Four experiments were conducted to evaluate the influence of changing the proportion of supplemental degradable intake protein (DIP) from urea on forage intake, digestion, and performance by beef cattle consuming either low-quality, tallgrass prairie forage (Exp. 1, 2, and 4) or forage sorghum hay (Exp. 3). Experiments 1, 2, and 3 were intended to have four levels of supplemental DIP from urea: 0, 20, 40, and 60%. However, refusal to consume the 60% supplement by cows grazing tallgrass prairie resulted in elimination of this treatment from Exp. 1 and 2. Levels of supplemental DIP from urea in Exp. 4 were 0, 15, 30, and 45%. Supplements contained approximately 30% CP, provided sufficient DIP to maximize digestible OM intake (DOMI) of low-quality forage diets, and were fed to cows during the prepartum period. In Exp. 1, 12 Angus x Hereford steers (average initial BW = 379) were assigned to the 0, 20, and 40% treatments. Forage OM intake, DOMI, OM, and NDF digestion were not affected by urea level. In Exp. 2, 90 pregnant, Angus x Hereford cows (average initial BW = 504 kg and body condition [BC] = 5.0) were assigned to the 0, 20, and 40% treatments. Treatment had little effect on cow BW and BC changes and calf birth weight, ADG, or weaning weight. However, pregnancy rate tended to be lowest (P = 0.13) for the greatest level of urea. In Exp. 3, 120 pregnant, crossbred beef cows (average initial BW = 498 kg and BC = 4.6) were assigned to the 0, 20, 40, and 60% treatments. Prepartum BC change tended (P = 0.08) to be quadratic (least increase for 60% treatment), although BW change was not statistically significant. Treatment effect on calf birth weight was inconsistent (cubic; P = 0.03), but calf ADG and weaning weight were not affected by treatment. Pregnancy rate was not affected by prepartum treatment. In Exp. 4, 132 pregnant, Angus x Hereford cows (average initial BW = 533 and BC = 5.3) were assigned to the 0, 15, 30, and 45% treatments. Prepartum BC loss was greatest (quadratic; P = 0.04) for the high-urea (45%) treatment, although BW loss during this period declined linearly (P < 0.01). Prepartum treatment did not affect pregnancy rate, calf birth weight, or ADG. In conclusion, when sufficient DIP was offered to prepartum cows to maximize low-quality forage DOMI, urea could replace between 20 and 40% of the DIP in a high-protein (30%) supplement without significantly altering supplement palatability or cow and calf performance.  相似文献

Recently, in vitro fertilization (IVF) in the horse has met with less than anticipated results. Various problems associated with equine IVF include: (1) the inability to collect large numbers of good quality oocytes, (2) the alteration of the zona pellucida associated with in vitro maturation of equine oocytes, and (3) the improper preparation of equine sperm cells for IVF of these oocytes. Therefore, this study was conducted to achieve fertilization via sperm injection of equine oocytes and to produce live offspring from this IVF procedure. Oocytes were collected by transvaginal ultrasound-guided oocyte retrieval procedures from early pregnant mares of mixed breeds (day 14 to day 70 of pregnancy) and were matured in vitro and subjected to intracytoplasmic sperm injection (ICSI). Injected oocytes were then cultured for 48 hours in either TCM-199 or P-1 medium (glucose and phosphate-free medium) supplemented with 15% fetal bovine serum. Cleavage rates for embryos cultured in the two culture media were different (47% vs. 63% in TCM-199 and P-1, respectively). Also, four Grade 1 embryos were surgically transferred into the oviducts of four recipient mares (one embryo/mare) at 48 hours post-ICSI, with three pregnancies (75%) developing as ultrasonically demonstrated by the presence of an embryonic vesicle in the uterine body by day 16 post-ICSI. On June 23rd one live filly was born after 328 days of gestation and subsequently, a second healthy filly was born after 319 days of gestation. To our knowledge, this is the first report of live foals resulting from in vitro fertilization (via ICSI) of in vitro matured oocytes recovered from pregnant mares using an efficient, repeatable transvaginal ultrasound-guided procedure.  相似文献

Six ruminally fistulated crossbred steers (BW = 369 kg) were used in a randomized complete block experiment to test the efficacy of continuous-infusion pumps and controlled-release boluses for administering external markers to predict fecal output. Steers, limit-fed chopped alfalfa hay at 2% of BW daily, were fitted with continuous-infusion pumps that administered Co-EDTA and YbCl3 solutions intraruminally. In addition, a controlled-release bolus containing Cr2O3 was inserted into the rumen of each steer. Fecal grab samples were taken every 6 h for 7 d during initial marker equilibration; after this period, fecal grab samples were taken every 3 h for 48 h to evaluate diurnal variation patterns. Steers were subsequently fitted with fecal bags for 7 d to allow total fecal collection. Grab samples also were taken during the total fecal collection period at 0600 (AM) and 1400 (PM). The marker X time effect was nonsignificant. Similarly, time of grab sampling (AM or PM) did not affect estimates of fecal output (P greater than .10), but the Cr2O3 bolus overestimated fecal output (P less than .05). Fecal marker concentrations during the 48-h profile showed little diurnal variation regardless of marker used. All markers equilibrated in the feces at approximately 100 to 120 h after initiating infusions. The continuous-infusion pumps evaluated were efficacious for administering markers for estimating total fecal output in limit-fed steers; however, the Cr2O3 boluses evaluated overestimated fecal output when the manufacturer's suggested release rate was used for fecal output calculations.  相似文献

