Previous studies have demonstrated inconsistent results on the impact of tillage systems on nitrogen (N) losses from field-applied manure. This study assessed the impact of no-tillage (NT) and conventional tillage (CT) systems on gaseous N losses, N2O:N2O + N2 ratios and NO3−-N leaching following surface application of cattle manure. The study was undertaken during the 2003/2004 and 2004/2005 seasons at two field sites in Nova Scotia namely, Streets Ridge (SR) in Cumberland County and the Bio-environmental Engineering Centre (BEEC) in Truro. Results showed that the NT system had higher (p < 0.05) NH3 losses than CT. Over the two seasons, manure incorporation in CT reduced NH3 losses on average by 86% at SR and 78% at BEEC relative to NT. At both sites and during both seasons, denitrification rates and N2O fluxes in NT were generally higher than in CT plots, presumably due to higher soil water and organic matter content in NT. Over the two seasons, mean denitrification rates at SR were 239 and 119 g N ha−1 d−1, while N2O fluxes were 120 and 64 g N ha−1 d−1 under NT and CT, respectively. At BEEC mean denitrification rates were 114 and 71 g N ha−1 d−1, while N2O fluxes were 52 and 27 g N ha−1 d−1 under NT and CT, respectively. Conversely, N2O:N2O + N2 ratios were lower in NT than CT suggesting more complete reduction of N2O to N2 under NT. When averaged across all soil depths, NO3−-N was higher (p < 0.05) in CT than NT. Nitrate-N decreased with depth at both sites regardless of tillage. In most cases, NO3−-N was higher under CT than NT at all soil depths. Similarly, flow-weighted average NO3−-N concentrations in drainage water were generally higher under CT. This may be partly attributed to higher denitrification rates under NT. Therefore, NT may be a viable strategy to remove NO3−-N from the soil, and thus, reduce NO3−-N contamination of groundwater. However, it should be noted that while the use of NT reduces NO3−-N leaching it may come with unintended environmental tradeoffs, including increased NH3 and N2O emissions. 相似文献

Our study objective was to determine the ability of milk urea-nitrogen concentrations ([MUN]) to predict fecal nitrogen concentrations ([Fecal N]) in commercial dairy herds. A total of 83 dairy herds were each visited 3 times within 48 h after a monthly herd milk test. For each farm visit, forages were sampled for nutrient analyses, which were entered into a computerized ration evaluator, and fecal samples were taken per rectum from each of 6 cows (2 early-, 2 mid-, and 2 late-lactation). Fecal samples were pooled, mixed, and analyzed for nitrogen content. Fecal nitrogen concentrations were compared with the routinely measured nutritional parameters from the ration evaluation, and the herd average [MUN] for the previous milk test date using mixed linear regression analyses. Total protein supplied in the ration was significantly positively associated with [Fecal N], but herd average [MUN] was not associated (P > 0.10) with [Fecal N]. 相似文献

Transportation causes stress in cattle that may alter numerous physiological variables with a negative impact on production and health. The objectives of the current study were to investigate the physiological effects of truck transportation and to characterize a pattern of phenotypes in the circulation that may aid in the early identification of stress-susceptible animals that often succumb to severe respiratory disease. Thirty-six young beef bulls (Aberdeen Angus, n = 12; Friesian, n = 12; and Belgian Blue x Friesian, n = 12) were subjected to a 9-h truck transportation by road. Blood (10 mL) was collected at -24, 0, 4.5, 9.75, 14.25, 24, and 48 h relative to the initiation of transportation (0 h). Plasma was collected for the assay of various metabolic, inflammatory, and steroid variables, and total leukocyte counts were determined in whole blood at each time point. Body weight and rectal temperature were recorded at -24, 9.75, and 48 h. Transportation decreased measures of protein metabolism in the plasma, including albumin (P = 0.002), globulin (P < 0.001), urea (P = 0.006), and total protein (P < 0.001), and increased creatine kinase (P < 0.001). The energy substrate beta-hydroxybutyrate was not changed (P = 0.27). Acute phase proteins haptoglobin and fibrinogen were both decreased (P < 0.001), whereas total leukocyte counts were elevated (P = 0.002). Circulating steroid concentrations were altered, because a classical acute increase in plasma cortisol was observed with the onset of transit (P < 0.001), in association with a decrease in dehydroepiandrosterone (P = 0.07), resulting in a profound increase in cortisol:dehydroepiandrosterone ratio (P < 0.001). Plasma testosterone was decreased, whereas plasma progesterone was increased (P < 0.001) in association with the increase in cortisol (P < 0.001). There was also an effect of breed for all variables except plasma urea, creatine kinase, and testosterone, perhaps indicating that a genetic component contributed to the physiological response to transportation stress, although without any clear trend. Taken together, this profile of physiological variables in the circulation of transportation-stressed bulls may aid in the future detection of disease-susceptible cattle after transportation. Maintenance of high levels of realized fertility (defined as the percentage of ewes that lamb) and appropriate levels of fecundity are critical for efficient sheep production. The optimal level of fecundity in most situations is well below the maximum attainable level and can be targeted by combining selection among and within breeds with use of an expanding array of single-gene mutations affecting ovulation rate and litter size. The heritability of litter size is approximately 0.10, allowing changes of up to 2%/year from simple mass selection. Mutations in several genes associated with the transforming growth factor β superfamily ( BMPRIB , GDF9 and sex-linked BMP15 ) can increase ovulation rates by 0.7–1.5 ova in heterozygous ewes. However, ewes that are homozygous for BMP15 or GDF9 mutations are sterile, so use of these mutations requires carefully structured breeding programmes. Improvements in fertility may be critical for autumn lambing or programmes that aspire to lamb throughout the year. Selection to improve fertility in spring matings has been successful; selected adult ewes have lambing rates of 80–85% in October and early November. The selected ewes have a dramatically reduced seasonal anestrus, and many ewes continue to cycle during spring and summer. Major genes affecting seasonal breeding have not been identified in sheep. Polymorphisms in the melatonin receptor 1a gene appear to be associated with seasonal breeding in some, but not all breeds. However, functional genomic studies of genes associated with circadian and circannual rhythms have potential to reveal additional candidate genes involved in seasonal breeding. 相似文献

