Background

The possibility for commercial mining of deep-sea manganese nodules is currently under exploration in the abyssal Clarion-Clipperton Fracture Zone. Nematodes have potential for biomonitoring of the impact of commercial activity but the natural biodiversity is unknown. We investigate the feasibility of nematodes as biomonitoring organisms and give information about their natural biodiversity.

Results

The taxonomic composition (at family to genus level) of the nematode fauna in the abyssal Pacific is similar, but not identical to, the North Atlantic. Given the immature state of marine nematode taxonomy, it is not possible to comment on the commonality or otherwise of species between oceans. The between basin differences do not appear to be directly linked to current ecological factors. The abyssal Pacific region (including the Fracture Zone) could be divided into two biodiversity subregions that conform to variations in the linked factors of flux to the benthos and of sedimentary characteristics. Richer biodiversity is associated with areas of known phytodetritus input and higher organic-carbon flux. Despite high reported sample diversity, estimated regional diversity is less than 400 species.

Conclusion

The estimated regional diversity of the CCFZ is a tractable figure for biomonitoring of commercial activities in this region using marine nematodes, despite the immature taxonomy (i.e. most marine species have not been described) of the group. However, nematode ecology is in dire need of further study.

This study was conducted to determine whether the daily consumption for 84 days of tyrosol and hydroxytyrosol, the main olive oil phenolic compounds, and olive oil mill wastewater (OMWW), a byproduct of olive oil production, rich in micronutrients, may improve bone loss in ovariectomized rats (an experimental model of postmenopausal osteoporosis) and in ovariectomized rats with granulomatosis inflammation (a model set up for senile osteoporosis). As expected, an induced chronic inflammation provoked further bone loss at total, metaphyseal, and diaphyseal sites in ovariectomized rats. Tyrosol and hydroxytyrosol prevented this osteopenia by increasing bone formation ( p < 0.05), probably because of their antioxidant properties. The two doses of OMWW extracts had the same protective effect on bone ( p < 0.05), whereas OMWW did not reverse established osteopenia. In conclusion, polyphenol consumption seems to be an interesting way to prevent bone loss.  相似文献

Duane's retraction syndrome (DRS) is a complex congenital eye movement disorder caused by aberrant innervation of the extraocular muscles by axons of brainstem motor neurons. Studying families with a variant form of the disorder (DURS2-DRS), we have identified causative heterozygous missense mutations in CHN1, a gene on chromosome 2q31 that encodes alpha2-chimaerin, a Rac guanosine triphosphatase-activating protein (RacGAP) signaling protein previously implicated in the pathfinding of corticospinal axons in mice. We found that these are gain-of-function mutations that increase alpha2-chimaerin RacGAP activity in vitro. Several of the mutations appeared to enhance alpha2-chimaerin translocation to the cell membrane or enhance its ability to self-associate. Expression of mutant alpha2-chimaerin constructs in chick embryos resulted in failure of oculomotor axons to innervate their target extraocular muscles. We conclude that alpha2-chimaerin has a critical developmental function in ocular motor axon pathfinding.  相似文献

Annual grasses are stronger competitors for available soil N than blue oak seedlings and soil microorganisms. However, little is known about the dynamics of N competition during annual grass senescence. We conducted a field experiment in a California oak woodland to study effects of annual grass senescence on N uptake by grasses, blue oak seedlings and soil microorganisms. Labeled N was applied at the beginning of April, May and of June in the form of 15NH4+ or 15N-glycine. Plants and soils were harvested after 5 days (15NH4+ and 15N-glycine treatments) and after 26 days (15NH4+ treatment only). We evaluated effects of N form, season and labeling period on N competition among oak seedlings, annual grasses and soil microorganisms. N forms did not affect competition among grasses, oak seedlings and soil microorganisms, but more 15N was incorporated into the soil organic N pool in the 15N-glycine treatments than in the 15NH4+ treatments. There were no seasonal (May vs June) effects on 15N recovery in blue oak seedlings and soil microorganisms. Plant samples from April harvest were lost. In June, when grasses were senescing, more 15N was found in the soil inorganic pool than in May. Extremely dry soils in June may have limited inorganic N availability to oak seedlings and soil microorganisms. After 26-day labeling period, 15N recovery in blue oak seedlings and the soil organic N pool significantly increased, while 15N recovery in both the soil microbial and inorganic N pools decreased compared to the 5-day labeling period. Although blue oak seedling biomass changed little from early May to late June, N concentrations in oak roots increased 53%. In contrast, annual grass biomass peaked in May, and then decreased rapidly. Our results suggest that blue oak seedlings and annual grasses have different temporal competitive abilities. Blue oak seedlings appear to have a long-term strategy for N competition. Blue oaks take up N slowly but steadily, increasing N uptake from 5 to 26 days. This extended time period has a greater positive effect on N uptake than does reduced grass uptake caused by senescence.  相似文献

