Halkieriids and wiwaxiids are cosmopolitan sclerite-bearing metazoans from the Lower and Middle Cambrian. Although they have similar scleritomes, their phylogenetic position is contested. A new scleritomous fossil from the Burgess Shale has the prominent anterior shell of the halkieriids but also bears wiwaxiid-like sclerites. This new fossil defines the monophyletic halwaxiids and indicates that they have a key place in early lophotrochozoan history.  相似文献

This study deals with the development and validation of an original PCR protocol to assess the presence of Fasciola hepatica in Galba truncatula its main intermediate host in Western Europe. In the present study two DNA extraction techniques are compared and a new multiplex PCR is described. The Chelex(?) DNA extraction technique showed to be more appropriate than the classical Phenol/Chloroform/Proteinase K based method because of the absence of toxic organic solvent, shorter duration and lower cost, and a higher reproducibility regarding DNA concentrations and wavelength ratios. The multiplex PCR was set up to amplify the lymnaeid internal transcribed spacer 2 sequence (500-600 bp) that act as an internal control and a 124 bp Fasciola sp. sequence that is repeated more than 300,000 times in fluke whole genome. Ninety six snails were pooled and 6 snails (6.25%) found positive for Fasciola sp. The limit of detection is lower than the minimal biological infestation unit (one miracidium). DNA extracts from Paramphistomum daubneyi, Dicrocoelium lanceolatum, and Fascioloides magna did not cross react.  相似文献

Aqueous natural organic matter (NOM) impacted by two contrasting human impacts was analyzed using by multiresponse fluorescence, decoupled with the resolution routine PARAFAC. The first site is Chalk River, Ontario, Canada, near a pit formerly used to dispose low-level wastes. The second site is the Grand River in Cambridge, south-central Ontario, which is impacted by urban activities and agriculture. Our analysis included raw water, plus fractions from ultrafiltration and solid-phase extraction (SPE). The fluorescence spectra of the NOM, resolved with PARAFAC, showed three common features: humic-like components, at excitation/emission wavelengths 325?C350/450?C475 nm, fulvic-like components at 325/380?C420 nm and protein-like components, at 275/300 nm. Ultrafiltration revealed that most of the NOM comprised fine material below 5,000 Da cut-off (<4% of the total) in the urban-impacted sites and the clean site at Chalk River, but the colloidal fraction (larger than 5,000 Da) was substantially higher in the contaminated water, with ??18?C26% of the total. The protein-like components in the contaminated Chalk River water were affected by ultrafiltration, but less so in the clean Chalk River sample and the urban-impacted waters. SPE preferentially removed the protein-like component in the contaminated Chalk River water (typically 89?C95% signal decrease), but had a limited effect on humic- and fulvic-like components elsewhere. In conclusion, multiresponse fluorescence provided new information on the NOM quality from two contrasting sites, aided by ultrafiltration and SPE. These results are consistent with the in situ production of NOM in the Chalk River contaminated site, and natural production at the other sites.  相似文献

Reasons for performing the study: The conventional arthroscopic approach to the palmar/plantar aspect of the distal interphalangeal joint (DIPJ) may result in the inadvertent penetration of the digital flexor tendon sheath (DFTS) and the navicular bursa (NB). This iatrogenic communication would be undesirable subsequent to arthroscopic lavage of a septic DIPJ. Hypothesis: A lateral/medial approach to the palmar/plantar aspect of the DIPJ will result in a significantly lower rate of inadvertent penetration of the DFTS and NB, whilst still providing adequate intra‐articular evaluation. Methods: The conventional palmar/plantar approach or a novel lateral/medial approach to the DIPJ was performed on cadaver fore‐ and hindlimbs (30 limbs/approach). Subsequently, India ink was injected into the dorsal pouch of the DIPJ, and the DFTS (n = 60) and NB (n = 20) were examined for the presence/absence of ink. In addition, observations of the number of attempts made to access the joint, evidence of iatrogenic intra‐articular trauma and occurrence of incomplete visualisation of the palmar/plantar pouch were recorded. Results: With the conventional approach, DFTS penetration was noted in 18/30 (60%) of the limbs, compared to 1/30 (3.3%) with the lateral/medial approach (P≤0.001). NB penetration was seen in 5/10 limbs with the palmar/plantar approach compared to 0/10 with the lateral/medial approach (P = 0.01). No significant differences were found between the approaches in the number of attempts made to access the joint, the incidence of iatrogenic intra‐articular trauma, or the occurrence of incomplete visibility of the palmar/plantar pouch. Conclusions: The novel lateral/medial approach to the DIPJ significantly decreases the risk of inadvertent penetration of the DFTS and NB. Potential relevance: The novel lateral/medial approach to the DIPJ is an effective technique to gain access to the palmar/plantar pouches, and is particularly advantageous for arthroscopic lavage of a septic DIPJ.  相似文献

