The neuropathology and host-parasite relationship of experimental infection with the RH-strain of Toxoplasma gondii were studied in 27 blue foxes (Alopex lagopus) aged 0 to 23 days, using light microscopy, including immunohistochemical staining, and transmission and scanning electron microscopy. All cases displayed multifocal necrotic lesions with numerous parasitic tachyzoites in the brain and spinal cord. The gray matter and the meninges were most seriously affected. Although a wide variety of cell types were parasitized, neurons and astrocytes seemed to be the main target cells. Individual parasitophorous vacuoles usually contained only a few tachyzoites, with rosette formations as a prominent feature. The present ultrastructural study supports the theory that the parasites actively invade the host cells by mechanisms that are different from those of phagocytosis. This is apparently the first report indicating that the formation of the network of tubular structures within the parasitophorous vacuole of T. gondii is associated with a transient, sack-like formation in the posterior end of the tachyzoites.  相似文献

Coho (Oncorhynchus kisutch) and chinook salmon (O. tschawyrscha) were reared in seawater pumped from the 600 m depth at the Natural Energy Laboratory of Hawaii. Fish were reared in either 100% deep water at 11 C, or a mixture of 55% deep and 45% surface waters at 17 C. Salmon flesh was analyzed for mercury, zinc, cadmium and lead. Mercury concentration in coho salmon reared at 11 C was significantly lower than the concentration in coho reared at 17 C (P < 0.05), but other heavy metal concentrations in tissue did not differ between the two species or between the two rearing temperatures. Compared with wild salmon from the northwest Pacific Ocean, salmon reared in Hawaii had significantly lower concentrations of mercury and lead (P < 0.05), but significantly higher concentrations of zinc and cadmium. These differences in heavy metal concentration between wild and cultured salmon were most readily explained by differences in the heavy metal content of diets and the lack of heavy metal concentrations in rearing water. The concentration of mercury in salmon tissue was at least 20 times less than the 1.0 μ/g U.S. Food and Drug Administration action level for mercury. Action levels have not been established for zinc, cadmium, and lead.  相似文献

Two feeding trials were conducted with juvenile largemouth bass Micropterus salmoides to evaluate alternative plant and animal source proteins for their ability to replace fish meal in practical diets. The first trial was designed to identify the most promising candidates. The second trial was conducted to evaluate how much of the fish meal could be replaced by those candidates. In Study 1, feed‐trained largemouth bass (3.1 ± 0.7 g) were randomly stocked into 18114‐L glass aquaria at 25 fish per aquarium. Fish were fed one of six experimental diets, each containing approximately 38% crude protein and 10% crude lipid, to apparent satiation twice daily. The control diet (CTL) contained 30% fish meal and 34.5% soybean meal. Diets 2–6 each contained 15% fish meal and at least 34.5% soybean meal with the remainder of the protein made up of either meat and bone meal (MBM), soybean meal (SBM), poultry by‐product meal (PBM), a 50150 mixture of blood meal and corn gluten meal (BM/CG), or 50150 mixture of hydrolyzed feather meal and soybean meal (FMISBM). There were three replicate aquaria per dietary treatment. After 12 wk, there was no significant difference (P > 0.05) among treatments in survival which averaged 92% overall. Only fish fed the PBM or BM/CG diets had average individual weights and feed conversion efficiencies that were not significantly different (P > 0.05) from the control diet (CTL). In Study 2, the formulation of the control diet (CTL) remained the same. Based on their performance in the first trial, PBM and BM/CG were chosen to now replace 75 or 100% of the fish meal. Fish were stocked at an average weight of 6.9 ± 1.7 g. After 11 wk, fish fed diets containing the BM/CG mixture at both levels were significantly smaller (P 5 0.05) than fish fed other diets and at 100% replacement survival was reduced. Fish fed diets containing poultry meal as the primary protein source performed as well as those fed the control diet (CTL). It appears that PBM can completely replace fish meal in diets for juvenile largemouth bass without adverse effects on growth, feed efficiency, or body composition.  相似文献

OBJECTIVE: To compare drilling, tapping, and screw-insertion torque, force, and time for the 4.5-mm AO and 6.5-mm Acutrak Plus (AP) bone screws, and to compare the mechanical shear strength and stiffness of a simulated complete lateral condylar fracture of the equine third metacarpal bone (MC3) stabilized with either an AO or AP screw. STUDY DESIGN: In vitro biomechanical assessment of screw-insertion variables, and shear failure tests of a bone-screw-stabilized simulated lateral condylar fracture. SAMPLE POPULATION: Eight pairs of cadaveric equine MC3s METHODS: Metacarpi were placed in a fixture and centered on a biaxial load cell in a materials-testing system to measure torque, compressive force, and time for drilling, tapping, and screw insertion. Standardized simulated lateral condylar fractures were stabilized by either an AO or AP screw and tested in shear until failure. A paired t test was used to assess differences between screws, with significance set at P < .05. RESULTS: Insertion and mechanical shear testing variables were comparable for AO and AP insertion equipment and screws. CONCLUSION: The 6.5-mm tapered AP screw can be inserted in equine third metacarpal condyles and is mechanically comparable with the 4.5-mm AO screw for fixation of a simulated lateral condylar fracture. CLINICAL RELEVANCE: Considering the comparable mechanical behavior, the potential for less-persistent soft-tissue irritation with the headless design, and the ability to achieve interfragmentary compression by inserting the screw in one hole drilled perpendicular to the fracture plane, the 6.5-mm tapered AP screw may be an attractive alternative for repair of incomplete lateral condylar fractures in horses.  相似文献

