Most plant-microbe interactions do not result in disease; natural products restrict non-host pathogens. We found that sulforaphane (4-methylsulfinylbutyl isothiocyanate), a natural product derived from aliphatic glucosinolates, inhibits growth in Arabidopsis of non-host Pseudomonas bacteria in planta. Multiple sax genes (saxCAB/F/D/G) were identified in Pseudomonas species virulent on Arabidopsis. These sax genes are required to overwhelm isothiocyanate-based defenses and facilitate a disease outcome, especially in the young leaves critical for plant survival. Introduction of saxCAB genes into non-host strains enabled them to overcome these Arabidopsis defenses. Our study shows that aliphatic isothiocyanates, previously shown to limit damage by herbivores, are also crucial, robust, and developmentally regulated defenses that underpin non-host resistance in the Arabidopsis-Pseudomonas pathosystem.  相似文献

Bioluminescence (light production) and fluorescence (re-emission of absorbed radiation as light) are found in an unaccountably diverse array of marine organisms, where their functions are largely unknown. Here we report a deep-sea siphonophore that twitches glowing lures to attract fish. This is rare evidence of bioluminescence used for prey attraction among nonvisual marine organisms. The lures also contain red fluorescent material that shifts the wavelength of emitted light. The existence of a red-luminescent invertebrate suggests that long-wavelength light plays a greater role in marine interactions than previously suspected.  相似文献

Hydrotalcites are used in technology as catalysts and anion exchangers and are important sinks for environmental contaminants. Their compositional variability makes it important, but difficult, to estimate their aqueous solubility. We report calorimetric measurements of the heats of formation of cobalt-aluminum hydrotalcite phases. The heat and free energy of formation from the elements are equal to those of mechanical mixtures of binary compounds, namely hydroxides and carbonates. The interlayer anion is much more important than the cation in determining the solubility of the hydrotalcite phase and its ability to contain or release heavy metals to the environment. Because hydrotalcites do not have an unreactive polymer as a structural core, their aqueous stability will change dramatically with composition, particularly with anion content. This simple mechanical mixture model allows prediction of aqueous solubilities and trace metal retention in a variety of geochemical settings.  相似文献

Chromosome banding analysis of human malignant melanoma has documented the nonrandom alteration of chromosome 6. To determine the relevance of chromosome 6 abnormalities in melanoma, a normal chromosome 6 was directly introduced into melanoma cell lines. The resulting (+6) microcell hybrids were significantly altered in their phenotypic properties in culture and lost their ability to form tumors in nude mice. The loss of the chromosome 6 from melanoma microcell hybrids resulted in the reversion to tumorigenicity of these cells in mice. The introduction of the selectable marker (psv2neo) alone into melanoma cell lines had no effect on tumorigenicity. These results support the idea that one or more genes on chromosome 6 may control the malignant expression of human melanoma.  相似文献

Local control of the domain orientation in diblock copolymer thin films can be obtained by the application of electric fields on micrometer-length scales. Thin films of an asymmetric polystyrene-polymethylmethacrylate diblock copolymer, with cylindrical polymethylmethacrylate microdomains, were spin-coated onto substrates previously patterned with planar electrodes. The substrates, 100-nanometer-thick silicon nitride membranes, allow direct observation of the electrodes and the copolymer domain structure by transmission electron microscopy. The cylinders aligned parallel to the electric field lines for fields exceeding 30 kilovolts per centimeter, after annealing at 250°C in an inert atmosphere for 24 hours. This technique could find application in nanostructure fabrication.  相似文献

About 240,000 square kilometers of Earth's surface is disrupted by mining, which creates watersheds that are polluted by acidity, aluminum, and heavy metals. Mixing of acidic effluent from old mines and acidic soils into waters with a higher pH causes precipitation of amorphous aluminum oxyhydroxide flocs that move in streams as suspended solids and transport adsorbed contaminants. On the basis of samples from nine streams, we show that these flocs probably form from aggregation of the epsilon -Keggin polyoxocation AlO4Al12(OH)24(H2O)12(7+)(aq) (Al13), because all of the flocs contain distinct Al(O)4 centers similar to that of the Al13 nanocluster.  相似文献

