Grapevine inflorescences (cv. Chardonnay) were found to be susceptible to infection by the berry rotting pathogens Colletotrichum acutatum and Greeneria uvicola responsible for ripe rot and bitter rot of grapes respectively. Infection of inflorescences on field-grown grapevines at mid-flowering led to subsequent berry rot at veraison. An application of the strobilurin fungicide Cabrio (active ingredient pyraclostrobin) at flowering reduced the incidence of ripe rot and bitter rot at veraison from 88% to 0% and from 86% to 2%, respectively. The infection of detached inflorescences was influenced by temperature and was greatest at 25–30°C for C. acutatum and 30°C for G. uvicola. Our results demonstrate for the first time that grapevine flowers are susceptible to C. acutatum and G. uvicola and that flower infections have the potential to lead to subsequent rotting of the grape berries. The findings have implications for the management of ripe rot and bitter rot of grapes.  相似文献

The correct measurement of spermatic concentration in boar semen is one of the basic tasks routinely performed at the Swine Artificial Insemination Center (CIAS). This article explains two experiments whose objectives were: Experiment 1: This trial had two objectives: (1) to evaluate the precision of the spermdensimeter (SPM) with relation to the Neubauer count chamber (CN) which is considered the standard technique in the field and (2) to correct the scale on the SPM table. Experiment 2: This trial also had two objectives: (1) to evaluate the time and repetitions required in the SPM and (2) to compare the SPM adjusted table produced in Expt 1 with the SPM standard table. The methods in Expt 1 were used to measure total spermatic concentration in 141 boar ejaculates. The average spermatic concentrations (10(6) spermatozoids/ml) and average dose number were 229.1 and 22.6 for CN, and 283.6 and 28.0 for SPM. These results indicate that the SPM has a tendency to overestimate spermatic concentration with relation to the CN. The SPM produced a smaller coefficient of variation and needed lesser examination time than the CN (p < 0.05). The SPM adjusted table created in Expt 1 produced similar results as the SPM and CN standard tables (2.96 billion live spermatozoids/dose vs 2.36 billion live spermatozoids/dose). Incorporation and evaluation of supplemental data from other CIAS regarding the use of the SPM adjusted table in the field entitled 'Adjusted Table for Karras Spermdensimeter LPS-FMVZ-USP' would be valuable to prove its efficiency.  相似文献

The identity of Colletotrichum acutatum as the causal pathogen of grape ripe-rot, which causes yield loss and a bitter taint that lowers wine quality in Australian subtropical wine-grape regions, was confirmed using species-specific primers. Cultural, morphological and molecular methods (RAPD-PCR and sequencing of parts of the 5·8S-ITS regions and the β-tubulin-2 gene) were used to determine the phylogenetic relationships of Australian C. acutatum isolates from wine grapes and other horticultural crops. A combination of RAPD-PCR and β-tubulin-2 gene data showed that all wine-grape ripe-rot isolates from northern regions of New South Wales (NSW) and Queensland belong to a proposed new C. acutatum group (A9), together with isolates from Australian strawberry, mango, blueberry and olive. The 5·8S-ITS sequences for these grape pathogens were identical to published sequences for an isolate from Cyclamen (the Netherlands) and differed by 1 bp from isolates from Capsicum (Taiwan) and orange (Costa Rica). The grape ripe-rot isolates from the Shoalhaven Valley (southern NSW) were clustered within two other C. acutatum groups: A2 and A5. In vitro infection studies showed that Australian C. acutatum isolates from almond, blueberry, chilli, grape, mango, olive, strawberry and tomato were able to infect grape and could also infect blueberry and strawberry, indicating a lack of host specificity. This lack of host specificity, the genetic similarity with non-grape isolates, and the fact that many of the non-grape hosts were isolated from wine-growing regions, suggest the potential for cross-infection between grape and other horticultural crops.  相似文献