84 FeLV-positive cats were continually treated with the paramunity inducer (immunomodulator) PIND-AVI (23 healthy cats once a week, 61 diseased cats up to 3 times a week), the frequency depending on their general condition. The ELISA control tests were repeated after 4 to 6 weeks. Paramunization was continued in cats still viremic until remission of viremia. 21 healthy cats already reacted negatively to the first control ELISA, the 2 others after 8 weeks of paramunization. Within an observation time of 18 to 30 months there were no relapses. 49 of the 61 diseased cats (80.3%) reacted negatively to the first control. 5 cats (8.2%) had died at the beginning of the treatment. 5 cats (8.2%) showed a remission of viremic after being paramunized continually for 7 to 20 weeks. 2 cats (3.3%) remained viremic throughout the observation time (more than 2 years). None of the animals had a relapse. Neither had any of 9 chronically diseased cats, known to have been viremic for 3 to 12 months at the beginning of paramunization. The importance of the results of these paramunization tests for the treatment of various retrovirus infections of human beings and animals is briefly discussed.  相似文献

A five-month-old male domestic shorthair cat presented with a history of vomiting for 4 days. An ultrasound examination of the abdomen was performed as part of the diagnostic workup. The gallbladder appeared to be bilobate. The cat died the following day from renal failure. Paranasal meningioma was diagnosed in a 5-year-old Appaloosa gelding. The mass occupied the right maxillary, frontal, and sphenopalatine sinuses but did not invade the calvarium. The diagnosis was based on histologic evaluation, positive immunohistochemical staining for vimentin and cytokeratin, and ultrastructural features including the presence of interdigitating spindle cells with numerous desmosomes.  相似文献

Wild raptors brought into an ex situ environment often have poor semen quality that is further compromised by urine contamination. Generally, it is believed that in birds, artificial insemination into the cloaca or caudal vagina of females requires large doses of high-quality spermatozoa to maximize fertility. In an effort to define and overcome some of the challenges associated with reproduction in wild raptors, the objectives of this study were to 1) evaluate the frequency, impact, and remediation of urine contamination in fresh ejaculates for the purpose of maintaining sperm motility and viability in vitro, and 2) develop a deep insemination method that allows low numbers of washed sperm to be placed directly into the magnum to increase the probability of producing fertilized eggs. The species evaluated include golden eagle (Aquila chrysoetos), imperial eagle (A. adalberti), Bonelli's eagle (Hiernaetus fasciatus), and peregrine falcon (Falco peregrinus). Semen samples were collected and pooled by species, and a minimum of 25 pooled ejaculates per species were evaluated for urine contamination, pH, sperm viability, and sperm motility; the samples were either unwashed or washed in neutral (pH 7.0) or alkaline (pH 8.0) modified Lake's diluent. Female golden eagles and peregrine falcons were inseminated via transjunctional, intramagnal insemination with washed spermatozoa from urine-contaminated samples. Urine contamination occurred in 36.8 +/- 12.8% (mean +/- SEM) golden eagle, 43.1 +/- 9.1% imperial eagle. 28.7 +/- 16.1% Bonelli's eagle, and 48.2 +/- 17.3% peregrine falcon ejaculates. The pH in urine-contaminated semen samples ranged from 6.48 +/- 0.3 to 6.86 +/- 0.2, and in noncontaminated samples it ranged from from 7.17 +/- 0.1 to 7.56 +/- 0.1. Sperm viability and motility were reduced (P < 0.05) in all species for unwashed vs. washed sperm after 30 min incubation at room temperature. Two peregrine falcon chicks and one golden eagle chick hatched after intramagnal insemination. This study demonstrates that urine contamination, a common and lethal acidifier in manually collected raptor ejaculates, can be circumvented by immediate, gentle seminal washing. Furthermore, these processed sperm, when deposited by transjunctional intramagnal insemination, can produce live young.  相似文献

The influence of dietary protein, methionine, and lysine on leather quality in blue fox pelts was studied. The pelt material originated from animals in two consecutive feeding trials (Exp. 1 and Exp. 2) with three protein levels: conventional, slightly lowered, and very low. The two lowest protein diets were fed as such or as supplemented with methionine or with lysine (lysine only in Exp. 2). The following physical leather properties were measured: breaking load (BRL), tensile strength (TEN), relative elongation at break (PEB), straining of skins at pelting, and shrinkage at dressing. A decline in the dietary protein content reduced BRL and, hence, leather firmness, and increased straining and the corresponding shrinking in Exp. 1. The supplemented methionine tended to improve leather strength and elasticity by increasing TEN and PEB in Exp. 1, whereas lysine elicited no response. Methionine supplementation at the slightly lowered protein level increased BRL in both experiments by almost 10% as compared with the respective non-supplemented diet. We conclude that with high protein quality diets, a level of 200 g/kg DM (as digestible protein) appears to be adequate for producing pelts with firm, elastic leather, provided that an adequate amount of methionine is included in the diet.  相似文献