A vertically moving biofilm system (VMBS) was developed to treat wastewater. In this system, the biofilm grows on a biofilm module consisting of plastic media that is vertically and repeatedly moved up into the air and down into the water. The objectives of this study were to investigate the oxygen transfer efficiency and industrial wastewater treatment performance of the VMBS. The oxygen transfer coefficient (K L a) depended on the movement frequency (n) of the biofilm module and was proportional to n 1.67. K L a values measured were within the range of 0.0001 to 0.0027 s-1. The VMBS exhibited good carbonaceous removal when treating industrial wastewater produced in a factory manufacturing synthetic fibres. Removal efficiency of filtered chemical oxygen demand (COD) and biological oxygen demand (BOD5) was up to 93.2 and 97.9%, respectively. The volumetric removal rates of filtered COD and BOD5 reached 1320 g COD m-3 day-1 and 700 g BOD5 m-3 day-1. The areal organic removal rates, based on the surface area of the biofilm substrata, were 16 g BOD5 m-2 day-1 and 39 g COD m-2 day-1. No clogging occurred during the experiment. The mean areal biofilm mass increased with increasing the mean areal BOD5 removal rate. The new biofilm process has such advantages as high carbonaceous oxidation, energy saving, simpleconstruction and easy operation for industrial wastewater treatment.  相似文献

A monitoring program for Vibrio harveyi and other potential bacterial pathogens of summer flounder was conducted at two facilities in the Northeast United States. Bacterial samples were collected from larval and juvenile fish and live-feed, and identified using API 20E biochemical profiles and 16S rDNA sequencing. Histopathological examinations were conducted in order to relate histological changes with the presence of potential bacterial pathogens. V. harveyi, Vibrio ichthyoenteri, and Photobacterium damselae subsp. damselae, three known pathogens of flatfish, were isolated from diseased summer flounder. Although a high proportion of juvenile summer flounder showed microscopic signs of disease, the presence of these potential bacterial pathogens in fish was not associated with large-scale mortalities. An outbreak of flounder infectious necrotizing enteritis with 30% cumulative mortality occurred when juveniles were transported to a new facility. Isolates of V. harveyi from the disease outbreak were genetically different from the isolates from the commercial hatchery. V. harveyi isolates from both facilities were pathogenic to summer flounder by intraperitoneal injection. P. damselae subsp. damselae, V. ichthyoenteri, and Vibrio scophthalmi, also found in fish with gross lesions, were not pathogenic to juvenile summer flounder by intraperitoneal injection. Our research shows that several potential bacterial pathogens are associated with morbidity and mortality in summer flounder larvae and juveniles, especially in situations of stress. Increased knowledge about the environmental conditions that lead to disease, as well as the interactions of V. harveyi with other microbial species, could lead to the development of management strategies in summer flounder farms.  相似文献

Abstract.— Many aquaculture studies are conducted in relatively small research ponds and the results are then extrapolated to larger commercial ponds. Implicit in this research is the assumption that there is no relationship between pond size and phytoplankton composition. Study objectives were to assess phytoplankton composition and biomass by several methods in 17 channel catfish Ictalurus punctatus ponds at the Aqua‐culture Research Station, Louisiana Agricultural Experiment Station in Baton Rouge, Louisiana, USA. Pond size ranged from 0.04–0.60 ha. Sampling occurred weekly from 10 September – 1 October 1997. Water temperatures coincided with a transition from summer to fall‐winter conditions. Biomass was assessed by cell counts and quantification of photopigments. Concentrations of dissolved off‐flavor compounds (2‐methylisoborneol and geosmin) were assessed by gas chromatography/mass spectroscopy of water column samples. Cell count data showed differences in dominant species, biovolume, and diagnostic pigment signatures among ponds. The smaller ponds had more diverse phytoplankton composition compared to the larger ponds, whereas chlorophyll levels were nearly an order of magnitude lower in the smaller ponds. Ultraplanktonic (2–20 μm) unicellular cyanobacteria dominated the numerical counts on most sampling dates; however, biovolume transformations of cell count data reduced the dominance of this component relative to cryptophytes, diatoms, and filamentous cyanobacteria. Pigment and microscopic analyses were well correlated. Unialgal isolates of dominant taxa from these samples indicated the presence of at least five genera of off‐flavor producers in these ponds; these taxa included Anabaena, Aphanizomenon, Pseudanabaena, as well as two species of Oscillatoria. Care in extrapolating results from smaller‐sized research ponds to larger commercial ponds is warranted, as is the potential for taxa other than Oscillatoria and Anabaena in forming off‐flavor compounds.  相似文献