The precise determination of the embryonic chronology is very important in reproductive biotechnologies, especially in estimating embryonic age. Thus, there is a need for greater knowledge and standardization for determining the chronology of embryonic development and functional morphology. We describe aspects of embryonic development in two domestic carnivores to add knowledge about organ peculiarities and for application in veterinary practice, in prenatal development and in the biotechnology fields. We found that the development of differential characteristics of embryonic organs occurs in the first trimester of pregnancy for both species. Thus, using the combination of the crown‐rump length, macroscopic analysis and optical microscopy, it is possible to predict gestational age more precisely in animals that lack a defined breed and establish an embryonic pattern.  相似文献

The potential for colloid-facilitated migration in contaminated sites is well known, and remediation techniques such as ultrafiltration are often considered for contaminant removal. Although this approach could be successful, the stability of the contaminant species in groundwater and the removal efficiency need to be investigated to ensure proper decontamination of moving aquifers. In our study, we have sampled contaminated groundwater near a former radioactive liquid disposal area at Chalk River, Ontario, Canada. Samples were taken in 2002 and 2004 to determine the behaviour of radiocontaminants by size fractionation using ultrafiltration, with emphasis on 60Co and 137Cs. The contaminant concentrations varied significantly for both contaminants in the two samples (34.5 and 25.5 Bq/l for 60Co, 25.5 and 97.2 Bq/l for 137Cs). On the other hand, the size fractionation (5,000 Daa nominal cut-off) remained consistent between the 2002 and 2004 samples, as most of the 60Co (72%–83%) remained in the filtrate, while almost all of the 137Cs (>98%) was retained along with the colloidal-sized material. Release of 60Co and 137Cs from the colloidal material yielded desorption coefficients (K D-des) of 7.8 × 105 and 1.7 × 108 ml/g for 60Co and 137Cs, respectively.  相似文献

In arid zones, the use of compost for plant production in forest nurseries is hindered by a lack of water. The main objectives of this study were (1) to evaluate the physical stability of composts produced from shredded branches of Acacia cyanophylla and A. cyclops subjected to a repeated drying and wetting cycles, similar to those used at the operational scale in nurseries in arid regions and to compare these composts with a standard peat-vermiculite (PV) substrate; (2) to identify the relevant substrate physical variables that correlate with seedling growth. Carob (Ceratonia siliqua L.) was cultivated during a production cycle of 27 weeks in a completely randomized block experiment. Substrate physical variables were measured at the beginning, middle and end of the experiment. Seedling growth variables were evaluated over the course of the production cycle, while gas exchange and water-relation variables were measured during a wetting and drying cycle at the end of the experiment. All three substrates produced vigorous seedlings with well-developed root systems that colonized the entire root plug. The growth of seedlings produced in the PV substrate was better than those grown in the compost-based substrates. No significant differences in gas exchange capacities and water relation variables were observed among the three substrates at the end of experiment with the exception of net photosynthesis, which was higher for the PV substrate at high substrate matric potential. Excessive drainage was negatively correlated with growth variables while water availability in the early growth phase and air porosity towards the end of experiment were positively correlated. Performance of the two composts could be increased by improving their initial structure and stability and by adjusting the irrigation regime.  相似文献
