基于高通量测序的数字基因表达谱技术研究进展

数字基因表达谱(digital gene expression profiling,DGE)是一种全面、快速地检测某一物种特定组织在特定状态下基因表达情况的高通量测序技术。通过对基因表达谱的生物信息学搜索、比较和分析,可从中获取基因转录、基因调控、信号转导通路、蛋白质功能等相关信息。该研究主要介绍了数字基因表达谱的原理以及在生物学和医学研究等方面的应用。     

白菜叶和茎中微量元素Cu、Mn含量的测定

为研究白菜的食品应用及工业应用价值,采用HNO3-HCl O4(4∶1)湿法消解样品,火焰原子吸收光谱法(FAAS)测定白菜叶和茎中微量元素Cu、Mn的含量。试验结果表明,白菜叶和茎中Mn、Cu含量存在明显差异,Mn的含量均大于Cu的含量,且这两种元素在叶中的含量均高于茎。     

Effects of microglia-derived IL-1β on differentiation of OPCs in corpus callosum of septic neonatal rats

AIM: To explore whether IL-1β inhibits the oligodendrocyte precursor cell (OPCs) differentiation and affects axonal myelination. METHODS: One-day-old SD rats were randomly divided into control group and LPS group (48 rats in each group). The rats in LPS group were intraperitoneally injected with 1 mg/kg LPS. The rats in control group were injected with an equal volume of PBS. The rats in each group were further divided into 3 h, 24 h, 3 d, 7 d, 14 d and 28 d subgroups after injection. The expression of IL-1β and IL-1R₁ in the rat corpus callosum at 3 h, 24 h, 3 d, 7 d was determined by double immunofluorescence and Western blotting. The myelin basic protein(MBP) expression in the rat corpus callosum at 14 d, 28 d after injection was also measured. In vitro, primary OPCs culture was performed and divided into control group, 30 μg/L IL-1β group, 30 μg/L IL-1β+IL-1Ra group and 30 μg/L IL-1Ra group. The expression of MBP in the OPCs induced differentiation for 3 d was observed by double immunofluorescence and Western blotting. RESULTS: The expression of IL-1β and IL-1R₁ in the rat corpus callosum at 3 h, 24 h, 3 d, 7 d after LPS injection was obviously increased and the expression of MBP in the rat corpus callosum at 14 d, 28 d in LPS group was obviously decreased compared with control group in vivo. The level of MBP was significantly decreased after IL-1β treatment for 3 d in vitro. However, IL-1Ra (IL-1R inhibitor) reversed the down-regulation of MBP expression. IL-1β inhibited the expression of p-ERK, ERK over-expression reversed the down-regulation of MBP expression compared with IL-1β group. CONCLUSION: IL-1β inhibits the differentiation of OPCs, which may be involved in ERK pathways, thus leading to axonal hypomyelination in the corpus callosum of septic neonatal rats.     

温室黄瓜新品种津优335号的选育

津优335号是以自交系A143-5为母本,以自交系B74A-7为父本配制而成的黄瓜一代杂种。耐低温弱光能力强,植株长势强,雌花节率高、连续结瓜能力强,瓜条生长速度快。瓜条商品性突出,品质好。抗枯萎病,中抗褐斑病、霜霉病和白粉病。越冬栽培不歇秧,单瓜质量约220g,总产量20000kg·(667m2)-1左右,适宜我国北方地区日光温室各茬口栽培。     

辣椒新品种海丰1052

海丰1052是以08-123为母本,F07-253-2-1为父本配制的早熟鲜食线椒一代杂种。始花节位为第7~8节,果实顺直,果面光滑,果肩略皱,辣味较强,青熟果亮绿色,老熟果鲜红色,果实纵径31.2cm,横径1.5cm,平均单果质量为32.6g,VC含量为672mg·kg-1,可溶性糖含量1.7g·kg-1,蛋白质含量为14.0g·kg-1,辣椒素含量为0.25g·kg-1。抗CMV、TMV,中抗疫病。一般每667m2产量3000kg左右。适合北方露地、秋延后大棚栽培,广东、广西、山西、陕西等地露地栽培。     

Cx43 is involved in electrical remodeling of atrial myocytes through regulating L-type calcium current