REASONS FOR PERFORMING STUDY: The mediators and signalling cascades important in the initiation of laminitis remain unclear. We therefore wanted to explore the genes and overall signalling mechanisms that play an important role in the developmental stage of laminitis. OBJECTIVE: To use a broad genomic screening technique to identify novel genes that are differentially regulated in the equine lamellae during the developmental period of laminitis. METHODS: Differential mRNA display (DRD) was performed to discover regulated genes, and real-time quantitative polymerase chain reaction (RT-qPCR) was then used to evaluate lamellar mRNA levels of a regulated gene (MAIL) and mediators related to that gene (IL-1beta and IL-6) in control horses (n = 5) and horses administered black walnut extract (BWE; n = 5). RESULTS: Using DRD, MAIL was identified as a regulated gene. RT-qPCR indicated a 4-fold increase in expression of the MAIL mRNA in BWE lamellae compared to controls. A 30-fold increase in IL-1beta, and a 160-fold difference in IL-6 mRNA expression was present in BWE lamellae. Differences in MAIL, IL-1beta and IL-6 mRNA expression were statistically significant between groups (P < 0.05). CONCLUSIONS AND POTENTIAL RELEVANCE: The data strongly support a role for inflammatory cytokines in the developmental stages of laminitis, possibly inducing the vascular and metabolic alterations reported to occur in the affected digit. These results potentially support the use of anti-inflammatory drugs in horses at risk of laminitis, and warrant further investigation of the link between systemic disease processes associated with laminitis and the reported digital inflammation.  相似文献

Three independent experiments were conducted each using 16 ruminally fistulated beef steers fed bermudagrass (8.2% CP, 71% NDF; Exp. 1), bromegrass (5.9% CP, 65% NDF; Exp. 2), or forage sorghum (4.3% CP, 60% NDF; Exp. 3) hays to evaluate the effects of increasing level of supplemental degradable intake protein (DIP) on forage utilization. In each experiment, steers were blocked by weight and assigned to one of four treatments, and hay was offered to each steer at 130% of average voluntary intake for the preceding 5-d period. Supplemental DIP (sodium caseinate) was placed in the rumen at 0700, immediately before feeding forage. Levels of DIP supplementation were .041, .082, and .124% BW; the control received no supplemental DIP. Following a 10-d adaptation, intake and total fecal output were measured for 7 d. In Exp. 1, neither forage OM intake (FOMI) nor fiber (NDF) digestion were influenced (P > or = .20) by increasing level of DIP supplementation. The DIP supplied by the bermudagrass hay was estimated to be 8.2% of the total digestible OM intake (TDOMI) for control steers. In Exp. 2, increasing level of supplemental DIP did not affect (P > or = .26) FOMI but tended to increase total OM intake linearly (TOMI; P = .10). The tendency for a rise in TOMI coupled with a slight numeric increase in digestion resulted in an increase (linear; P = .06) in TDOMI. In the treatment group in which the maximum TDOMI was observed (supplemental DIP treatment of .082% BW), total DIP intake constituted approximately 9.8% of the TDOMI. In Exp. 3, FOMI, TOMI, organic matter digestion (OMD), and TDOMI were improved (P < .01) by increasing amounts of supplemental DIP. Although there was some evidence of a tendency for a decrease in the magnitude of change in TDOMI in response to increasing DIP supplementation, a clear plateau was not achieved with the levels of supplement provided. When the highest level of supplemental DIP was fed, DIP constituted approximately 12.8% of the TDOMI. In conclusion, significant variation was observed among forage in the amount of DIP needed to maximize intake and digestion when expressed in relationship to the digestible OM.  相似文献

