The objective of this work was to study the changes that occur in the Leydig cells of rats exposed to continuous light. The laboratory rat is considered a non-photoperiodic species because exposure to short photoperiod has little or no effect on the reproductive status. However, exposure of adult female rats to constant light induces polycystic ovaries, indicating that extreme changes in the photoperiod affect the reproductive function seriously. Adult male rats were placed under continuous light conditions for a duration of 15 weeks. After this period, the animals were killed and testicles were dissected and processed by routine histologic protocols. Follicle-stimulating hormone (FSH) and luteinizing hormone (LH) serum levels were determined by radioimmunoassay (RIA). The visualization of antigens was achieved by the streptavidin-peroxidase immunohistochemical method. Antibodies against chromogranin A, S-100 protein, P substance, synaptofisin, neurofilament protein-200, gliofibrillary acidic protein and neurone-specific enolase were used. The mean LH serum concentration was significantly lower, while the mean FSH level was significantly higher in treated animals. The expression of S-100, NSE, CrA, SP and SYN was significantly lower in treated animals. In conclusion, the constant light exposure acting directly at the pituitary level decreases LH secretion. The increased FSH secretion may be due to a partial reduction of the negative androgen feedback in the pituitary gland. Moreover, the constant light exposure affects the expression of some immunomarkers in Leydig cells, possibly because of the changes found in the gonadotrophin level and feedback mechanism.  相似文献

Long-term agroecosystem experiments can be defined as large-scale field experiments more than 20 years old that study crop production, nutrient cycling, and environmental impacts of agriculture. They provide a resource for evaluating biological, biogeochemical, and environmental dimensions of agricultural sustainability; for predicting future global changes; and for validating model competence and performance. A systematic assessment is needed to determine the merits of all known experiments and to identify any that may exist in tropical and subtropical environments. The establishment of an international network to coordinate data collection and link sites would facilitate more precise prediction of agroecosystem sustainability and future global change.  相似文献

Increased labor costs and reduced labor pools for hop production necessitate the development of strategies that improve efficiency and automation of hop production. One solution for reducing labor inputs is the use of “short-trellis” hop varieties. Unfortunately, little information exists on the genetic control of this trait in hop, and there are no known molecular markers available for selection. This preliminary study was enacted to identify QTLs associated with expression of short-stature growth phenotype using SNPs identified within genome-assembled scaffolds. A bi-parental mapping population of 87 offspring was obtained from the cross, “Pioneer × 25/95/15”. Genotyping-by-sequencing was performed on parents and offspring. SNPs were identified using TASSEL v3.0 with either ‘Teamaker’ reference genome or ‘Shinsuwase’ genome. The genetic map derived from ‘Teamaker’ SNPs was far superior and was used for all further analysis. QTL analysis identified eight QTLs linked to short stature with five showing strong statistical association based upon three different statistical analyses. All eight QTLs were found on linkage group one. Evaluation of scaffolds containing SNP markers located at or surrounding QTL regions (±1 cM) identified 67 putative genes—several of which are known structural genes. A genome-wide scan of SNP markers identified an additional marker found on a scaffold containing a putative gene (Aspartyl protease family protein) known to induce dwarf characteristics in other species. Further validation of significantly associated markers on different populations is necessary prior to implementation in marker-assisted selection.  相似文献

The aim of this study was to characterize cytoskeletal intermediate filament proteins and glycoconjugates of syncytiotrophoblast, cytotrophoblast and decidual cells of feline endotheliochorial placenta. Samples from 12 normal pregnant female cats, after 45 ± 5 days of gestation, were obtained removing the uterine horns by hysterectomy. Sections were processed for routine observation and for immunohistochemistry using anticytokeratin, antivimentin and antidesmin antibodies. In addition, lectin histochemistry was performed using a panel of several biotinylated lectins to characterize glycosides expression profile. Cytotrophoblast and syncytiotrophoblast showed immunoreactivity only with acidic and basic cytokeratins. Decidual cells were only positive to vimentin, consistent with their origin from endometrial fibroblasts. Trophoblast expressed a broad population of glycans, highly exposing terminal N‐acetyl glucosamine residues and non‐sialylated galactose and N‐acetyl galactosamine oligomers. Oligosaccharides bound by Phaseolus vulgaris erythroagglutinin were the only highly branched Nlinked residues evidenced in cats, and they were restricted to the syncytium. Unlike results reported on humans, mice and rats on lectin affinity of decidual cells, sialid acids and complex Nlinked oligosaccharides were not demonstrated in cats. Glycosylation of proteins determines many of their final properties, thus becoming essential for the embryo‐maternal dialogue during implantation and placentation. Changes in glycosylation pattern have been related to pathological pregnancies in other species. Hence, the knowledge about glycosylation profile of the normal cat placenta may lead to a better understanding of both normal and pathological reproductive events.  相似文献

Dryland ecosystems have long been considered to have a highly heterogeneous distribution of nutrients and soil biota, with greater concentrations of both in soils under plants relative to interspace soils. We examined the distribution of soil resources in two plant communities (dominated by either the shrub Coleogyne ramosissima or the grass Stipa hymenoides) at two locations. Interspace soils were covered either by early successional biological soil crusts (BSCs) or by later successional BSCs (dominated by nitrogen (N)-fixing cyanobacteria and lichens). For each of the 8 plant type×crust type×locations, we sampled the stem, dripline, and 3 interspace distances around each of 3 plants. Soil analyses revealed that only available potassium (Kav) and ammonium concentrations were consistently greater under plants (7 of 8 sites and 6 of 8 sites, respectively). Nitrate and iron (Fe) were greater under plants at 4 sites, while all other nutrients were greater under plants at less than 50% of the sites. In contrast, calcium, copper, clay, phosphorus (P), and zinc were often greater in the interspace than under the plants. Soil microbial biomass was always greater under the plant compared to the interspace. The community composition of N-fixing bacteria was highly variable, with no distinguishable patterns among microsites. Bacterivorous nematodes and rotifers were consistently more abundant under plants (8 and 7 sites, respectively), and fungivorous and omnivorous nematodes were greater under plants at 5 of the 8 sites. Abundance of other soil biota was greater under plants at less than 50% of the sites, but highly correlated with the availability of N, P, Kav, and Fe. Unlike other ecosystems, the soil biota was only infrequently correlated with organic matter. Lack of plant-driven heterogeneity in soils of this ecosystem is likely due to (1) interspace soils covered with BSCs, (2) little incorporation of above-ground plant litter into soils, and/or (3) root deployment patterns.  相似文献
