Capillary gas chromatography (GC) with flame ionization detection was used to determine the cellular fatty acid profiles of various food-borne microbial pathogens and to compare the fatty acid profiles of spores and vegetative cells of the same endospore-forming bacilli. Fifteen bacteria, representing eight genera (Staphylococcus, Listeria, Bacillus, Yersinia, Salmonella, Shigella, Escherichia, and Vibrio) and 11 species were used to compare the extracted fatty acid methyl esters (FAMEs). Endospore-forming bacilli were processed to obtain pure spores and whole cell FAMEs for GC analysis. A data set for each bacterial agent was prepared using fatty acid profiles from five replicates prepared on different days. The results showed that these fatty acid intensity profiles were unique for each of the 11 species and that they could be used as a fingerprint for the organisms. The cellular fatty acid profiles for Bacillus anthracis and Bacillus cereus show that there are two branched chain fatty acids, iso 17:1 omega10c and 17:1 anteiso, which are unique in these species. Iso 17:1 omega10c is present in B. cereus vegetative cells and spores but is not observed in B. anthracis. The 17:1 anteiso fatty acid is present in B. anthracis cells but not in B. cereus cells. Fatty acids 16:0 2OH and 17:0 iso 3OH are present in B. anthracis and B. cereus spores but not in the vegetative cells. In summary, analysis of FAMEs from bacteria and spores can provide a sensitive procedure for the identification of food-borne pathogens.  相似文献

We estimated the total inorganic fluxes of nitrogen (N), sulfur (S), chloride (Cl?, sodium (Na+, calcium (Ca2+, magnesium (Mg2+, potassium (K+ and hydronium (H+. The resistance deposition algorithm that is programmed as part of the CALMET/CALPUFF modeling system was used to generate spatially-distributed deposition velocities, which were then combined with measurements of urban and rural concentrations of gas and particle species to obtain dry deposition rates. Wet deposition rates for each species were determined from rainfall concentrations and amounts available from the National Acid Deposition Program (NADP) monitoring network databases. The estimated total inorganic nitrogen deposition to the Tampa Bay watershed (excluding Tampa Bay) was 17 kg-N ha?1 yr?1 or 9,700 metric tons yr?1, and the ratio of dry to wet deposition rates was ~2.3 for inorganic nitrogen. The largest contributors to the total N flux were ammonia (NH3 and nitrogen oxides (NO x at 4.6 kg-N ha?1 yr?1 and 5.1 kg-N ha?1 yr?1, respectively. Averaged wet deposition rates were 2.3 and 2.7 kg-N ha?1 yr?1 for NH4 + and NO3 ?, respectively.  相似文献

A national dataset of water chemistry collected for critical loads mapping is used to make a regional assessment of surface water nitrate concentrations in Great Britain. The primary data are dominated by high concentrations in lowland regions Where N inputs are dominated by non-atmospheric sources. Land cover data are used to screen out sites with potential catchment sources of N, allowing the evaluation of nitrate leaching due to atmospheric deposition alone. In the screened dataset several upland regions show elevated nitrate concentrations, notably Wales, the Pennines, Cumbria, Galloway and the Cairngorms, and there is a clear relationship between surface water nitrate and total N deposition.  相似文献

