An understanding of the basic defect in the inherited disorder cystic fibrosis requires cloning of the cystic fibrosis gene and definition of its protein product. In the absence of direct functional information, chromosomal map position is a guide for locating the gene. Chromosome walking and jumping and complementary DNA hybridization were used to isolate DNA sequences, encompassing more than 500,000 base pairs, from the cystic fibrosis region on the long arm of human chromosome 7. Several transcribed sequences and conserved segments were identified in this cloned region. One of these corresponds to the cystic fibrosis gene and spans approximately 250,000 base pairs of genomic DNA. 相似文献
In many genetic disorders, the responsible gene and its protein product are unknown. The technique known as "reverse genetics," in which chromosomal map positions and genetically linked DNA markers are used to identify and clone such genes, is complicated by the fact that the molecular distances from the closest DNA markers to the gene itself are often too large to traverse by standard cloning techniques. To address this situation, a general human chromosome jumping library was constructed that allows the cloning of DNA sequences approximately 100 kilobases away from any starting point in genomic DNA. As an illustration of its usefulness, this library was searched for a jumping clone, starting at the met oncogene, which is a marker tightly linked to the cystic fibrosis gene that is located on human chromosome 7. Mapping of the new genomic fragment by pulsed field gel electrophoresis confirmed that it resides on chromosome 7 within 240 kilobases downstream of the met gene. The use of chromosome jumping should now be applicable to any genetic locus for which a closely linked DNA marker is available. 相似文献
In the western tropical Pacific, the interannual migration of the Indonesian Low convective system causes changes in rainfall that dominate the regional signature of the El Ni?o-Southern Oscillation (ENSO) system. A 96-year oxygen isotope record from a Tarawa Atoll coral (1 degrees N, 172 degrees E) reflects regional convective activity through rainfall-induced salinity changes. This monthly resolution record spans twice the length of the local climatological record and provides a history of ENSO variability comparable in quality with those derived from instrumental climate data. Comparison of this coral record with a historical chronology of EI Ni?o events indicates that climate anomalies in coastal South America are occasionally decoupled from Pacific-wide ENSO extremes. Spectral analysis suggests that the distribution of variance in this record has shifted among annual to interannual periods during the present century, concurrent with observed changes in the strength of the Southern Oscillation. 相似文献
Nitrogen-fixing species can dramatically increase soil acidity and organic matter content, and potentially alter biogeochemical P dynamics. We compared ecosystem P cycling in adjacent stands of N2-fixing red alder (Alnus rubra Bong.) and non-fixing Douglas-fir (Pseudotsuga menziesii Franco) in order to determine whether P-cycling rates within stands were related to soil P forms as measured by sequential P fractionation. Above-ground annual P uptake was 61% greater in the red alder stand, although soil available P, as measured by Bray (NH4F–HCl) extraction, was only 10% of that found in the Douglas-fir stand. Total ecosystem P in the alder stand was only 69% of that found in the Douglas-fir stand, and could indicate a pre-establishment difference between stands. However, the percentage of total soil P released by Bray or NaOH extraction was also lower in the alder stand, which suggests that differences in total P alone did not control the patterns observed in P fractions. Concentrations of inorganic P sorbed to Fe and Al minerals and contained in Fe minerals and apatite were greater under Douglas-fir, while organic P was slightly greater under red alder. While fluxes of P in litterfall, uptake and resorption were 94, 60 and 292% higher in the alder stand, soil extractable fractions meant to represent available P were lower under alder. Static measures of available P do not appear to adequately reflect P supply, and the development of techniques to assess P turnover is needed to better understand cycling and plant availability of P. 相似文献
The response of a forest to nutrient and moisture stresses is reflected in nutritional, physiological, and structural changes that include efficiency of nutrient use, translocation and cycling of nutrients, transpiration, retention of foliage, below-ground and above-ground allocation of carbon, as well as the structural development of the forest stand and its growth characteristics. This article reviews the relationship of forest ecosystems to nutrient and moisture stresses and addresses the means by which productivity can be enhanced by altering nutrient and moisture regimes.
Considerable research has focused on optimizing productivity by minimizing nutrient and moisture stresses. Research involved in nutrient additions has led to the use of commercial fertilizers to improve forest productivity. The results suggest that many forests are deficient in N and P and, to a lesser extent, S, K, Mg and trace elements. The duration of response for most nutrient additions is, however, relatively brief and the efficiency of the tree in using fertilizer is relatively poor. Long-term correction of nutrient deficiencies is seldom achieved with chemical fertilizers. However, N added through symbiotic fixation or, on a more limited scale, through addition of municipal and industrial waste by-products, can provide an excellent long-term growth response.
It is seldom feasible to change the moisture regime of a forest ecosystem through irrigation. However, field trials involving irrigation have demonstrated that moisture stress can limit productivity. There are various ways of minimizing moisture stress without irrigation, including mulching, removing ground-cover vegetation, and changing the spatial characteristics of the forest cover.
Research trials show that forest ecosystems will respond to moisture and nutrient additions; however, these responses and interactions between nutrients and moisture are typically poorly understood, and many questions remain unanswered: Does fertilization increase moisture-use efficiency of a forest or simply improve the nutrition of the site? Does improving the moisture regime of a site improve productivity primarily by decreasing moisture stress or by increasing nutrient availability and the rate of nutrient uptake? Is there a synergism in growth response with the addition of both nutrients and moisture? The linkages between nutrients and moisture appear inseparable and confound experimentation in this field. Answers to these questions and issues need to be found for the future development of plantation forestry. 相似文献
