Vitamin D metabolism: the role of kidney tissue 总被引:30,自引:0,他引:30
The appearance of a polar metabolite of 25-hydroxychotecalciferol has been studied in the intestinal mucosas of nephrectomized rats and rats which have been made uremic by ureter ligation. In confirmation of previous work by Fraser and Kodicek, it was found that nephrectomy prevents the appearance of this metabolite in the intestine. On the other hand, equivalent amounts of the metabolite were found in uremic rats and in sham-operated controls, showing that the production of this metabolite is independent of the uremic state of the animals. In addition, it was shown that the metabolite produced in vitro by kidney homogenates is identical to that found in vivo in the intestine. 相似文献

Microbial growth in soil is mostly limited by lack of carbon (C). However, adding fresh, C-rich litter can induce nitrogen (N) limitation. We studied the effect of alleviating C and N limitation in high-pH (> 8) soils, soils expected to favor bacterial over fungal growth. Nitrogen limitation was induced by incubating soils amended with C-rich substrate (starch or straw) for 4 weeks. Limiting nutrients and the effects of alleviating limitation were then studied by adding C (as glucose) or N (as NH4NO3) and measuring microbial growth and respiration after 4 d. In non-amended, C-limited soils, adding C but not N increased both microbial respiration and bacterial growth. In N-limited, substrate-amended soils, adding C increased respiration, whereas adding N increased both microbial respiration and growth. Inducing N limitation by amending with straw was most easily detected in increased fungal growth after the addition of N, whereas with starch, only bacterial growth responded to alleviating N limitation. Compared to earlier results using a low-pH soil, the effect of substrate used to induce N limitation was more important than pH for inducing bacterial or fungal growth after alleviating N limitation. Furthermore, we found no evidence that alleviating N limitation resulted in decreased respiration concomitant with increased microbial growth in soil, suggesting no drastic changes in C use efficiency. 相似文献

The purpose of this study was to assess the effects of charcoal and earthworm presence in contrasting soil types of northern Japan using the biologically based phosphorus (BBP) extraction method, which employs a variety of plant P acquisition strategies. Using soils developed in serpentine and sedimentary parent materials, we tested the interactive effects of Eisenia japonica (Michaelsen) earthworms and 500 kg ha?1 of dwarf bamboo charcoal (Sasa kurilensis (Rupr.) Makino et Shibata) in a microcosm incubation that lasted four weeks. Soils were extracted in parallel after the incubation with the BBP method using 0.01 M CaCl2 (soluble P), 0.01 M citric acid (chelate-extractable P), 0.02 phosphatase enzyme units ml?1 (enzyme-extractable organic P), and 1.0 M HCl (mineral occluded P). Dwarf bamboo charcoal alone contained up to 444 mg total BBP kg?1 prior to application to soil microcosms. Treatment effects in soil microcosms were highest in sedimentary soil types and where charcoal was combined with earthworms (15.97 mg P kg?1 ± SE 1.23 total inorganic BBP). Recalcitrant inorganic P (HCl extracted) in combination treatments yielded the highest single inorganic BBP measure (12.41 mg kg?1 ± SE 1.11). Our findings suggest that charcoal, as a legacy of wildfire, and native earthworm activity may help stimulate cycling of recalcitrant inorganic BBP pools. 相似文献