Changes in total yellow pigment (TYP) content and carotenoid composition were examined at different stages of pasta processing. Semolina samples were milled from durum genotypes with and without the Lpx‐B1.1 gene deletion and then processed into dry pasta. Significant pigment loss (12.8–15.3%) based on TYP content was observed from semolina to dough in genotypes without the gene deletion. Such loss remained low (2.0–2.8%) for genotypes with the Lpx‐B1.1 gene deletion. Extrusion and drying processes did not result in substantial pigment loss. The overall pigment loss (from semolina to dried pasta) of genotypes with the gene deletion was 9.1–12.8%, in comparison with 19.0–21.7% in genotypes without the deletion. Changes in carotenoids examined by ultra‐performance liquid chromatography showed that lutein decreased gradually from representing 80% of total carotenoids to 70% of total carotenoids during pasta processing. The reduction of lutein was mostly during dough mixing, with a decrease of 16.7% in genotypes with the Lpx‐B1.1 deletion and 27.8% in genotypes without the deletion. Minor carotenoids increased during pasta drying, possibly at the expense of lutein. Results of this study showed that although breeding for elevated yellow pigments is the key, pasta color can be further improved by reducing pigment losses at different stages of pasta processing through selection of genotypes with Lpx‐B1.1 deletion and applying a vacuum during mixing and extrusion processes.  相似文献

An unexpected three-stage melting transition has been observed in two-dimensional (2D) free-standing liquid-crystal films by in situ electron-diffraction and optical-reflectivity measurements. These data suggest the existence of two phases between the 2D solid and liquid: a hexatic phase and, at a higher temperature, an intermediate liquid phase with hexatic-like positional correlations ( approximately 40 angstroms) but no long-range orientational order. Previous high-resolution heat-capacity measurements have revealed a divergent-like anomaly at the hexatic-liquid transition that sharply contradicts the predictions of 2D melting theories. The observation of an intermediate isotropic phase may alter our understanding of 2D melting and lead to reconciliation between current experiments and theories.  相似文献

A review of agronomic and genetic approaches as strategies for the mitigation of acrylamide risk in wheat and potato is presented. Acrylamide is formed through the Maillard reaction during high-temperature cooking, such as frying, roasting, or baking, and the main precursors are free asparagine and reducing sugars. In wheat flour, acrylamide formation is determined by asparagine levels and asparagine accumulation increases dramatically in response to sulfur deprivation and, to a much lesser extent, with nitrogen feeding. In potatoes, in which sugar concentrations are much lower, the relationships between acrylamide and its precursors are more complex. Much attention has been focused on reducing the levels of sugars in potatoes as a means of reducing acrylamide risk. However, the level of asparagine as a proportion of the total free amino acid pool has been shown to be a key parameter, indicating that when sugar levels are limiting, competition between asparagine and the other amino acids for participation in the Maillard reaction determines acrylamide formation. Genetic approaches to reducing acrylamide risk include the identification of cultivars and other germplasm in which free asparagine and/or sugar levels are low and the manipulation of genes involved in sugar and amino acid metabolism and signaling. These approaches are made more difficult by genotype/environment interactions that can result in a genotype being "good" in one environment but "poor" in another. Another important consideration is the effect that any change could have on flavor in the cooked product. Nevertheless, as both wheat and potato are regarded as of relatively high acrylamide risk compared with, for example, maize and rice, it is essential that changes are achieved that mitigate the problem.  相似文献

Canine granulocytic ehrlichial organisms were transmitted from an infected dog from Missouri to two male, 10-month-old dogs by an intravenous injection of whole blood. Physical or behavioral abnormalities were not detected during the 98 days of evaluation other than a mild pyrexia from Day 18 to 20. Ehrlichial morulae were found in blood granulocytes of Dog 1 from Day 13 to 44 and of Dog 2 from Day 14 to 34 with the peak rickettsemia occurring on Day 16 for both dogs. By Day 21 after inoculaiuon, both dogs had positive titers to Ehrlichia canis. The highest titers for both dogs were found 63 days after inoculation, after which the titers decreased. Most of the hematologic abnormalities (i.e., neutropenia, lymphocytosis, thrombocytopenia) and fever occurred between 18 and 24 days after inoculation. The pathologic bases of these abnormalities were not investigated but their concurrent presence suggested an association with the dogs' immunologic responses to the granulocytic ehrlichial agent. Results from the study indicated that the canine granulocytic ehrlichial agent of Missouri may produce subclinical infections and suggested that dogs may be able to clear the organism without antimicrobial therapy.  相似文献
