An ELISA was developed and tested to detect antibodies to Eperythrozoon suis in swine. Results were compared with those of the indirect hemagglutination (IHA) test. Antigen isolated from swine heavily infected with E suis was used for both tests. Comparison of the ELISA with the IHA test revealed a significant (P less than 0.001) correlation between results. Of 114 samples obtained from 9 swine infected with E suis, 87.7% were seropositive (titer greater than or equal to 200) via the ELISA, and 80.7% were seropositive (titer greater than or equal to 20) via the IHA test. The sensitivity of the ELISA was greater than that of the IHA test. All blood samples obtained from specific-pathogen-free swine tested negative for E suis antibody. Cross-reactions were not observed between E suis antigen and antisera against various swine and cattle disease agents using ELISA. We concluded that the ELISA may be used for rapid and effective diagnosis of infection with E suis in swine.  相似文献

A theoretical model of the United States demand for hardwood plywood imports was developed. Demand derives from activity in the construction industry where costs are minimised subject to a generalised Cobb-Douglas technology. This static model was then modified to reflect dynamic adjustments of imports to prices and housing starts. The resulting model was estimated with monthly data for the period January, 1974 to December, 1979. The choice of either c.i.f., f.a.s. or customs data did not significantly affect the statistical results. No significant substitution was apparent between imported hardwood plywood and domestic hardwood or softwood plywood. Import data appeared to be explained by expectations on housing starts and relative prices. Importers' expectations were represented by Cagan's adaptive expectations model and by Almon's polynomial lag equation. The latter model, relating imports to (i) housing starts and (ii) import prices, gave significantly better results. The maximum lag was of 9 months, with a mean lag of 4·5 months. The long-term elasticity of imports with respect to housing starts was estimated at 1·10 (±0·12). The long-term elasticity with respect to real price was ?1·98 (±0·24), using the price of all commodities as the deflator.  相似文献

The sensitivity and error limits of a conventional in ovo neutralization test for antibody to distemper virus were compared with the same parameters of a single dilution-probit test for neutralizing antibody.With the former test, antibody was demonstrable in ferrets 8 days following a single subcutaneous injection of virus while probit analysis of data from the single dilution test revealed the appearance of antibody as early as 4 days post-immunization. The error terms of the single dilution test were relatively large as compared to those of the conventional test for antibody.Distinctly biphasic antibody responses were observed in ferrets following a single subcutaneous injection of distemper virus.  相似文献

柑桔类的严重病虫主要由苗木带入新区,造成危害。因而培育不带有这些病虫的苗木(简称无病虫苗木),在隔离地点建立果园并采取防疫措施,殊为预防新果区发生此类病虫危害的根本办法。(林孔湘1975) 1974年春,我们开始采用热水消毒种子和采用洗刷接穗芽条并用蒸汽和药剂消毒接穗芽条的方法,在隔离地点培育各种柑桔类的无病虫实生苗和嫁接苗。1976年第一批无病虫苗木出圃,栽植在各地。在隔离和防疫条件不同的情况下,这些新果园发生的病虫危害有明显差异。隔离及防疫条件较好的果园,一年之后检查,除潜叶蛾外,没有发现任何其他病虫。而与有病虫的柑桔树接近的或虽有隔离而不注意防疫的果园,很快或不久便受到各种病虫侵害。试验结果表明,用无病虫苗木在隔离地点建立果园,并采用严格防疫措施,可长期不发生严重病虫的危害,既可保证不会发生黄龙病和斑黄病等类似病毒病的危害,又可节省喷药防病虫的费用。这是在国内外尚未见报道的一项技术革新。  相似文献

An ideal national resistance monitoring program should deliver a precise estimate of the resistance situation for a given combination of bacteria and antimicrobial at a low cost. To achieve this, decisions need to be made on the number of samples to be collected at each of different possible sampling points. Existing methods of sample size calculation can not be used to solve this problem, because sampling decisions do not only depend on the prevalence of resistance and sensitivity and specificity of resistance testing, but also on the prevalence of the bacteria, and test characteristics of isolation of these bacteria. Our aim was to develop a stochastic simulation model that optimized a national resistance monitoring program, taking multi-stage sampling, imperfect sensitivity and specificity of diagnostic tests, and cost-effectiveness considerations into account. The process of resistance testing of Campylobacter spp. isolated from cloacal swab samples from poultry was modeled using a Markov Chain Monte Carlo model. Different sampling scenarios on the number of flocks to be tested, the number of birds from each flock, and the number of campylobacter colonies submitted to susceptibility testing were evaluated regarding the precision of the resulting prevalence estimate. Precision of the prevalence estimate was defined as the absolute difference between apparent and true prevalence of resistance. A partial budget approach was utilized to find the most cost-effective combination of samples to obtain a defined precision of the prevalence estimate. For a sampling scenario testing 100 flocks, five birds per flock, and one campylobacter colony per sample, the median error of the prevalence estimate was 2.5%, and 95% of the simulations resulted in an error of 7% or less. When the total number of samples was kept constant, maximizing the number of flocks tested, and only testing one bird per flock resulted in the most precise prevalence estimate. Submitting more than one campylobacter colony to resistance testing did not improve the prevalence estimate. Partial budget analysis indicated that the most cost-effective strategy was testing of two birds per flock, and submitting one colony per sample to resistance testing.  相似文献

