OBJECTIVE: To evaluate a method of aerobic bacteriologic culture of epidermal collarette specimens from dogs with superficial pyoderma and compare results with those for aerobic bacteriologic culture of abdominal skin specimens in healthy dogs. DESIGN: Prospective study. ANIMALS: 22 dogs with epidermal collarettes and 24 healthy dogs. PROCEDURE: Dry sterile cotton swabs were rolled across epidermal collarettes or hairless areas of abdominal skin in healthy dogs and submitted for aerobic bacteriologic culture. Hemolytic colonies of gram-positive-staining cocci were tested for catalase production, and if results were positive, a coagulase test was performed. Colonies with coagulase activity were tested for the ability to ferment mannitol. Antimicrobial susceptibility testing was performed on all Staphylococcus spp that were isolated. RESULTS: S. intermedius was isolated from collarettes in 18 of 22 dogs with superficial pyoderma but not from healthy dogs. Estimated sensitivity and specificity of the culture method were 81.8% and 100%, respectively. There were no significant differences in the ability to culture S. intermedius, the number of S. intermedius isolates without resistance to antimicrobials, and the number of S. intermedius isolates resistant to penicillin G when comparing dogs with superficial pyoderma for the first time and dogs with recurrent pyoderma, dogs that did or did not receive concurrent antimicrobials, and dogs with and without underlying allergic disease. CONCLUSIONS AND CLINICAL RELEVANCE: Bacteriologic culture of epidermal collarette specimens was a simple and reliable method for identification of S. intermedius in dogs with superficial pyoderma, regardless of history of pyoderma or current antimicrobial use. 相似文献

During the Near Earth Asteroid Rendezvous (NEAR)-Shoemaker's low-altitude flyover of asteroid 433 Eros, observations by the NEAR Laser Rangefinder (NLR) have helped to characterize small-scale surface features. On scales from meters to hundreds of meters, the surface has a fractal structure with roughness dominated by blocks, structural features, and walls of small craters. A selection program in three tropical maize populations aimed to improve tolerance of mid-season to late season drought environments
while maintaining grain yield (GY) potential. The selection process employed other attributes that included maintaining a
constant anthesis date (AD) and, under drought, shortening the anthesis-silking interval (ASI) and increasing ear number per
plant (EPP). Three-mode (genotypes × environments × attributes) pattern analysis, which consists of clustering and ordination,
should be able to collectively interpret these changes from ten evaluation trials. Mixture maximum likelihood clustering identified
four groups that indicated the populations' performance had changed with selection. Groups containing the advanced cycles
of selection were higher yielding in most environments and had lower ASI and higher EPP, particularly in drought environments.
Check entries with no selection for drought tolerance remained grouped with the initial cycles of selection. A 3 × 2 × 3 (genotypes
by environments by attributes) principal component model explained 70% of the variation. For the first environmental component,
ASI was shown to be highly negatively correlated with both GY and EPP while anthesis date (AD) was virtually uncorrelated
with other traits. The second environmental component (explaining 10% of the variation) contrasted droughted and well-watered
environments and showed that EPP and GY were better indicators of this contrast (in terms of changes in population performance)
than were AD or ASI. Three-mode analysis demonstrated that improvements with selection occurred in both droughted and well-watered
environments and clearly summarised the overall success of the breeding program.

To accurately assess the risks of metal contaminants in soil, the bioavailability of the metals need to be considered. The bioavailable concentrations determined from homogenized, dried soils are not necessarily representative of in situ conditions of undisturbed field soils. In this study, we investigated the accumulation of metals in wheat (Triticum aestivum) grown in undisturbed contaminated (Pb, Zn, Cu, and Cd) soil cores, in relation to total soil, leachate, and diffusive gradients in thin films (DGT)-labile metal concentrations. Despite the fact that many of the samples contained metal concentrations above guideline values, no significant effects were observed on wheat growth. The bioavailability of metals in the most contaminated samples was estimated to be medium to low, possibly explaining why few effects were observed in the bioassay. For Cu, Zn, and especially for Pb, leachate and DGT-labile concentrations were better predictors of uptake by wheat than total concentrations based on correlations. It is suggested that DGT and leachate concentrations in combination with bioassays in undisturbed soil cores can be used to account for metal bioavailability in soil. This study showed that particulate (i.e., physical) toxicity was responsible for rainbow trout deaths in bioassays with two separate solid wastes. This conclusion was based on: (1) fish necropsies which indicated physical damage to gills but no evidence of chemical damage to liver or kidney, (2) chemical analyses which indicated that levels of Priority Pollutants and other target compounds were too low to cause the observed toxicity, (3) structural and chemical analyses of the waste particles which showed that these consisted of inert materials, and (4) the use of centrifugation techniques to remove most of the suspended particulate material in bioassay tanks resulting in an elimination of most of the toxicity. The particles associated with the lethal effects were approximately 5 to 10 μm in size. Regulatory testing of solid wastes must distinguish physical and chemical toxicity since disposal options can vary depending on the mode of toxicity. For instance, chemical toxicity raises concern regarding leaching through soils into groundwater, whereas if physical particles are responsible for toxicity, such leaching is not of concern. 相似文献

Adult wild-trapped opossums were infected with Leishmania donovani (Khartoum strain, WR 378) and evaluated as an animal model of visceral leishmaniasis. All infected opossums died within 32 days. Loss of body fat, hepatomegaly, and petechiae of skin and abdominal musculature were seen at necropsy. Microscopically, numerous amastigote-laden macrophages were seen in histologic sections of liver, spleen, and lymph nodes; fewer parasite-laden macrophages were in the bronchial-associated lymphoid tissues and renal glomeruli. Hematological findings included thrombocytopenia (terminal), neutropenia, and lymphopenia. Blood lymphocyte blastogenesis in response to concanavalin A and phytohemagglutinin was decreased markedly at day 24 post-infection (PI). Serum antibodies (1:40 dilution) to promastigotes of L. donovani were detected in five of eight infected opossums tested on days 10 and 24 PI. Total bilirubin concentrations and alanine aminotransferase and aspartate aminotransferase activities were increased after day 25 PI. A major challenge in predicting Earth's future climate state is to understand feedbacks that alter greenhouse-gas forcing. Here we synthesize field data from arctic Alaska, showing that terrestrial changes in summer albedo contribute substantially to recent high-latitude warming trends. Pronounced terrestrial summer warming in arctic Alaska correlates with a lengthening of the snow-free season that has increased atmospheric heating locally by about 3 watts per square meter per decade (similar in magnitude to the regional heating expected over multiple decades from a doubling of atmospheric CO2). The continuation of current trends in shrub and tree expansion could further amplify this atmospheric heating by two to seven times. 相似文献