There is limited understanding of the carbon (C) storage capacity and overall ecological structure of old-growth forests of western Montana, leaving little ability to evaluate the role of old-growth forests in regional C cycles and ecosystem level C storage capacity. To investigate the difference in C storage between equivalent stands of contrasting age classes and management histories, we surveyed paired old-growth and second growth western larch (Larix occidentalis Nutt)–Douglas-fir (Pseudostuga menziesii var. glauca) stands in northwestern Montana. The specific objectives of this study were to: (1) estimate ecosystem C of old-growth and second growth western larch stands; (2) compare C storage of paired old-growth–second growth stands; and (3) assess differences in ecosystem function and structure between the two age classes, specifically measuring C associated with mineral soil, forest floor, coarse woody debris (CWD), understory, and overstory, as well as overall structure of vegetation. Stands were surveyed using a modified USFS FIA protocol, focusing on ecological components related to soil, forest floor, and overstory C. All downed wood, forest floor, and soil samples were then analyzed for total C and total nitrogen (N). Total ecosystem C in the old-growth forests was significantly greater than that in second growth forests, storing over 3 times the C. Average total mineral soil C was not significantly different in second growth stands compared to old-growth stands; however, total C of the forest floor was significantly greater in old-growth (23.8 Mg ha−1) compared to second growth stands (4.9 Mg ha−1). Overstory and coarse root biomass held the greatest differences in ecosystem C between the two stand types (old-growth, second growth), with nearly 7 times more C in old-growth trees than trees found on second growth stands (144.2 Mg ha−1 vs. 23.8 Mg ha−1). Total CWD on old-growth stands accounted for almost 19 times more C than CWD found in second growth stands. Soil bulk density was also significantly higher on second growth stands some 30+ years after harvest, demonstrating long-term impacts of harvest on soil. Results suggest ecological components specific to old-growth western larch forests, such as coarse root biomass, large amounts of CWD, and a thick forest floor layer are important contributors to long-term C storage within these ecosystems. This, combined with functional implications of contrasts in C distribution and dynamics, suggest that old-growth western larch/Douglas-fir forests are both functionally and structurally distinctive from their second growth counterparts. 相似文献

This study was designed to evaluate effects of different combinations of cryoprotectants on the ability of vitrified immature buffalo oocytes to undergo in vitro maturation. Straw and open‐pulled straw (OPS) methods for vitrification of oocytes at the germinal vesicle stage also were compared. The immature oocytes were harvested from ovaries of slaughtered animals and were divided into three groups: (i) untreated (control); (ii) exposed to cryoprotectant agents (CPAs); or (iii) cryopreserved by straw and OPS vitrification methods. The vitrification solution (VS) consisted of 6 m ethylene glycol (EG) as the standard, control vitrification treatment, and this was compared with 3 m EG + 3 m dimethyl sulfoxide (DMSO), 3 m EG + 3 m glycerol, and 3 m DMSO + 3 m glycerol. Cryoprotectants were added in two steps, with the first step concentration half that of the second (and final) step concentration. After warming, oocyte samples were matured by standard methods and then fixed and stained for nuclear evaluation. Rates of MII oocytes exposed to CPAs without vitrification were lower (54.3 ± 1.9% in EG, 47.5 ± 3.4% in EG + DMSO, 36.8 ± 1.2% in EG + glycerol and 29.9 ± 1.0% in DMSO + glycerol; p < 0.05) than for the control group (79.8 ± 1.3%). For all treatments in each vitrification experiment, results were nearly identical for straws and OPS, so all results presented are the average of these two containers. The percentages of oocytes reaching telophase‐I or metaphase‐II stages were lower in oocytes cryopreserved using all treatments when compared with control. However, among the vitrified oocytes, the highest maturation rate was seen in oocytes vitrified in EG + DMSO (41.5 ± 0.6%). Oocytes cryopreserved in all groups with glycerol had an overall low maturation rate 19.0 ± 0.6% for EG + glycerol and 17.0 ± 1.1% for DMSO + glycerol. We conclude that the function of oocytes was severely affected by both vitrification and exposure to cryoprotectants without vitrification; the best combination of cryoprotectants was EG + DMSO for vitrification of immature buffalo oocytes using either straw or OPS methods. 相似文献

A tunable Kondo effect has been realized in small quantum dots. A dot can be switched from a Kondo system to a non-Kondo system as the number of electrons on the dot is changed from odd to even. The Kondo temperature can be tuned by means of a gate voltage as a single-particle energy state nears the Fermi energy. Measurements of the temperature and magnetic field dependence of a Coulomb-blockaded dot show good agreement with predictions of both equilibrium and nonequilibrium Kondo effects. 相似文献