The aim of this work was to determine whether a cervical dilation protocol (CDP) composed of only oxytocin can be used to perform transcervical (non-surgical) embryo transfer in sheep (NSET) without affecting the viability of the corpus luteum (CL). Likewise, we evaluated whether a cervical transposing test with a Hegar dilator (CT Hegar test), performed at oestrous time, could be used to screen ewes for NSET (greater or lower chances to transpose the cervix). For that, oestrous and ovulation synchronization was performed in 25 Santa Inês ewes to induce the dioestrous condition. Animals went through the following CDP in a crossover design: E + OX, oestradiol benzoate (100 µg intravenously [IV]) and oxytocin (100 IU IV); OX, oxytocin (100 IU IV); and SAL, saline solution (IV). Using a Hegar dilator, cervical transposing attempts were performed at oestrous (D0) and dioestrous time (D8). The viability of the CL (morphology, luteal blood flow and progesterone values) was evaluated by ultrasonography (colour Doppler and B-mode) and by serum progesterone measurement from D7 to D13. The cervical transposing rate was lower for the SAL group (64%; 16/25; p < .05) and did not differ between the E + OX (88%; 22/25, p > .05) and OX (84%; 21/25, p > .05) groups. No treatment affected the CL viability. The CT Hegar test showed a high sensitivity (85.7%–93.3%), satisfactory accuracy (72%–84%), low false-negative rate (6.7%–14.6%), but high false-positive rate (46%–66.7%). In conclusion, a CDP protocol composed exclusively of oxytocin can lead to good cervical transposing rates and does not affect the viability of the CL. In addition, a screening test (CT Hegar) performed at oestrus can identify ewes for which cervical transposing will likely not occur at NSET.  相似文献

The factors affecting herring recruitment are still poorly understood, complicating the prediction of stock dynamics and the choice of operational management strategies. We investigated effects of intrinsic (SSB) and extrinsic factors (physical and biological environments, including competition and predation) on recruitment of the spring and fall spawning components of each of the two herring stocks occurring in the Gulf of the St. Lawrence between 1971 and 2014. Effects of potential explanatory factors on recruit (age 2) abundance were tested using Generalized Additive Models. Model fit was significantly improved by incorporating both physical and biological environmental variability, but effects of herring SSB and predation were not significant. Indices of zooplankton abundance and phenology explained more variance in recruitment than physical indices. Our results emphasize the dominance of bottom‐up processes over SSB in the regulation of herring recruitment. Environmental variability did not seem to act uniformly on the recruitment of either stock or their respective spawning components. A long‐term trend of decreasing recruitment in spring spawners was associated with a long‐term decline in abundance of cold water copepods. In fall spawners, optimal recruitment was dependent on warmer environmental conditions combined with an adequate supply (species composition and phenology) of zooplankton. These results provide the first empirical evidence that spring and fall spawning herring are adapted to contrasting environmental conditions and shed light on the potential mechanisms linking herring recruitment to key zooplankton community characteristics and phenology. Management strategies can be improved by incorporating this new knowledge on environmental drivers of herring recruitment.  相似文献

As an important component of organic fertilizers, animal faeces require methods for determining diet effects on their microbial quality to improve nutrient use efficiency in soil and to decrease gaseous greenhouse emissions to the environment. The objectives of the present study were (i) to apply the chloroform fumigation extraction (CFE) method for determining microbial biomass in cattle faeces, (ii) to determine the fungal cell-membrane component ergosterol, and (iii) to measure the cell-wall components fungal glucosamine and bacterial muramic acid as indices for the microbial community structure. Additionally, ergosterol and amino sugar data provide independent control values for the reliability of the microbial biomass range obtained by the CFE method. A variety of extractant solutions were tested for the CFE method to obtain stable extracts and reproducible microbial biomass C and N values, leading to the replacement of the original 0.5 M K2SO4 extractant for 0.05 M CuSO4. The plausibility of the data was assessed in a 28-day incubation study at 25 °C with cattle faeces of one heifer, where microbial biomass C and N were repeatedly measured together with ergosterol. Here, the microbial biomass indices showed dynamic characteristics and possible shifts in the microbial community. In faeces of five different heifers, the mean microbial biomass C/N ratio was 5.6, the mean microbial biomass to organic C ratio was 2.2%, and the mean ergosterol to microbial biomass C ratio was 1.1‰. Ergosterol and amino sugar analysis revealed a significant contribution of fungi, with a percentage of more than 40% to the microbial community. All three methods are expected to be suitable tools for analysing the quality of cattle faeces.  相似文献

In order to investigate the pathogenesis of neosporosis following a primary infection in late pregnancy, cattle were subcutaneously challenged with 5 × 108Neospora caninum (NC1 isolate) tachyzoites at day 210 of gestation and serial necropsies were then carried out at 14, 28, 42 and 56 days post-infection (dpi). No abortions occurred and all the foetuses were viable at the time of euthanasia. There was a high rate of vertical transmission, as parasites were detected by immunohistochemical labelling and PCR in all the foetuses from 28 dpi. Focal necrotic lesions were observed in the placentomes of the placenta from 28 dpi and showed resolution during later time points, denoted by infiltration of inflammatory cells at 42 dpi and fibrosis at 56 dpi. Foetuses at 28 and 42 dpi showed scarce and isolated lesions which are unlikely to represent a threat to foetal viability. No lesions were observed in the foetuses at 14 or 56 dpi suggesting control of the infection and resolution of the lesions by maternal and foetal immune responses. Once infection was established, it could not be cleared from the host and vertical transmission of the parasite occurred in all infected hosts. Parasite was detected in the placenta at 28 dpi, while in previous experimental infections of cattle at day 70 and 140 of gestation using the same challenge model, it was already present at day 14 post infection. This suggests that a change in the maternal immune response plays a crucial role in limiting the initial infection during the last term of pregnancy.  相似文献

The response of four naturally occurring biotypes of Epilobium ciliatum and four sources of Senecio vulgaris to the herbicide atrazine were compared with their susceptibility to the powdery mildews Sphaerotheca epilobii and Erysiphe cichoracearum. Biotypes that were resistant to atrazine were also resistant to mildew. Mechanisms that might explain the association between atrazine resistance and mildew resistance are discussed, along with possible implications of these findings for farmland ecology and for the production of herbicide- and mildew-resistant crop plants.  相似文献