During an on-going amphibian ecology study, a free-ranging American bullfrog (Rana catesbeiana) metamorph was captured in a pitfall trap adjacent to a constructed farm pond at the Plateau Research and Education Center (PREC) on the Cumberland Plateau near Crossville, Tennessee, USA. Grossly, the right eye was approximately 50% the size of the left. Stereo and light microscopic examination revealed two granulomas within the orbit. Electron microscopic examination revealed virus particles scattered throughout one structure but mostly aggregated toward the center. Subsequent PCR and sequencing (GenBank accession Number EF175670) confirmed frog virus 3 (FV3). This represents the first report of a malformation in an anuran associated with FV3. 相似文献

Our objective of this study was to investigate the relationship between milk urea-nitrogen concentrations ([MUN]) and first-service breeding success (FSBS) in a large number of commercial dairy herds, using various timings on [MUN]. All commercial dairy herds in Prince Edward Island on monthly milk testing (n=198) formed the sampling frame. Milk components, [MUN], 24-h milk production, and breeding data for all cows from these farms were gathered electronically from a central database. A first service between 1 June 1999 and 31 May 2000 was classified successful (FSBS=1) if it was the cow's last service and she calved 270-290 d later. Mixed logistic-regression modeling was used to determine the association between FSBS (the outcome variable) and the [MUN] closest to first service, controlling for other possible confounders and clustering effects of cows within the study herds. The final dataset included 2787 successful and 3015 unsuccessful first services. A change in [MUN] on the test closest to first service from 10 to 20 mg/dL was associated with a 13.9% reduction in the odds of FSBS (controlled for parity, milk production and days in milk). 相似文献

It has been proposed that hydrogen produced from basalt-ground-water interactions may serve as an energy source that supports the existence of microorganisms in the deep subsurface on Earth and possibly on other planets. However, experiments demonstrated that hydrogen is not produced from basalt at an environmentally relevant, alkaline pH. Small amounts of hydrogen were produced at a lower pH in laboratory incubations, but even this hydrogen production was transitory. Furthermore, geochemical considerations suggest that previously reported rates of hydrogen production cannot be sustained over geologically significant time frames. OBJECTIVES: To confirm the hydrophobicity of the luminal surface of the canine stomach and to elucidate the ultrastructure of the lining imparting that property. DESIGN AND PROCEDURES: Gastric and duodenal mucosal samples from eight dogs were collected immediately after euthanasia and subjected to contact angle measurement using a goniometer. Other samples were examined by electron microscopy following a fixation procedure known to preserve phospholipids and oligolamellar structures. RESULTS: Contact angles for the canine gastric mucosal surface (85.1 +/- 5.5) were significantly greater (P < 0.0001) than for the duodenum (24.0 +/- 1.7). Electron microscopy revealed the existence of surfactant as abundant osmiophilic phospholipid material within the gastric and duodenal mucosae. CONCLUSION: We have confirmed the hydrophobic nature of the canine gastric mucosa whereas the luminal surface of the duodenum is hydrophilic. We propose that the water-repellent nature of the canine gastric lining contributes to the 'gastric mucosal barrier' and is imparted by an oligolamellar layer of surface-active phospholipid ('gastric surfactant') adsorbed to the surface. Both gastric and duodenal mucosae may also utilise phospholipids as an intercellular defense mechanism in the event that tight junctions are breached by acid. It is tempting to speculate that a deficiency of gastric phospholipids may predispose dogs to ulcers. Objective To develop a lameness model to assess the efficacy of analgesics for alleviating pain, swelling and systemic signs of inflammation in sheep. Procedures The response to subcutaneous injection of 0.1 or 0.2 mL turpentine in a forelimb pastern (n = 4 ewes per dose) was examined at 0, 3, 6, 24, 48 and 72 h. In a second experiment, responses were measured at 0, 2, 4, 6, 8, 10, 12 and 24 h in ewes receiving 0.1 mL turpentine ± meloxicam 1 mg/kg IV at 0 h (n = 6 per group). Responses measured included forceplate pressure, skin temperature, limb circumference, nociception, leucocyte count, neutrophil : lymphocyte ratio, haptoglobin and daily feed intake. Results Turpentine injection caused a decrease in weight borne on the treated limb, increased skin temperature, increased sensitivity at the injection site and leucocytosis by 2 h and increased limb circumference by 4 h. Weight borne and sensitivity of the injected limb returned to control levels after around 24 h, whereas tissue swelling, elevated skin temperature and elevated haptoglobin levels persisted for at least 72 h. Treatment with meloxicam improved weight borne by and tolerance to pressure exerted on the turpentine‐injected limb. Conclusions The local and systemic signs of inflammation and pain, temporary reduction in function of the affected limb and partial amelioration of some of these changes by the dose of meloxicam used here suggest that injection of turpentine in the lower forelimb provides a suitable model for examining the efficacy of analgesics for alleviation of pain and inflammation in sheep. 相似文献
