Accurate identification of bovine parainfluenza type 3 virus in bovine respiratory disease requires dependable, sensitive, and specific techniques for detection in affected animals. Immunohistochemical testing can be a rapid and reliable means of demonstration of virus in tissues from suspect cases; however, this procedure is dependent upon the quality of the antisera directed against the viral antigens. The production of rabbit polyclonal and murine monoclonal antibodies directed against bovine parainfluenza type 3 virus and techniques for their use in fresh-frozen and formalin-fixed paraffin-embedded tissues in immunofluorescence and immunoperoxidase-based immunohistochemical tests are described.  相似文献

Four rumen-fistulated steers averaging 400 kg in body weight were used in a 4 X 4 Latin square arrangement with 18-d periods to investigate the effect of treating soybean meal (SBM) with formaldehyde on nitrogen (N) utilization and ruminal fermentation. Experimental diets, on a dry matter basis, consisted of 42% corn silage, 48.5% cracked corn-mineral mixture and 9.5% SBM treated with 0, .3, .6 or .9% formaldehyde by weight. Dry matter and organic matter digestibilities were not affected by treatment. Formaldehyde treatment of SBM resulted in a linear decrease in N digestibility (P less than .005) and urinary N excretion (P less than .01) and a quadratic increase (P less than .05) in N retention. The depression in apparent N digestibility was small when SBM was treated with .3% formaldehyde. This level of formaldehyde treatment also had little effect on in vitro enzymatic hydrolysis of SBM. Ruminal ammonia-N concentrations were lower (P less than .05) in steers fed formaldehyde-treated SBM. Ruminal pH was lower (P less than .05) at 6 and 8 h postfeeding while volatile fatty acid concentrations were higher (P less than .05) at 8 and 12 h postfeeding for steers fed untreated SBM. Propionic acid (mol/100 mol) decreased linearly (P less than .05) with increasing level of formaldehyde treatment. Urea-N concentrations in plasma were decreased (P less than .001) and plasma-free essential amino acid concentrations were increased (P less than .10) by formaldehyde treatment. We examined 18 horses with clinical signs of chronic obstructive pulmonary disease (COPD) using physical examination, cytological and bacteriological evaluation of bronchoalveolar fluid, and percutaneous lung biopsy. In 16 cases, histological examination of lung tissue confirmed the diagnosis of COPD. Two horses were excluded: one had uncomplicated bacterial pneumonia and in the other a satisfactory lung biopsy could not be obtained. In horses with COPD, the most common historical complaint was coughing, which was reported in 88%. The most frequently detected abnormal finding on physical examination was abnormal lung sounds; these were detected in 69% of horses at rest and in 88% of horses breathing deeply into a bag. A novel finding was that 29% of horses had lung sounds that were quieter than would be expected for the degree of respiratory effort. Horses with COPD had increased percentages of neutrophils and decreased percentages of lymphocytes and macrophages in their bronchoalveolar lavage fluid. Bronchiolar neutrophil infiltration and peribronchiolar mast cell accumulation in lung biopsy tissue had the highest correlation with clinical condition. The severity of pathological changes in biopsies of lung did not predict whether the horse would die in the two to four year follow-up period. Horses that died in the follow-up period were more severely affected clinically at initial presentation than horses that were alive at the end of the follow-up period.  相似文献

Two cases of nonsuppurative myeloencephalitis are reported which clinically and pathologically resemble equine protozoal myeloencephalitis. Lesions in both horses were associated with Toxoplasma-like organisms visible in microscopic sections. Clinical signs and lesions in one case primarily involved the brain and in the other case principally involved the spinal cord and associated meninges. Positive identification of the organisms was not achieved; however, the etiological agent is unlikely to be a species of Toxoplasma. Recently published studies suggest a species of Sarcocystis is involved.  相似文献

The frequency of claw regrowth, bony remnants, and complications in cats that underwent forelimb onychectomy using laser, scalpel, or guillotine techniques were evaluated. Eighty-seven client-owned cats were recruited from 27 veterinary clinics in the Canadian Atlantic provinces. At least 1 year after onychectomy the cats underwent a physical examination, gait analysis, and radiographic evaluation by 1 of 2 authors. There was no significant difference in the frequency of claw regrowth among the 3 methods (P = 0.283). Significantly more cats had bony remnants following guillotine onychectomy (P < 0.001). Bony remnants were significantly associated with claw regrowth (P = 0.001). Cats that underwent laser onychectomy had significantly less post-operative complications (P = 0.023). The long-term outcome was not significantly different among the 3 methods. We conclude that leaving remnants of the third phalanx in situ is associated with an increased frequency of claw regrowth. Laser onychectomy may be preferred to reduce the risk of post-operative complications, bony remnants, and claw regrowth.  相似文献

This article describes and contrasts the public health response to two human rabies cases: one organ recipient diagnosed within days of symptom onset and the transplant donor who was diagnosed 18 months post‐symptom onset. In response to an organ‐transplant‐related rabies case diagnosed in 2013, organ donor and recipient investigations were conducted by multiple public health agencies. Persons with potential exposure to infectious patient materials were assessed for rabies virus exposure. An exposure investigation was conducted to determine the source of the organ donor's infection. Over 100 persons from more than 20 agencies spent over 2700 h conducting contact investigations in healthcare, military and community settings. The 564 persons assessed include 417 healthcare workers [5.8% recommended for post‐exposure prophylaxis (PEP)], 96 community contacts (15.6% recommended for PEP), 30 autopsy personnel (50% recommended for PEP), and 21 other persons (4.8% recommended for PEP). Donor contacts represented 188 assessed with 20.2% recommended for PEP, compared with 5.6% of 306 recipient contacts recommended for PEP. Human rabies cases result in substantial use of public health and medical resources, especially when diagnosis is delayed. Although rare, clinicians should consider rabies in cases of encephalitis of unexplained aetiology, particularly for cases that may result in organ donation.  相似文献