ObjectiveTo determine if general anesthesia with sevoflurane and laparoscopic surgery changed gastric and small bowel propulsive motility or pH in dogs.Study designProspective, controlled trial.AnimalsTwelve, 19–24 months old, female, Treeing Walker Hound dogs, weighing 23–30 kg.MethodsDogs were anesthetized for a median of 8.5 hours during another study to determine the minimum alveolar concentration of sevoflurane using a visceral stimulus. Gastric and small bowel motility were determined using a sensor capsule that measures pressure, pH and temperature. Gastric transit time and motility index were calculated. For 8/12 dogs, gastric motility, pH and transit time were measured. In 4/12 dogs, small bowel motility and pH were measured.ResultsAnesthesia decreased gastric and small bowel motility but did not change luminal pH. Mean gastric contraction force decreased from median (range) 11 (8–20) to 3 (1–10) mmHg (p < 0.01) and gastric motility index decreased from 0.63 (0–1.58) to 0 (0–0.31; p = 0.01). Frequency of contractions did not change, 3.7 (1.6–4.4) versus 2.8 (0.1–5.1) contractions minute?1 (p = 0.1). Gastric motility returned to normal 12–15 hours following anesthesia. Gastric emptying was prolonged from 12 (5.3–16) to 49 (9.75–56.25) hours (p < 0.01). Mean small bowel contraction force decreased from 34 (24–37) to 3 (0.9–17) mmHg (p < 0.02) and motility index decreased from 3.75 (1–4.56) to 0 (0–1.53; p = 0.02). Frequency of contractions did not change, 0.5 (0.3–1.4) versus 1.4 (0.3–4.6) contractions minute?1 (p = 0.11). Small bowel motility returned within 2 hours after anesthesia. Laparoscopy did not result in changes to gastric or small bowel parameters beyond those produced by general anesthesia.Conclusions and clinical relevanceThe force of gastric and small bowel contractions decreased during sevoflurane anesthesia for laparoscopy. Although gastric motility returned to normal within 12–15 hours the impairment of gastric emptying lasted 30–40 hours, predisposing dogs to postoperative ileus.  相似文献

Elevation of the triiodothyronine (T3) content of striped bass (Morone saxatilis) eggs by maternal T3 injection confirms the uptake of T3 by oocytes. The resulting offspring were influenced favorably by the T3, as seen in quantitative indices of development. As reported previously, larvae from T3-supplemented eggs raised under laboratory conditions exhibited increased body area, length, dry weight, and rates of swimbladder inflation and survival, compared to controls. Also, the T3 content of unfertilized oocytes correlated positively and highly significantly with survival to two weeks of age within individual cohorts (Brownet al., 1988). In the present study, the survival of experimental and control striped bass was monitored through the fingerling stage, under hatchery production conditions. The rate of recovery of maternally T3-treated cohorts from pond-culture was approximately fourfold that of controls. The striking effects of T3 enrichment of eggs on offspring indicate the potential contribution of maternal hormones in striped bass development, and suggest possible applications in aquaculture.  相似文献

Two experiments were conducted to determine the influence of dietary roughage concentration and feed intake on finishing steer performance and ruminal metabolism. In Exp. 1, 126 steers (334 kg) were used in a completely randomized design and fed (120 d) diets of steam-rolled wheat without roughage or containing 5, 10 or 15% roughage (50% alfalfa hay:50% corn silage). Steers fed 5 or 10% roughage gained faster (quadratic, P less than .05) and were more efficient (quadratic, P less than .05) than steers fed 15% or no roughage. In Exp. 2, six ruminally cannulated steers (447 kg) were used in a 6 x 6 latin square design and fed (twice daily) diets of steam-rolled wheat without roughage or containing 5 or 15% alfalfa hay at twice or three times NE required for maintenance. Increasing dietary roughage increased (linear, P less than .01) ruminal liquid passage 38%, indigestible ADF passage 63%, Yb-labeled wheat passage 75% and fiber fill 31%. The rate of in situ starch digestion tended to increase (linear, P = .16), and ruminal VFA concentration was 40 mM higher (P less than .01) at 4 h after feeding with increased roughage. Increased feed intake increased (P less than .05) ruminal starch fill, fiber fill, liquid fill and liquid passage 23%, Yb-labeled wheat passage 50% and Dry-labeled hay passage 20%. It reduced protozoa five- to sixfold (P less than .01) but doubled total bacterial counts (P less than .01). Ruminal NH3N was lower (P less than .01) and total VFA concentration was 50 mM higher (P less than .01) at 4 h after feeding. The acetate:propionate ratio was reduced from 2.3 to 1.3 (P less than .01) with increased intake. Adding roughage to a steam-rolled wheat diet increased passage and tended to increase rate of starch digestion; increased feed intake with its associated effects on ruminal fill and passage dramatically shifted the microbial population and fermentation end products.  相似文献

Serums from 4 species of deer and 1 species of antelope were analyzed for various components in order to define an animal disease model for sickle cell disease in people. Animal species included black-tailed deer (Odocoileus hemionus columbianus), mule deer (Odocoileus hemionus), sika deer (Cervus nippon nippon), fallow deer (Dama dama), and pronghorn antelope (Antilocapra americana). The mean serum values for total bilirubin, total protein, albumin, creatinine, urea nitrogen, and electrolytes were similar in all species and were in the normal range for human beings. Cholesterol and uric acid values for all animals were lower than those for people. Alkaline phosphatase values in the 4 cervid species were higher than in the pronghorn antelope. Values for glutamic oxalacetic transaminase were lower in the cervids than in the pronghorn antelope. Lactic dehydrogenase values were similar in the 5 species. High activities for glutamic oxalacetic transaminase and lactic dehydrogenase in the 5 species probably related to muscle mass and great muscular activity.  相似文献