AIM: To investigate whether the association of connexin 43(Cx43) and L-type calcium channel involved in the pathogenesis of atrial fibrillation (AF). METHODS: The biochemical assays and whole-cell patch-clamp technique were used to study the expression of Cx43 in human atrial tissue. The co-localization of Cx43 and L -type calcium channel, and the regulation of L-type calcium current in atrial myocytes were investigated. RESULTS: The expression of Cx43 at mRNA and protein levels was decreased in human atrial tissues of AF patients. In cultured atrium-derived myocytes (HL-1 cells), knockdown of Cx43 significantly inhibited the mRNA expression of L-type calcium channel α1c subunit, as well as L-type calcium current. Co-localization of Cx43 with L-type calcium channel α1c subunit in mouse atrial myocytes was observed. CONCLUSION: The decrease in Cx43 is involved in the pathogenesis of AF, probably through reducing the L-type calcium current in atrial myoctyes by co-localization with L-type calcium channel, thus representing the potential pathogenesis in atrial fibrillation.     

Expression of Jagged2/Notch3 signaling molecules in pulmonary hypertensive rats induced by monocrotaline

AIM: To study the expression of Jagged2/Notch3 signaling molecules in pulmonary vascular wall of pulmonary hypertensive rats induced by monocrotaline. METHODS: SD rats were randomly divided into normal control group (C group,n=15), solvent control group (S group,n=15) and monocrotaline model groups (M group,n=15). The model of pulmonary hypertension was established by a single subcutaneous injection of monocrotaline (50 mg/kg). The rats in S group were given a single subcutaneous injection of the same dose of solvent. After 4 weeks, the pulmonary vascular remodeling was assessed by HE staining, and the mean pulmonary artery pressure (mPAP) and right ventricular systolic pressure (RVSP) were determined by right heart catheterization. The expression of Jagged2/Notch3/Hes5 molecules in the pulmonary vascular wall was detected by immunohistochemical method and real-time PCR. RESULTS: Compared with S group and C group, the percentage of medial wall thickness of smaller arteries in model group increased significantly (P<0.01). The levels of mPAP and RVSP in M group were significantly higher than those in S group and C groups (P<0.01). The results of real-time PCR showed that the expression of Jagged2, Notch3 and Hes5 was significantly increased in M group compared with S group and C group. The data from immunohistochemical detection indicated that Jagged2 mainly expressed in the intima of small lung artery, Notch3 and Hes5 mainly expressed in the medial smooth muscle cells. Compared with S group and C group, the expression of Jagged2 and Notch3 was significantly increased in the lung small arteries of M group. CONCLUSION: The activation of Jagged2/Notch3 signaling pathway might play an important role in the formation of pulmonary hypertension.     

华灰莉木提取物对柑橘全爪螨的杀螨活性

以华灰莉木为试材,采用溶剂冷浸法提取植物成分,溶剂液-液萃取法对植物成分进行初步分离,Potter喷雾法测定杀螨活性,研究华灰莉木提取物和初步分离物对柑橘全爪螨的影响。结果表明:华灰莉木叶和枝条甲醇提取物对柑橘全爪螨雌成螨有较高的触杀毒力,致死中浓度(LC50)分别为5.5334g/L和3.9682g/L。叶的杀螨活性物质主要集中在水萃取物中,属于极性强的物质;枝条的杀螨活性物质主要集中在石油醚萃取物中,属于极性弱的物质。叶提取物的水萃取物和枝条提取物的石油醚萃取物对柑橘全爪螨雌成螨的LC50分别为3.2970g/L和1.7372g/L。     

水稻籼爪交和粳爪交杂种优势的研究Ⅰ.杂种农艺性状的表现

研究了籼爪交和粳爪交杂种在长沙、三亚两地农艺性状和干物质产量的表现. 由引进爪哇稻品种所配的籼/爪和粳/爪交杂种F1代表现为植株高大, 生长茂盛, 具有强大的干物质产量优势. 绝对干物质产量(单株穗期干重)为: 籼爪杂种＞粳爪杂种＞对照(籼籼交杂种), 干物质生产率(单株日干物质产量)为: 籼爪杂种＞粳爪杂种＞对照. 供试籼     

超级杂交稻模式株型的光合优势

以三系杂交稻汕优63为对照，比较研究表明，具有超级杂交稻模式株型的培矮64S/E32有以下生理优势。（1）叶面积指数前期较小，中期稳健，后期衰减缓慢、维持较高水平；（2）抽穗后剑叶和倒2叶的比叶重显著或极显著大于对照；（3）后期上3叶叶绿素含量衰减缓慢，变化平稳；（4）上3叶单位叶面积气孔数目比对照多19.2%，达极显
