The effect of available C and N on the rate of wheat (Triticum aestivum L.) straw decomposition by microorganisms was determined using electrolytic respirometers under laboratory conditions. Three lots of spring wheat straw containing 1.13, 0.79 and 0.18% N were used. Cold-water soluble C and N were leached from the wheat straw and the respiration from the leached and non-leached straws (1.5 g of each), with and without added N, were compared in a sand system for 881 h. The fraction of soluble C declined as straw N content declined (total soluble C was 14.0, 11.4 and 8.9%, respectively, for the three straws). The fraction of soluble N declined as straw N content declined, but the proportion of total N that was soluble varied with initial straw N content. We postulated from decomposition data for the various treatments that the amount of microbial biomass produced and the overall rate of wheat straw decomposition in the early stages is largely dependent on the size of the soluble C pool and an intermediately-available C pool that is not cold-water soluble but decomposes within the first few days of decay. Our results from this study imply that the amount of N immobilized during wheat straw decomposition is dependent on the amount of available C from both the primary and secondary pools present in the straw. The data also suggest that the soluble C pool and the intermediately-available C pool were metabolized simultaneously.  相似文献

Field studies were conducted for three seasons (1978–1979, 1979–1980 and 1981–1982) on a Palouse silt loam near Pullman, Washington, to compare the effects of broadcast and deep banding of nitrogen (N) fertilizer beneath winter wheat (Triticum aestivum L.) seed on N uptake and dry matter production of downy brome (Bromus tectorum L.) and jointed goatgrass (Aegilops cylindrica Host.), and on N uptake, dry matter production and grain yields of winter wheat. Three tillage systems were used: conventional tillage; shallow roto-tilling, or no-tillage prior to planting. Rates of N were 0, 65, 130 and 190 kg N ha−1 as ammonium nitrate. Additional plots were maintained free of weeds at the 130 kg N ha−1 rate. In 1983–1984, deep banding of the fertilizer between rows in a paired-row configuration was compared to surface-broadcast N fertilizer using N rates of 0, 45, 90 and 135 kg N ha−1. There were no significant differences between broadcast and deep-band application of N on grass weed N uptake or dry matter production with mold-board plowed or no-tillage, but there was greater weed growth with surface-broadcast N with shallow roto-tilling. Wheat N uptake, growth and grain yields were consistently higher with band-applied N compared to broadcast N. The yield response to banding N was the same with or without the presence of grass weeds.  相似文献

Four multicannulated Holstein steers (initial BW 424 +/- 16 kg) were used in a 4 x 4 Latin square to determine the influence of protein supplementation on forage intake, site and extent of digestion, and nutrient flow in steers consuming dormant bluestem-range forage (2.3% CP). Treatments were 1) control, no supplement; 2) 1.8 kg of low-protein supplement, 12.8% CP (Low-CP); 3) 1.8 kg of moderate-protein supplement, 27.1% CP (Mod-CP); and 4) 2.7 kg of dehydrated alfalfa pellets, 17.5% CP (Dehy). The Dehy supplement was fed to provide the same amount of CP/d as Mod-CP, and all supplements provided similar amounts of ME/d. Forage DMI was increased (P less than .05) by feeding Mod-CP and Dehy. Ruminal OM digestibility was 39% greater (P less than .05) for the Mod-CP and Dehy supplementations than for the Low-CP supplementation and control. Ruminal CP digestibility was negative for all treatments, and control (-326%) was less (P less than .05) than supplemented treatments (average -27%). Total tract OM digestibility was greatest (P less than .10) for steers fed Mod-CP and least for control steers; Low-CP and Dehy steers were intermediate. Total tract NDF digestibility tended (P = .15) to be less with Low-CP than with Mod-CP and Dehy. Duodenal N flow was greater (P less than .05) with Mod-CP and Dehy than with Low-CP and control. In summary, supplementation with Mod-CP increased forage intake, digestion, and duodenal N flow compared with Low-CP or control; however, the response was similar when Mod-CP and Dehy supplements were fed to provide equivalent amounts of CP and ME daily.  相似文献

Clinical isolates (n = 63) of Pseudomonas aeruginosa obtained from various sites in 63 horses were compared using ERIC2 RAPD PCR to determine their genetic relatedness. Resulting banding patterns (n = 24 genotypes) showed a high degree of genetic heterogeneity amongst all isolates examined, indicating a relative non-clonal relationship between isolates from these patients, employing this genotyping technique. This study characterised 63 clinical isolates into 24 distinct genotypes, with the largest cluster (genotype E) accounting for 10/63 (15.9%) of the isolates. ERIC2 RAPD PCR proved to be a highly discriminatory molecular typing tool of P. aeruginosa in isolates recovered from horses. With the adoption of several controls to aid reproducibility, this technique may be useful as an alternative to PFGE, particularly in epidemiological investigations of outbreaks where speed may be a significant parameter. This is the first report of clonal heterogeneity amongst P. aeruginosa from horses and demonstrated that ERIC RAPD PCR is a rapid method for the examination of this species in horses, which may be useful in outbreak analysis.  相似文献

