ABSTRACT: Susceptibility of sheep to scrapie, a transmissible spongiform encephalopathy of small ruminants, is strongly influenced by polymorphisms of the prion protein gene (PRNP). Breeding programs have been implemented to increase scrapie resistance in sheep populations; though desirable, a similar approach has not yet been applied in goats. European studies have now suggested that several polymorphisms can modulate scrapie susceptibility in goats: in particular, PRNP variant K222 has been associated with resistance in case-control studies in Italy, France and Greece. In this study we investigated the resistance conferred by this variant using a natural Italian goat scrapie isolate to intracerebrally challenge five goats carrying genotype Q/Q 222 (wild type) and five goats carrying genotype Q/K 222. By the end of the study, all five Q/Q 222 goats had died of scrapie after a mean incubation period of 19 months; one of the five Q/K 222 goats died after 24 months, while the other four were alive and apparently healthy up to the end of the study at 4.5 years post-challenge. All five of these animals were found to be scrapie negative. Statistical analysis showed that the probability of survival of the Q/K 222 goats versus the Q/Q 222 goats was significantly higher (p = 0.002). Our study shows that PRNP gene mutation K222 is strongly associated with resistance to classical scrapie also in experimental conditions, making it a potentially positive target for selection in the frame of breeding programs for resistance to classical scrapie in goats.  相似文献

Domestic cats are preferred models for normal physiology and several human diseases. In the present study feline fetal fluids and membranes were evaluated as possible sources of MSCs. Samples were recovered from 4 pregnant queens after ovarian-hysterectomy. Gestational sacs were separated from uterine wall; after allantoic and amniotic fluids aspiration and chorion-allantois and amniotic membranes separation, all cell lineages were cultured into 25 cm(2) flasks, in DMEM/TCM199, in a 5% CO(2) incubator at 38.5 °C. At passage 3, chondrogenic, osteogenic and adipogenic differentiation ability were evaluated by culturing cell monolayers in differentiating media for 21 days. Cellular characterization with CD90, CD44, CD105, CD73, CD34, CD14, CD45, was performed by flow cytometry. In all samples, adherent fibroblastoid spindle-shaped cells were observed. Positive von Kossa and Alizarin Red staining confirmed osteogenesis. Alcian blue staining of matrix glycosaminoglycans illustrated chondrogenesis, and positive Oil Red O lipid droplets within cell cytoplasm suggested adipogenesis. All cell lines isolated were positive for CD90, CD44, CD105 and negative for CD34, CD14 and CD45; as unexpected and different from human cells, feline cells resulted negative for CD73. Based on this preliminary results, fetal fluids and membranes could represent an alternative sources for mesenchymal stem cells in feline species.  相似文献

The capability of native bacterial strains isolated from Lolium perenne rhizosphere to behave as plant growth promoting bacteria and /or biocontrol agents was investigated. One strain (BNM 0357) over 13 isolates from the root tips of L. perenne resulted proved to be nitrogenase positive (ARA test) and an IAA producer. Conventional tests and the API 20E diagnostic kit indicated that BNM 0357 behaves to the Enterobacteriaceae family and to the Enterobacter genus. Molecular identification by 16S rRNA sequence analysis indicated that BNM 0357 had the highest similarity to Enterobacter ludwigii (EN-119). Isolate BNM 0357 had the capability to solubilize calcium triphosphate and to antagonize Fusarium solani mycelial growth and spore germination. Strain BNM 0357 also showed the ability to improve the development of the root system of L. perenne. This study disclosed features of E. ludwigii BNM 0357 that deserve further studies aimed at confirming its putative importance as a PGPR.  相似文献

Developing long-term chronologies of tree-ring anatomical features to evaluate climatic relationships within species might serve as an annual proxy to explore and elucidate the climatic drivers affecting xylem differentiation. Pinus leucodermis response to climate was examined by analyzing vertical xylem resin ducts in wood growing at high elevation in the Apennines of peninsular Southern Italy. Early- and latewood tree-ring resin duct chronologies, spanning the 1804–2010 time period, were constructed. We analyzed the relationships between resin duct chronologies and climate over the last century using correlation and response function analyses. Overall, results showed that ring width and resin duct relationships differed between early- and latewood, which indicated conditions affecting growth were not associated with resin duct production. Results also revealed differential responses to climate between early- and latewood resin duct chronologies. A notable observation was a positive and stable relationship between latewood resin duct number chronology and July maximum temperature throughout the twentieth century. This result suggested resin ducts might be a suitable proxy to evaluate P. leucodermis response to climate in the study area.  相似文献