OBJECTIVE: To determine the prevalence of dorsal metacarpal disease in 2-year-old Thoroughbred horses by prerace inspection and to evaluate the effectiveness of a dorsal metacarpal disease detection program on the predictability of race performance (finishing in the order predicted by punters) and the distance between the first- and last-placed horse in a field. DESIGN: A retrospective study using client-owned horses. METHODS: A program of prerace detection of dorsal metacarpal disease using digital palpation of the dorsal aspect of the third metacarpal bone of both forelimbs of 2-year-old Thoroughbreds racing at Ascot racecourse was instituted by the Western Australian Turf Club in December 1995. Horses that had significant pain reaction to digital palpation were considered to have pain due to dorsal metacarpal disease, withdrawn from that day's race and not permitted to race for 6 weeks. The race records of all 2-year-old Thoroughbreds racing at Ascot racecourse from December 1993 until December 1995 were compared with the race results from December 1995 until March 1997, after initiation of the dorsal metacarpal disease detection program. RESULTS: After initiation of the program, horses were racing more predictably over 1100 m (P = 2.09) and 1200 m (P = 3.48). These horses finished closer to the winner over 1100 m (P = 3.39) and over 1200 m (P = 3.13). In 1000 m races, the trend for horses to race more predictably (P = 1.07) and for there to be less distance between first and last (P = 1.72) was not statistically significant. CONCLUSIONS: The program of prerace digital palpation for dorsal metacarpal disease was considered to be beneficial in 2-year-old Thoroughbreds racing over 1100 m and 1200 m, in that they raced more predictably.  相似文献

Plasma levels of fenbendazole (FBZ) and its sulphoxide (OFZ) and sulphone (FBZ.SO2) metabolites were measured in goats and sheep during low-level administration of FBZ given by intraruminal infusion or formulated into a urea-molasses feed supplement block (UMB). In experiment 1, 6 goats and 6 sheep were offered UMB containing 0.5 g FBZ/kg (MUMB) and individual block consumption was measured daily for 18 days. In experiment 2, some of the same animals (n=4 for each species) received FBZ by intraruminal infusion at 1, 1.5 and 3 mg/kg liveweight per day for 7 days at each dosage. FBZ, OFZ and FBZ.SO levels were determined in plasma collected every 3 days in experiment 1 and on days 4, 5 and26 of each infusion period in experiment 2. In both experiments, higher equilibrium levels were observed for the three metabolites in sheep than in goats. Significant linear relationships were observed between the daily FBZ dosages and the plasma levels of the three metabolites in both species. The regression coefficients were significantly higher in sheep than in goats for FBZ and OFZ but not for FBZ.SO2, and they were also significantly higher during MUMB administration than during infusion for all three metabolites in both species. FBZ is a suitable anthelmintic for incorporation into a MUMB formulation for use in livestock production systems where responses to molasses urea supplementation have been demonstrated and gastrointestinal parasitism impairs productivity. Infection of young roots of maize (Zea mays L.) by the parasitic plant Striga hermonthica (Del.) Benth. was examined. Attachment to and penetration of roots occurred within 1–2 days after inoculation. Subsequent growth through the cortex to the host stele and proliferation of parasite xylem tissue was commonly completed by 3–4 days after inoculation. Histochemical staining showed that young maize roots do not contain major wall-thickening components. However, an increase in cell wall fluorescence and endodermal cell wall thickness was often seen at the site of infection and in the surrounding maize root tissue at 3 days after inoculation. This host response was variable and did not prevent rapid and successful penetration by the parasite. In contrast, uninfected roots of Pennisetum setosum (Sw.) L. Rich., a species resistant to S. hermonthica, had substantial thickening of the inner endodermal cell walls and exhibited further cell wall thickening at the stele upon infection. Examination of infections on both hosts demonstrated the presence of autofluorescent material at the host-parasite interface. This material was thicker and more extensive at the P. setosum-S. hermonthica interface than at the maize-S. hermonthica interface, and contained polyphenols and lignin. Examination of the host-parasite xylem connections in maize revealed substantial invasion of the host stele by both parenchyma and tracheary elements. In a few cases of P. setosum infection, parasite cells entered the stele; however, this did not lead to successful establishment of the parasite.  相似文献

A 4-year-old Thoroughbred gelding with severe left hindlimb lameness and colic-like signs lasting 30–60 min post-exercise was diagnosed with unilateral internal aorto-iliac thrombosis. The thrombus was identified using transrectal ultrasonography in the terminal aorta and was initially thought to extend into the left external iliac artery. However, attempted thrombectomy via a left femoral arteriotomy performed under general anaesthesia using thrombectomy and angioplasty catheters was unsuccessful, as the thrombus was located in the left internal iliac artery. Transrectal ultrasound evaluation was repeated monthly for 12 months post-operatively during which time the gelding remained asymptomatic at rest. As removal of thrombi in the external but not internal iliac artery may be possible, correct diagnosis of the location of the thrombus is critical in preventing unnecessary attempts at surgical correction. The aetiology remains unknown, but altered haemodynamics within the abdominal aorta with mechanical stresses during intense exercise are proposed as a cause of thrombus formation. The sequential monitoring of this case also conflicts the assumption that thrombi in this location propagate over time as partial regression was observed 6 months after referral and 9 months after onset of clinical signs.  相似文献
