ABSTRACT We examined the effects of fresh and composted animal manures on the development of root rot, dieback, and plant death caused by Phytophthora cinnamomi. Fresh chicken manure, or chicken manure composted for 5 weeks before incorporation into the potting mix (25%, vol/vol), significantly reduced pathogen survival and the development of symptoms on Lupinus albus seedlings. Chicken manure composted for 2 weeks was less suppressive. Cow, sheep, and horse manure, whether fresh or composted, did not consistently suppress populations of P. cinnamomi or disease symptoms at the rates used (25%, vol/vol). All composts increased organic matter content, total biological activity, and populations of actinomycetes, fluorescent pseudomonads, and fungi. Only chicken manure stimulated endospore-forming bacteria, a factor that was strongly associated with seedling survival. Fallowing the potting mix for an additional 8 weeks after the first harvest increased the survival of lupin seedlings in a second bioassay, with survival rates in chicken manure compost-amended potting mix exceeding 90%. These data suggest that the ability of composted manure to stimulate sustained biological activity, in particular the activity of endospore-forming bacteria, is the key factor in reducing disease symptoms caused by P. cinnamomi. Supporting these results, the survival of rooted cuttings of Thryptomene calycina was significantly higher in sand-peat potting mix following amendment with commercially available chicken manure (15% vol/vol). However, this protection was reduced if the potting mix was steam pasteurized before amendment, indicating that suppression was due to endogenous as well as introduced microbes. Chicken manure compost incorporated at 5% (vol/vol) or more was strongly phytotoxic to young Banksia spinulosa plants and is not suitable as an amendment for phosphorus-sensitive plants.  相似文献

The intracellular survival of virulent Mycobacterium bovis and avirulent M. bovis BCG in ferret alveolar macrophages was investigated. In addition, the effects of endogenous and exogenous modulators of macrophage oxidative function on bacterial survival and growth in vitro were determined. Ferret macrophages limited the initial growth of BCG, while virulent M. bovis replicated within macrophages. Intracellular bacterial survival was unaffected by the addition of specific inhibitors of macrophage oxidative function. A T-cell supernatant (TCS), derived from mitogen-stimulated lymphocyte cultures, activated ferret macrophages for heightened oxidative burst performance. However, macrophages activated by TCS, bacterial LPS or a combination of both, failed to control infection, and actually enhanced the intracellular survival of M. bovis. These results are discussed in relation to the role of macrophages in mediating tuberculosis-related pathogenesis, with respect to the fact that ferrets are important wildlife vectors of bovine tuberculosis in New Zealand.  相似文献

The structure of the sperm of Liza dumerili was studied by light microscopy and scanning and transmission electron microscopy.

The sperm have a kidney-shaped nucleus. The “hilus” faces posteriorly and the proximal centriole is situated within the nuclear pit. Nuclear rotation takes place during spermiogenesis and the sperm are therefore related to those of other teleosts showing Type I spermiogenesis. No acrosome is evident. The midpiece is small but distinct and contains four spherical mitochondria. The flagellum enters the midpiece eccentrically and the axoneme runs obliquely towards the distal centriole to which it is connected. The flagellum conforms to the 9 + 2 flagellar pattern.  相似文献

The 12-36 h post blood meal (PBM) oocyte in C. theileri is characterized by a dramatic increase in the quantity of protein with concomitant changes in the adjacent follicular epithelium. The cytoplasm shows a preponderance of mitochondria and rough endoplasmic reticulum in proportion to free ribosomes. Numerous Golgi complexes, which bud off secretory granules, are also found scattered in the cytoplasm. These changes are concerned with the deposition of the vitelline membrane and by 36 h PBM, a continuous vitelline membrane is formed at the follicle cell-oocyte interface. Vitellogenesis appears to be completed by 48 h PBM and the final maturation stages are concerned solely with deposition of the chorionic membrane. In C. theileri the chorionic membrane seems to be synthesized in a similar manner to the vitelline membrane; namely from the aggregation of secretory droplets and only proceeds once the vitelline membrane formation is completed. The chorionic membrane is made up of an endochorion and an exochorion. Although the structural complexity of the follicular epithelium is maintained at 48 h PBM, its appearance at 60 h PBM is indicative of degenerative changes. By 60 h PBM synthesis of both the vitelline and chorionic membranes has been completed and vitellogenesis has ceased. This stage then represents the complete maturation of the ovum in C. theileri.  相似文献

OBJECTIVE: To compare the accuracy of reduction and biomechanical characteristics of acetabular osteotomies repaired with luted and nonluted reconstruction plates. STUDY DESIGN: In vitro study. ANIMALS: Pelves removed from 12 adult greyhounds. METHODS: Acetabular osteotomies were created and repaired with a 6-hole, 2.7-mm reconstruction plates in 24 cadaver hemipelves. Ten hemipelves each were assigned to group I and group II. An impression cast of each acetabulum in group I was made before luting (preluting cast). Group I plates were then elevated, luted, and replaced. A second cast of each acetabulum in group I was then made (postluting cast). Step, gap, and total areas of articular osteotomy incongruence were determined from the casts. Group I (luted plate repairs) and group II (nonluted plate repairs) hemipelves were loaded ventral-to-dorsal using a materials-testing machine. Stiffness, yield load, and maximal load sustained were determined. RESULTS: Mean gap and total area of articular osteotomy incongruence for group I preluted plate repairs (7.1 mm(2) and 8.6 mm(2), respectively) were significantly greater than for group I postluted plate repairs (4.1 mm(2) and 5.1 mm(2), respectively). Mean stiffness and maximal load for group I (681 N/mm and 2,555 N, respectively) were significantly greater than for group II (360 N/mm and 1,730 N, respectively). Mean step area and mean load at yield values were not significantly different between groups. CONCLUSIONS: Luted plate repairs of osteotomized acetabulae result in improved reduction and are stiffer and stronger than nonluted plate repairs. CLINICAL SIGNIFICANCE: Plate luting may improve the accuracy of reduction of acetabular fractures where anatomic reduction is required. Plate luting may also increase the stiffness and strength of fracture repairs and arthrodeses.  相似文献

O bjectives : To ascertain whether cavalier King Charles spaniels (CKCSs) have a proportionately smaller caudal fossa compared with other small dogs and with Labradors. To evaluate if cerebellar herniation in CKCS correlates with caudal fossa volume.
M ethods : In this retrospective study, three-dimensional images were created from magnetic resonance imaging brain series of 117 dogs (split into three groups: CKCS, Labradors and small breeds) from which the volumes of the fossae and brain parenchyma were calculated. These volumes were transformed into percentages of total cranial cavity and parenchyma volumes, respectively. The percentages were statistically compared among the groups. The percentage of herniated cerebellum in the CKCS was compared using linear regression with the caudal fossa and parenchyma percentages.
R esults : Cavalier King Charles spaniels had a proportionately smaller caudal fossa compared with Labradors (P=0·002) but not to small breeds (P=0·103). Their caudal fossa parenchyma was proportionately the same volume as Labradors (P=0·976) but greater than small breeds (P=0·005). No relationship was found for the per cent of cerebellum herniated.
C linical S ignificance : The results support mesoderm insufficiency or craniosynostosis as the pathogenesis of Chiari-like malformation (CM) in CKCS. It presents evidence for overcrowding of the caudal fossa due to a mismatch of brain parenchyma and fossa volumes as to why CKCS and not other small dogs are affected.  相似文献
