The adenosine monophosphate (AMP)-activated protein kinase (AMPK) regulates whole-body and cellular energy balance in response to energy demand and supply. AMPK is an αβγ heterotrimer activated by decreasing concentrations of adenosine triphosphate (ATP) and increasing AMP concentrations. AMPK activation depends on phosphorylation of the α catalytic subunit on threonine-172 (Thr(172)) by kinases LKB1 or CaMKKβ, and this is promoted by AMP binding to the γ subunit. AMP sustains activity by inhibiting dephosphorylation of α-Thr(172), whereas ATP promotes dephosphorylation. Adenosine diphosphate (ADP), like AMP, bound to γ sites 1 and 3 and stimulated α-Thr(172) phosphorylation. However, in contrast to AMP, ADP did not directly activate phosphorylated AMPK. In this way, both ADP/ATP and AMP/ATP ratios contribute to AMPK regulation.  相似文献

Cows were vaccinated simultaneously by the intramuscular and intramammary route with formolised Staphylococcus aureus cells of strains BB, Mexi and 3528. Vaccination resulted in slight increases in serum agglutinin titres, but the levels of the agglutinins in milk and colostrum were not higher in vaccinated cows than in the unvaccinated controls. Vaccination did not result in higher levels of IgM, IgG1, IgG2 or IgA immunoglobulins in serum, colostrum or milk as compared with controls. Vaccinated cows, particularly those in which strain 3528 was used, showed some resistance to infection following challenge with low numbers of viable S aureus Mexi, but there was no resistance to infection when similar numbers of the virulent BB strain were used for challenge. It is concluded that vaccination is unlikely to prevent infection of the bovine udder by S aureus.  相似文献

Over the last three decades, farming systems in Europe and Australia have seen a decline in legume plantings, leading to reduced soil carbon and fertility, and an increase in plant disease, reliance on industrial nitrogen fertilizer and herbicides. In Australia, one reason for this decline has been the movement towards sowing crops and forages into dry soil, before the opening rains, as a consequence of climate variability. This practice predicates against the survival of rhizobial inoculants, and hence generates uncertainty about legume performance. The research reported here was initiated to improve the robustness of a specific forage legume/rhizobia symbiosis to increase nitrogen fixation in low pH, infertile soils. Rhizobial strains (Rhizobium leguminosarum biovar viciae) from Pisum sativum L. were sourced from acid soils in southern Italy and southern Australia. Strains were evaluated for N fixation on the forage legumes P. sativum, Vicia sativa and Vicia villosa, then for survival and persistence in acid soils (pHCa 4.6). Fourteen of the strains produced a higher percentage of nitrogen derived from the atmosphere (%Ndfa) compared to commercial comparator strain SU303 (<78%). Twenty‐two strains survived sufficiently into the second season to form more nodules than SU303, which only achieved 3% of plants nodulated. Elite strains WSM4643 and WSM4645 produced six times more nodulated plants than SU303 and had significantly higher saprophytic competence in acid soil. These strains have the ability to optimize symbiotic associations with field peas and vetch in soils with low fertility, carbon and pH that are restrictive to the current commercial strain SU303.  相似文献

Climate variability and current farming practices have led to declining soil fertility and pH, with a heavy reliance on fertilizers and herbicides. The addition of forage and grain legumes to farming systems not only improves soil health but also increases farm profitability through nitrogen (N) fertilizer cost offsets. However, the formation of effective symbioses between legumes and rhizobia can be unreliable and is considered at risk when combined with dry sowing practices such as those that have been designed to obviate effects of climate change. This research was initiated to improve the robustness of the legume/rhizobia symbiosis in low pH, infertile and dry soils. Production from two cultivars of field pea (Pisum sativum) and two species of vetch (Vicia spp.), and symbiotic outcomes when inoculated with a range of experimental rhizobial strains (Rhizobium leguminosarum biovar viciae), was assessed in broad acre field trials which simulated farmer practice. New rhizobia strains increased nodulation, N fixation, produced more biomass and higher seed yield than comparator commercial strains. Strain WSM4643 also demonstrated superior survival when desiccated compared to current commercial strains in the laboratory and on seed when delivered as inoculant in peat carriers. WSM4643 is a suitable prospect for a commercial inoculant in Australia and other agricultural areas of the world where growing peas and vetch on soils generally considered problematic for this legume/rhizobia symbiosis. A particular advantage of WSM4643 may be that it potentiates sowing inoculated legumes into dry soil, which is a contemporary response by farmers to climate variation.  相似文献