Individual experimental animals used in our studies on bovine leukemia virus (BLV) are routinely screened for the presence of antibodies to the three bovine lymphotropic retroviruses. We utilized these screening methods to examine frozen sera from eight herds for antibodies to BLV, bovine visna virus (BVV) and bovine syncytial virus (BSV). Serum samples from 235 animals in four dairy and four beef herds were analyzed. Detection methods used included indirect fluorescent antibody tests of virus-infected cell cultures (BLV, BSV, BVV) and agar gel immunodiffusion (BLV). Sera from the BLV-infected animals in the dairy herds showed the highest single (50%, 49/97) and multiple (30%, 29/97) infections compared with 5% (7/138) and less than 1% (1/138), respectively in the beef herds. Single BVV infections were not detected in the dairy herds, but 11% (11/97) of the sera contained antibodies to BVV plus BLV or BSV. Five sera from beef cattle had antibodies only to BVV and four were obtained from one herd. Only one beef serum of the 138 tested demonstrated multiple antibodies (BLV, BVV).  相似文献

Our objective was to find herd factors associated with pigs testing seropositive for Salmonella. Data were collected from 359 finishing-pig herds in Germany, Denmark, Greece, The Netherlands and Sweden, between 1996 and 1998. Pigs fed non-pelleted feed (dry or wet) had 2- and 2.5-times lower odds of seropositivity, compared to pigs fed pelleted feed. The protective effect of non-pelleted feed over pelleted feed may be ascribed to the structure and composition. Also, pigs that were given whey (to drink or as the liquid part of the diet) had 2.6-times lower odds to test seropositive than pigs not getting whey. Pigs produced in batches in herds with hygienic-lock facilities had >3-times lower odds for testing seropositive compared to pigs in herds where only one or neither factor was present. In herds where the caretaker(s) washed hands consistently before tending to the animals, pigs had 1.5-times lower odds of seropositivity than pigs in herds where the caretaker did not. Pigs which were able to have snout contact with pigs in neighbouring pens (because pen separations were either open or too low) had 1.7-times higher odds to test seropositive compared to pigs for which such contact was prevented. Pigs in herds recruiting from more than three supplier herds had three-times higher odds to test seropositive than pigs in herds which breed their own replacement stock or recruit from a maximum of three supplier herds.  相似文献

Recent studies have documented that grapes and grape juices are equally cardioprotective as red wine. The existing reports implicate that the skin and seeds of the grapes containing polyphenolic antioxidants are instrumental for the cardioprotective properties of grapes. The present study examines if the flesh of grapes also possesses any cardioprotective abilities. Three groups of randomly selected rats were fed, water only (control), flesh of the grapes (2.5 mg/kg b. wt.) or the skins (2.5 mg/kg b. wt.) for 30 days. At the end of the 30 days, isolated perfused hearts were made ischemic for 30 min followed by 2 h of reperfusion in the working mode. The results demonstrated that both flesh and skin of the grapes could protect the hearts from ischemic reperfusion injury as evidenced by improved postischemic ventricular recovery and reduced myocardial infarct size. High performance liquid chromatography (HPLC) revealed that skin and flesh contained comparative amounts of glucose, fructose, tartaric acid, malic acid, shikimic acid, and trans-caftaric acid. In addition, the flesh contained reduced amounts (compared to skin) of cis-coutaric, trans-coutaric, caffeic, p-coumaric, cinnamics, and catechin/epicatechin. Total polyphenolic index was also lower in flesh compared to skin. The anthocyanins were present exclusively in the skin. Electron paramagnetic resonance (EPR) spectrometry of hydroxy radicals indicated that both flesh and skins possessed equal amount of ROS scavenging activities. Total malonaldehyde content in the heart was reduced comparatively with either flesh or skin. The results indicate for the first time that the flesh of grapes are equally cardioprotective as skin, and antioxidant potential of skin and flesh of grapes are comparable with each other despite of the fact that flesh does not possess any anthocyanin activities.  相似文献