This article focuses on the pharmacologic properties of drugs commonly used in the treatment of neonatal septicemia and its complications. Rational therapy demands an awareness of not only the pharmacology of individual drugs but also the interactions and anticipated fate of such drugs in the rapidly changing physiologic environment of the neonate. Further research in the area of equine neonatal pharmacology should greatly assist our understanding of the impact of the disease state on the unique physiology of the newborn and should allow us to better predict the ultimate fate of drugs commonly used for such purposes. Careful dosing and close monitoring of pharmacologic effects are critical for a successful outcome. In the future, newer therapeutic strategies that are safe and efficacious may provide a means to circumvent many of the problems currently encountered with treating the septicemic newborn foal. 相似文献

- 1. Cymodocea nodosa's leaf length distribution was studied as an easily measurable indicator to monitor and conserve Macedonian, North Aegean, Greek coastal habitats.
- 2. Three Cymodocea meadows off the eastern Kavala Gulf coast (Nea Karvali, Erateino, Agiasma), with that of Nea Karvali close to an industrial area being the most degraded, were sampled during the seagrass main growing season in July 2004. Two further meadows, one pristine to less degraded (Brasidas, Gulf of Kavala) and one degraded (Biamyl, Inner Thessaloniki Gulf), were sampled as benchmarks in July 2005. The results were evaluated using Gaussian fit curves, and non‐parametric and nested parametric ANOVA on a hierarchy of spatial scales: area (tens of metres), site (hundreds of metres) and meadow (kilometres).
- 3. Frequency (%) distribution of leaf length values and CymoSkew index variation were best associated with anthropogenic stress. Frequency (%) distribution of adult and intermediate photosynthetic leaf length values revealed a unimodal distribution possible to be fitted, at least at pristine to less degraded meadows, by normal distribution (R2>0.5).
- 4. Statistically significant variation was estimated for CymoSkew index, a quantitative expression of leaf length asymmetry, on the meadow scale (P<0.001). Biamyl (3.82) and Nea Karvali (3.64) were indicated as heavily degraded meadows, Erateino (2.93) as a degraded meadow, Agiasma (2.18) as a meadow with the first signs of degradation, and Brasidas (1.68) as a pristine to less degraded meadow. These results in combination with other meadow specific biotic parameters were used to suggest a preliminary angiosperm ‘Ecological Status Classes’ classification scheme useful for the implementation of WFD in the north Aegean Sea.

BACKGROUND: Chemokine expression in airway epithelium and bronchoalveolar lavage fluid (BALF) cells of horses with recurrent airway obstruction (RAO) is increased. HYPOTHESIS: For RAO-affected horses that are stabled and fed a pelleted ration, the addition of oral dexamethasone further improves pulmonary function and reduces inflammatory gene expression in pulmonary cells. ANIMALS: Twelve RAO-affected horses. METHODS: In a randomized cross-over experiment, the effect of feeding pellets in lieu of hay to stabled, RAO-affected horses was compared with the effect of feeding pellets and administering a 21-day decreasing dose regimen of oral dexamethasone on the expression (by kinetic polymerase chain reaction) of interleukin-8 (IL-8), chemokine (C-X-C motif) ligand 2 (CXCL2), IL-1beta, IL-6, and beta-actin in the BALF cells and of IL-8, CXCL2, 2 IL-1 receptor (IL-1R2), Toll-like receptor 4 (TLR4), and glyceraldehyde 3-phosphate dehydrogenase in the bronchial epithelium 2 days after the final dose. RESULTS: Both treatments reduced airway neutrophilia and breathing efforts but the addition of dexamethasone was associated with fewer treatment failures. Compared with feed changes alone, dexamethasone administration further reduced the expression of IL-8, CXCL2, and IL-1beta in the BALF cells 3.3-, 2.5-, and 4.7-fold, respectively. In the airway epithelium, both treatments were equally efficacious in reducing the expression of IL-8 and CXCL2 expression relative to pretreatment values, but either treatment failed to alter the expression of IL-1R2 and TLR4. CONCLUSIONS AND CLINICAL IMPORTANCE: For a rapid and consistent improvement in pulmonary function and a reduction in inflammatory gene expression of the BALF cells, a decreasing dose of oral dexamethasone in combination with feed alterations is more efficacious for horses that must remain stabled. 相似文献