We describe movement patterns of hatchery-raised, juvenile, spring chinook salmon, Oncorhynchus tshawytscha, using a two-state Markov chain model. The existence of two states, moving and holding, is suggestedby anecdotalinformation from a large radio telemetry study; yet adequate observations of these small-scale fish behaviors are not available for estimating transition probabilities directly. Instead, we estimate the transition probability matrix from travel times within each of 11 river segments using a method of moments approach. Bootstrapped confidence intervals are presented. Results suggest that fish behavior in the region of the confluence between the Grande Ronde and Snake Rivers includes many transitions between moving and staying while fish behavior in the Snake River is more likely to include long periods of staying.  相似文献

CASE HISTORY: Eight mohua, or yellowheads (Mohoua ochrocephala), were held in a large open aviary over the summer months of 2003–2004, following their capture for captivebreeding purposes. Two birds died of transportation trauma shortly after arrival, one became ill and died a month later, and another four died within a 2-week period in February 2004. The eighth bird also became ill at this time but survived for a year following treatment with chloroquine and doxycycline.

CLINICAL AND PATHOLOGICAL FINDINGS: The affected birds were depressed, lethargic and dyspnoeic. Necropsy of three birds showed a slightly pale and swollen liver and spleen. Impression smears of the liver of one bird revealed schizonts resembling Plasmodium spp. within the cytoplasm of many hepatocytes, which was confirmed histopathologically. Similar protozoal organisms were seen within splenic histiocytes and pulmonary endothelial cells of 5/6 birds. Electron microscopy identified these as protozoal schizonts containing merozoites of similar size and structure to those of Plasmodium spp.

DIAGNOSIS: The birds were infected with a protozoal haemoparasite resembling Plasmodium spp.; asexual stages within hepatocytes and endothelial cells of the lung and spleen were typical of this organism.

CONCLUSIONS AND CLINICAL RELEVANCE: The mohua captured from west Otago were highly susceptible to avian malaria as they came from an isolated population that was likely to be naïve and have had no previous contact with this organism. The birds were probably infected by bites from mosquitoes feeding off local populations of blackbirds subsequently found to be infected with Plasmodium spp.  相似文献

Water temperature can have a profound influence on development and distribution of aquatic species. Salmon are particularly vulnerable to temperature changes because their reproductive and early development life phases are spent in freshwater river systems where temperature fluctuates widely both daily and seasonally. Flow regulation downstream of dams can also cause temperature regime changes, which in turn may spur local adaptation of early life‐history traits. In a common garden laboratory incubation experiment, we exposed spring Chinook salmon (Oncorhynchus tshawytscha) embryos to four temperature regimes: warm stable, cold stable, daily variation and below dam. We found that fry from warmer thermal regimes emerged earlier than those from colder regimes both in terms of calendar date and temperature units and that warmer temperatures caused fry to emerge less developed. There was also a significant effect of family on both emergence timing and development level at emergence. By combining measurements of physiological and behavioural traits at emergence and interpreting them within a reaction norm framework, we can better understand which populations might be more vulnerable to altered thermal regimes.  相似文献

The European Phytosanitary Research and Coordination network (Euphresco, www.euphresco.net ) is a network of organisations which were initiated as EU funded ERA‐NET projects in 2006 and 2010. The goal of the network is to strengthen the networking of research activities carried out at national or regional level and the mutual opening of research programmes. In 2014, having received EU funding twice, Euphresco members decided to continue to work together as a long‐term and self‐sustaining network. The coordination of Euphresco is now hosted within the European and Mediterranean Plant Protection Organization (EPPO), and in addition to consolidating the network and its procedures, it is also preparing the ground for future developments.  相似文献