Worldwide soil carbon (C) losses associated with agricultural expansion and intensification have contributed significantly to increased atmospheric CO2. Soil disturbances resulting from land use changes were shown to modify the turnover of C and the formation of soil organic matter. A native semiarid shrub-steppe ecosystem recently converted into an irrigated agricultural development in the Columbia Basin of Washington State was evaluated for several abiotic indicators that might signal changes in an ecosystem during the initial stages of conversion and disturbance. Soil samples were collected in March of 2003 and 2004 from nine sites that included native shrub-steppe and agricultural fields converted in 2001 and 2002. Disturbance from conversion to irrigated crop production influenced total organic C and nitrogen (N) storage, C and N mineralization, and C turnover. Cultivated fields had greater concentrations of total organic C and N and higher cumulative C and N mineralization than native sites after 3 years of cultivation. Soil organic C was divided into three pools: an active pool (C a) consisting of labile C (simple sugars, organic acids, the microbial biomass, and metabolic compounds of incorporated plant residues) with a mean residence time of days, an intermediate or slow pool (C s) consisting of structural plant residues and physically stabilized C, and a resistant fraction (C r) consisting of lignin and chemically stabilized C. Extended laboratory incubations of soil with measurements of CO2 were used to differentiate the size and turnover of the C a and C s functional C pools. The active pools were determined to be 4.5 and 6.5% and slow pools averaged 44 and 47% of the total C in native and cultivated fields, respectively. Cultivation, crop residue incorporation, and dairy manure compost amendments contributed to the increase in total soil C.  相似文献

The distribution of dissolved and particulate 234Th in the upper 200 m of the water column was obtained for three stations in the Ross Sea off Victoria Land and Terra Nova Bay. At site 24a, close to the retreating ice margin, all the sampled depths showed deficiencies in 234Th relative to the equilibrium with 238U. These are related to uptake of 234Th onto sinking particles. Residence times of 234Th in solution and of particulate 234Th were 130-247 days and 8.1-6.6 days, respectively. A high particle flux (1.23-5.03 g m-2 d-1) was calculated at this station. At the other two sites (11c and 15c), 234Th depth profiles are irregular, probably due to the release of dissolved 234Th by decomposing particles at certain depths, or to the contribution from lateral advection. Bulk mass fluxes measured by floating traps at stations 11c and 15c are very low (66-138 mg m-2 d-1). Also fluxes of organic carbon and nitrogen, and biogenic silica are reported for these two sites. The calculated fluxes are discussed with respect to methodologies and to the dynamics of the ice margin retreat.  相似文献

Three experiments were conducted to compare soybean meal/sorghum grain (SBM/SG), alfalfa hay or dehydrated alfalfa pellets (DEHY) as supplemental protein sources for beef cattle grazing dormant range forage. In Exp. 1 (35-d digestion study), 16 ruminally cannulated steers were stratified by weight (average BW 259 kg) and assigned randomly within stratification to: 1) control, no supplement; 2) SBM/SG (25% CP) fed at .48% BW; 3) alfalfa hay (17% CP) fed at .70% BW; or 4) DEHY (17.4% CP) fed at .67% BW. Steers receiving protein supplements displayed at least a twofold increase in forage intake (P less than .10). In addition, steers supplemented with DEHY consumed approximately 15% more forage (P less than .10) than SBM/SG- or alfalfa hay-supplemented steers. Digestible DM intake (kg/d), however, was similar between alfalfa hay- and DEHY-supplemented steers and 20% greater (P less than .10) than for SBM/SG-supplemented steers. In Exp. 2, 82 mature, nonlactating Hereford x Angus cows (average BW 489 kg) were assigned randomly to SBM/SG, alfalfa hay or DEHY supplement treatments, which were replicated in three pastures. Cows supplemented with DEHY gained more weight (P less than .05) during the first 84 d of supplementation and displayed the least amount of weight loss at calving (d 127; P less than .05) and just prior to breeding (P less than .10). In contrast, calving interval (361 d) and pregnancy rate (94%) were unaffected (P greater than .10) by dam's previous supplemental treatment. In Exp. 3, one block (pasture) of cows from Exp. 2 was selected at random and grazing behavior was monitored during week-long periods in January and February. A treatment X time interaction (P less than .05) occurred for total time spent grazing; treatments did not differ in January, but cows supplemented with alfalfa hay spent less time grazing in the February grazing period. In conclusion, DEHY and alfalfa hay appear to be at least as effective as SBM/SG as a supplemental protein source for pregnant grazing cows when supplements are fed on an equal CP and ME basis.  相似文献

An antibody elicited to a distorted N-methyl porphyrin catalyzed metal ion chelation by the planar porphyrin. At fixed Zn2+ and Cu2+ concentrations, the antibody-catalyzed reaction showed saturation kinetics with respect to the substrate mesoporphyrin IX (2) and was inhibited by the hapten, N-methylmesoporphyrin IX (1). The turnover number of 80 hour-1 for antibody-catalyzed metallation of 2 with Zn2+ compares with an estimated value of 800 hour-1 for ferrochelatase. The antibody also catalyzed the insertion of Co2+ and Mn2+ into 2, but it did not catalyze the metallation of protoporphyrin IX (3) or deuteroporphyrin IX (4). The antibody has high affinity for several metalloporphyrins, suggesting an approach to developing antibody-heme catalysts for redox or electron transfer reactions.  相似文献