Monospecific and bispecific genera are of special concern as they represent unique phylogenetic/evolutionary trajectories within larger clades. In addition, as phylogenetically older taxa are supposed to be exposed to higher rarity and extinction risk, monospecific and bispecific genera may be intrinsically more prone to extinction risks than multispecies genera, although extinction risks also depend on the ecological and biological strategy of the species. Here, the distribution across biogeographical zones and the levels of threat to 2 speciose orders of mammals (monospecific and bispecific genera of Rodentia and Soricomorpha) are investigated in order to highlight major patterns at the worldwide scale. In Rodentia, 39.7% of the genera (n = 490) were monospecific and 17.9% were bispecific. In Soricomorpha, 44.4% of the total genera (n = 45) were monospecific and 15% were bispecific. There was a positive correlation between the number of monospecific genera and the total number of genera per family. Peaks of monospecific and bispecific genera richness were observed in Neotropical, Oriental and Afrotropical regions in rodents and in the Palearctic region in soricomorphs. Range size was significantly uneven across biogeographic region in rodents (with larger ranges in Nearctic and Oriental regions and smaller ranges in the Australian region), but there was no difference across biogeographic regions in terms of range size in soricomorphs. Most of the monospecific and bispecific genera occurred in forest habitat in both taxa. The frequency distribution of the monospecific and bispecific genera across IUCN categories did not differ significantly from the expected pattern using the total rodent genera and the multispecies genera.  相似文献

The ingestion and retention of microplastics of filter-feeder organisms represent a risk for the final consumers and the environment. Biomonitoring is necessary to deal with the effects of plastic material pollution. The selection of the monitored organisms strongly affects the relevance of the results and the understanding of the environmental conditions. The results discussed in this paper highlight the differences in the estimate of microplastic pollution depending on the species subject of study. Ascidia spp. specimens retained a value five-fold higher (0.62 MP/g) than bivalve species (Crassostrea gigas 0.11 MP/g; Mytilus galloprovincialis 0.05 MP/g; Anomia ephippium 0.12 MP/g).  相似文献

Notwithstanding the wide range of biological and pharmacological activities reported for sweet basil (Ocimum basilicum L.), many discrepancies are still present in the evaluation of its health-promoting properties. These discordances could be at least in part due to insufficient details of qualitative and quantitative composition, connected to the ample variability of this species. Furthermore, many investigations have been carried out in vitro, with few data available on the effectiveness in biological systems. In this study, the protective effect of essential oils and water-soluble extracts derived from three different cultivars of sweet basil has been evaluated in cultured cardiomyocytes. To verify the effectiveness of supplemented oils/extracts in counteracting oxidative damage, cardiomyocytes were stressed by the addition of hydrogen peroxide. The results indicate that (a) in vitro antioxidant activity is not predictive of biological activity and (b) basil can yield extracts with substantially different protective effects, in relation to composition and extraction techniques. Variation among different cultivars has also been detected.  相似文献

The catalytic behavior of a mixture of pectic enzymes, covalently immobilized on different supports (glass microspheres, nylon 6/6 pellets, and PAN beads), was analyzed with a pectin aqueous solution that simulates apple juice. The following parameters were investigated: the rate constant at which pectin hydrolysis is conducted, the time (tau(50)) in which the reduction of 50% of the initial viscosity is reached, and the time (tau(comp,dep)) required to obtain complete depectinization. The best catalytic system was proven to be PAN beads, and their pH and temperature behavior were determined. The yields of two bed reactors, packed or fluidized, using the catalytic PAN beads, were compared to the circulation flow rate of real apple juice. The experimental conditions were as follows: pH 4.0, T = 50 degrees C, and beads volume = 20 cm(3). The initial pectin concentration was the one that was present in our apple juice sample. No differences were observed at low circulation rates, while at higher recirculation rates, the time required to obtain complete pectin hydrolysis into the fluidized reactor was found to be 0.25 times smaller than in the packed bed reactor: 131 min for the packed reactors and 41 min for the fluidized reactors.  相似文献

Paxillin and p130CAS are two adaptor proteins localized at the focal adhesions which play an important role in cell signaling, cell motility and oncogenic transformation. In this study we evaluated the levels of paxillin and p130CAS in feline and canine mammary tumor tissues at different stages of malignancy. The results obtained by Western blotting analysis showed no significant differences in the amounts of paxillin and p130CAS between normal and non-invasive tumor tissues. By contrast, mammary tumor tissues with the invasive phenotype showed lower levels of paxillin P < 0.01 and higher levels of p130CAS P < 0.001 than normal tissues. The decrease P < 0.001 of the amount of paxillin and the increase P < 0.001 of p130CAS levels were correlated with the progression stage of malignancy. Since paxillin and p130CAS are involved in regulating cell migration, our results suggest that low levels of paxillin together with high levels of p130CAS expression may cause certain breast cancers to be more motile and possibly more aggressive. Thus, both paxillin and p130CAS may represent useful prognosticators of feline and canine breast cancer malignancy.  相似文献
