Creatine phosphokinase in thyroid: isoenzyme composition compared with other tissues

Considerable creatine phosphokinase activity was demonstrated in thyroid glands of several species. After starch gel electrophoresis, thyroid exhibited a creatine phosphokinase isoenzyme pattern different from that of muscle, heart, and brain. The isoenzyme repertory of various tissues and a hybrid enzyme produced from muscle and brain enzyme are consistent with the postulated dimeric structure of creatine phosphokinase.     

Diisopropylphosphorofluoridate and Tabun: enzymatic hydrolysis and nerve function

Squid nerve contains an enzyme that hydrolyzes the nerve gas Tabun at about one-tenth the rate it hydrolyzes diisopropylphosphorofluoridate (DFP), and at about one-third to one-fourth the rate it hydrolyzes Sarin and Soman. Tabun is a more potent inhibitor of acetylcholinesterase than is DFP, is both lipid-and water-soluble, and penetrates readily into the squid giant axon in its inhibitory form. The failure of Tabun to block or markedly decrease the conducted action potential in the squid axon makes it likely that the blocking of conduction caused by DFP is probably not due to inhibition of acetylcholinesterase. Sub-strate specificity with regard to organophosphate metabolism by squid enzyme has possible implications for the disposal and detoxication of nerve gases in the ocean.

Complementary DNA sequencing: expressed sequence tags and human genome project

Automated partial DNA sequencing was conducted on more than 600 randomly selected human brain complementary DNA (cDNA) clones to generate expressed sequence tags (ESTs). ESTs have applications in the discovery of new human genes, mapping of the human genome, and identification of coding regions in genomic sequences. Of the sequences generated, 337 represent new genes, including 48 with significant similarity to genes from other organisms, such as a yeast RNA polymerase II subunit; Drosophila kinesin, Notch, and Enhancer of split; and a murine tyrosine kinase receptor. Forty-six ESTs were mapped to chromosomes after amplification by the polymerase chain reaction. This fast approach to cDNA characterization will facilitate the tagging of most human genes in a few years at a fraction of the cost of complete genomic sequencing, provide new genetic markers, and serve as a resource in diverse biological research fields.     

Autumn bloom of antarctic pack-ice algae

An autumn bloom of sea-ice algae was observed from February to June of 1992 within the upper 0.4 meter of multiyear ice in the Western Weddell Sea, Antarctica. The bloom was reliant on the freezing of porous areas within the ice that initiated a vertical exchange of nutrient-depleted brine with nutrient-rich seawater. This replenishment of nutrients to the algal community allowed the net production of 1760 milligrams of carbon and 200 milligrams of nitrogen per square meter of ice. The location of this autumn bloom is unlike that of spring blooms previously observed in both polar regions.

Adaptation of the membrane lipids of a deep-sea bacterium to changes in hydrostatic pressure

The fatty acid composition of the cell membrane of the barophilic marine bacterium CNPT3 was found to vary as a function of pressure. Greater amounts of unsaturated fatty acids were present in bacteria growing at higher pressures. The results suggest adaptations in the membrane lipids to environmentally relevant pressures. This response to pressure appears to be analogous to temperature-induced membrane adaptations observed in other organisms.     

High genetic diversity of Fusarium circinatum associated with the first outbreak of pitch canker on Pinus patula in South Africa

The disease known as pitch canker results from infection of Pinus species by the fungus Fusarium circinatum. This fungus also causes a serious root disease of Pinus seedlings and cuttings in forestry nurseries. Pinus radiata and P. patula are especially susceptible to the pathogen, but there are no records of pitch canker on P. patula in established plantations. To date, only planting material of this tree species in nurseries or in plantations at the time of establishment have been infected by F. circinatum. Symptoms of pitch canker have recently emerged in an established P. patula plantation in South Africa and this study sought to determine whether the symptoms were caused by F. circinatum. Isolates from cankers were identified as F. circinatum using morphology and DNA-based diagnostic markers. Microsatellite markers were then used to determine the genetic diversity of a collection of 52 isolates. The entire population included 17 genotypes representing 30 alleles, with a greater number of genotypes collected from younger (three- to six-year-old) than older (12- to 19-year-old) trees. Both mating types of F. circinatum were present, but no evidence of sexual recombination was inferred from population genetic analyses. This is the first record globally of pitch canker on P. patula trees in managed plantations. It is of significant concern to South Africa, where P. patula is the most important Pinus species utilised for plantation forestry.     

筇竹与黄皮树人工混交林中筇竹地上部分生物量模型构建

本研究以筇竹与黄皮树人工混交林中筇竹地上部分为研究对象,测定分析了1~4年生分株地上部分各构件生物量及含水率,建立人工筇竹分株地上部分各构件的生物量及总生物量模型,以期为人工筇竹林的经营管理及其碳汇项目的开发提供科学依据。结果表明：随着筇竹分株年龄的增加,各构件含水率和生物量均逐渐减少,筇竹1~4年生分株地上部分平均含水率分别为57.62%、53.40%、50.01%、42.66%,平均生物量分别为133.99、123.31、109.76、85.39 g/m²;各年龄分株地上部分生物量的分配均呈现出秆＞枝＞叶的变化规律。不同年龄分株的胸径与秆、枝、叶生物量及地上部分总生物量均有极显著相关性(P＜0.01)。以胸径为自变量建立的各年龄筇竹分株地上部分总生物量模型的决定系数(R²)均在0.93以上,具有较高的可信度,也有着较强的适用性,可用于类似立地条件下的筇竹分株生物量估测。
