【目的】随着养殖业集约化和规模化的发展,舍内环境对畜禽生长的影响日益突出。在密闭舍饲条件下,不断产生有毒有害气体,其中氨气的危害最大,降低畜禽生产性能,威胁动物健康。为此开展慢性氨气应激对肉鸡血清代谢物影响的研究,从小分子物质和代谢途径方面探究氨气导致机体代谢发生的变化,为舍内氨气的合理调控提供数据支持。【方法】试验采用单因素完全随机设计,将96只21日龄健康AA肉鸡随机分成对照组和试验组,每组设4个重复,每个重复12只鸡。对照组氨气浓度为0 mg·kg-1,试验组氨气浓度为45 mg·kg-1。试验在人工模拟呼吸舱内进行,呼吸舱采用全自动化控制温湿度等养殖条件,采用网上平养,自由采食和饮水,24h光照,试验期为21d。在肉鸡42日龄时,从每个重复中随机抽取2只,翅静脉采血,分离血清。将血清样品预处理后,采用气相色谱/质谱联用技术(gas chromatograph tandem mass spectrometer technology, GC-MS)检测代谢产物,利用质谱数据库对其进行鉴定。通过主成分分析、偏最小二乘法判别分析、正交偏最小二... 相似文献

An epizootic of equine infectious anemia (EIA) involved 35 horses on a farm in south Georgia. During a 126-day period, 21 of these horses became seropositive for EIA. After the initial diagnosis in July, the horses were tested every 7 to 10 days. At least one additional horse was found to be seropositive on each testing day. As soon as they were determined to be seropositive, the horses were removed from the herd and sent to slaughter. The removal of the seropositive horses, however, did not stop the epizootic. We believe the initial infection was from a 7-year-old stallion that recently had been purchased or from 1 of 2 mares that were seropositive for EIA on the first test. Two trials were conducted to determine the influence of prefast and postfast dietary protein concentration on feed intake of steers (avg wt 250 kg) and the N and P metabolism of lambs (avg wt 50 kg). In Trial 1, crossbred steers were fed prefast diets containing 8, 12 or 16% crude protein (CP) for 14 d before being deprived of feed and water for 24 h. Next, they received feed for 24 h and again were deprived for 48 h. Finally, they were fed diets containing either 10 or 15% CP. Steers fed the 16% CP prefast diet that were switched to the 10% CP realimentation diet tended to have lower feed intakes than the other fasted groups. In Trial 2, 16 crossbred lambs housed in metabolism stalls were fed prefast and postfast diets containing either 11 or 16% CP both before and after a 72-h fast in a 2 x 2 factorial arrangement. Nitrogen and P balance during realimentation tended to be affected by both the prefast and postfast dietary protein concentration. Results of this study indicate that CP content of the postfast diet alters repletion of nutrients lost during a feed and water deprivation period and can affect realimentation feed intake. The subacute toxic effects of cyclopiazonic acid (CPA; given orally) were characterized in the dog (CPA was purified from cultures of Aspergillus flavus). Four groups of dogs were given CPA in gelatin capsules for 90 days at the following dosage levels: 0.05, 0.25, 0.5, and 1.0 mg/kg of body weight; a 5th group was used as controls. All dogs administered the 0.5 and 1.0 mg of CPA/kg dosages and 1 dog given the 0.25 mg of CPA/kg dosage died or were humanely killed before the scheduled termination of the study. Clinical signs of intoxication appeared 2 to 44 days after dosing was started and consisted of anorexia and, in 1 to 2 days, vomiting, diarrhea, pyrexia, dehydration, weight loss, and CNS depression. Grossly, the entire alimentary tract had diffuse hyperemia with focal areas of hemorrhage and ulceration. Other lesions were renal infarcts, necrotizing epididymitis, and ulcerative dermatitis. Microscopic lesions included ulceration, necrosis, vasculitis, lymphoid necrosis, karyomegaly in several organs, and decreased mitotic activity in intestinal crypt epithelium. Ulcerative and necrotic lesions were usually associated with vascular lesions. Clinical pathologic changes were leukocytosis, neutrophilia, lymphopenia, monocytosis, and increased serum alkaline phosphatase activity. 相似文献

Little is known about the analgesic action of buprenorphine (BUP) in cats. Relative to man, the cat has a more alkaline oral pH, which may make this an effective route for administering BUP in this species. This study aimed to assess and compare the pharmacokinetics and pharmacodynamics of sublingual (S‐L) and IV administration of BUP. Thermal threshold (TT) was measured and blood samples were collected following IV or S‐L administration (20 µg kg?1) of the injectable formulation. Six cats (five spayed females, one castrated male, 4.1–6.6 kg) were used. Each cat received both treatments in a randomized cross‐over study design with 1 month between experiments. Twenty‐four hours prior to each study, the lateral thorax of each of the cats was shaved, cephalic and jugular catheters placed, and oral pH measured. On the day of the study, TT was measured using a ‘thorax‐mounted’ thermal threshold‐testing device specifically developed for cats. The cats were free to move around. Skin temperature was recorded before each test, then the heater activated. When the cat responded by flinching, turning, or jumping, the stimulus was terminated and the threshold temperature was recorded. The thermal threshold cut‐off point was 55.5 °C. Three baseline thresholds were recorded before treatment with S‐L or IV (via cephalic catheter) BUP (20 µg kg?1). Blood was withdrawn (jugular) at 1, 2, 4, 6, 10, 15, 30, 45, 60 minutes and at 2, 4, 6, 8, 12, and 24 hours post‐administration. TT was measured every 30 minutes?6 hours, 1–12 hours, and at 24 hours post‐administration. Plasma was immediately separated, stored at ?20.5 °C, and assayed within 4 months using a commercially available 125I radioimmunoassay. Threshold data were analyzed using anova with a repeat factor of time. No adverse effects were noted. Pupils were dilated for up to 9 hours post‐BUP. Behavioral changes were calm euphoria. Measured oral pH was 9 in each cat. Pre‐treatment mean threshold (±SD) was 41.2 ± 0.9 °C in the S‐L group and 40.8 ± 0.85 °C in the IV group. There were no significant differences between the groups with respect to thresholds over time (p = 0.72). Thresholds were significantly increased from 30 to 360 minutes in both the groups (>44.615 °C). Peak plasma BUP (Cmax) was lower (11 ± 6.7 ng mL?1vs. 92.9 ± 107.9 ng mL?1) and occurred later (Tmax) (30 minutes vs. 1 minute) after S‐L compared to IV administration, respectively. BUP (20 µg kg?1)‐administered S‐L or IV provided antinociception between 30 and 360 minutes after administration. Plasma levels did not correspond to TT. 相似文献
