotic integrity and habitat quality were negatively correlated with the
extent of agriculture and positively correlated with extent of wetlands and forest. Correlations were strongest at the catchment
scale (IBI with % area as agriculture, r2=0.50, HI with agriculture, r2=0.76), and tended to become weak and non-significant at local scales. Local riparian vegetation was a weak secondary predictor
of stream integrity. In this watershed, regional land use is the primary determinant of stream conditions, able to overwhelm
the ability of local site vegetation to support high-quality habitat and biotic communities. 相似文献

This study was initiated on the South Fork Edisto River in South Carolina to investigate and compare the impacts of two different harvest methods (helicopter and rubber-tired skidder) on the ecological structure and function of a blackwater forested wetland. The two harvest treatments were chosen to represent a broad spectrum of potential impacts and were compared to an undisturbed site. Following harvest in 1991, functional responses in vegetative productivity, herpetofaunal populations, and soil and water quality were evaluated in 1992 and 1993. Herbaceous biomass was greater on the helicopter and skidder treatments than on the undisturbed control. In general, the biomass measured on the skidder treatment was not significantly different from the biomass measured on the helicopter treatment. Higher decomposition rates were noted in the harvested areas as compared to the control. This was mainly attributed to higher soil temperatures, which accelerated microbial activity. Increased decomposition rates may have been responsible for elevated levels of ammonium and organic carbon observed in ground water samples. A total of 29 species of herpetofauna (10 amphibians and 19 reptiles) were observed during the study. Salamanders were found more often in the undisturbed control. Reptiles were observed more frequently in the harvested area. Although herpetofaunal species composition was different between harvest and control, indices of diversity were similar. 相似文献

Purpose To determine whether ultraviolet (UV) radiation can modulate expression and regulation of matrix metalloproteinases (MMP) in the canine cornea and to examine the expression of MMPs in canine chronic superficial keratitis (CSK). Methods Immunohistochemistry for MMP‐2 and MMP‐9 was performed on samples of CSK. In vitro, canine corneal epithelial cell (CEC) and stromal cell cultures were exposed to UV‐irradiation. Following 2, 8 or 24 h, cells were harvested. MMP expression was examined by zymography, and RT‐PCR was used to examine expression of Slug and Snail. CEC cultures treated with an EGFR inhibitor or a p38 inhibitor were UV‐exposed and harvested 24 h later to examine expression of MMPs, Slug and Snail. Results Canine CSK had increased immunopositivity for both MMP‐2 and MMP‐9 compared to normal canine corneas. In vitro, CEC and stromal cell cultures exposed to UV showed generally increased expression of MMP‐2, ‐9, Slug, and Snail; this response was dose and time dependent. Inhibition of the EGFR pathway did not prevent increased expression of MMP‐2, ‐9, Slug or Snail in UV‐exposed CEC; however, p38 inhibition did attenuate UV induction. Conclusions We have found increased expression of MMPs in clinical samples of CSK compared to normal corneas. In addition, we have shown that there is a temporal association and dose dependency between UV exposure and production of MMPs, Slug, and Snail. These findings suggest that overexpression of MMPs due to UV‐exposure may be linked to changes in the cornea that allow an influx of inflammatory cells and vascularization. 相似文献
