Two interferon gamma (IFN-gamma) assays, the IFN-gamma enzyme immunoassay (EIA) and the IFN-gamma bioassay and an absorbed ELISA were used to screen 6 cattle herds for Johne's disease. Each herd had a history of Johne's disease but the majority of infected animals did not show clinical signs. The disease status of the cattle, which were removed from the herds, was confirmed by bacteriological culture of faeces or histopathological examination and culture of tissues collected at necropsy. The sensitivities of the IFN-gamma assays and the absorbed ELISA were determined using test results from infected animals. The sensitivity of the IFN-gamma EIA in detecting subclinical (71.8 to 93.3%) and clinical animals (100%) was not significantly different. However, the IFN-gamma bioassay and the absorbed ELISA were more sensitive in detecting cattle with advanced infections (80%) than those that were subclinically affected (16.7 to 33.3%).  相似文献

There is no doubt that nutrient deficiencies and excesses can influence immune responses. Despite advances in the development of therapeutic agents, a competent immune system is crucial for prevention and resolution of disease. Leukocytes are some of the most metabolically active cells in animals. We know that growing animals require nutrients to sustain growth and reach their full potential, whether that is size, strength, speed, or agility. We do not expect kittens to grow or greyhounds to run without provision of adequate nutrients. To expect the immune system to function in the face of nutrient deficiency neglects our knowledge of the physiological needs of metabolically active tissues. Immune responses may be modified through dietary manipulations. However, the clinician must answer the question "to what end or consequence will the response be changed?" No "golden" nutrient ensures an appropriate immune response in all conditions, just as no "golden" drug heals all diseases. Our knowledge of nutrient-immune system interactions is increasing. Yet, for all of the current knowledge of nutrient effects on immunity, the words of Dr R.K. Chandra hold sterling advice, "Moderation is a good dictum in biology and medicine, and it applies equally to nutritional immunology."  相似文献

大豆起源于中国,中国是世界上大豆遗传资源最为丰富的国家,也曾经是世界上最主要的大豆生产国和出口国。半个多世纪以来,科学技术的进步带动了一些美洲国家大豆产业的迅猛发展,中国的大豆生产量由世界首位降到美国、巴西之后,从大豆出口国变为大豆净进口国,大豆成为中国农产品贸易中最令国际社会关心的贸易作物。随着农业生物技术的快速发展,知识产权保护的法律法规的国际化,尤其是我国加入WTO以后,大豆研究、开发、生产领域遇到了来自比其他作物前所未有的挑战。作为我国最重要的植物蛋白、食用油及动物饲料来源的大豆作物,有效地利用有关的法律法规保护我国大豆种质资源及新品种已经迫在眉睫。本文着重介绍了与大豆遗传资源、新品种保护相关的我国法律法规,进行了大豆保护的可专利性分析,并对大豆研发过程中的知识产权保护问题作了初步探讨,以促进对大豆新品种的申请保护,提高我国应对来自国外竞争的抗衡能力。  相似文献

An in vitro cellular assay for bovine tuberculosis has recently been developed. This assay detects gamma-interferon released in response to specific antigen in a whole blood culture system. The bio-assay previously described for the detection of bovine gamma-interferon (IFN-gamma) has now been replaced with a sandwich enzyme immunoassay (EIA) which utilises two monoclonal antibodies to bovine IFN-gamma. The EIA detects less than 25pg/ml of recombinant bovine IFN-gamma and is specific for biologically active bovine IFN-gamma; and does not detect bovine alpha or beta interferon. IFN-gamma from sheep, goat and buffalo, but not from pig, deer or man, are also recognised by the EIA. The bovine IFN-gamma EIA when used in conjunction with the whole blood culture system has resulted in a simple, rapid and sensitive in vitro assay for specific cell mediated immune responsiveness to M. bovis infection in cattle.  相似文献

The sensitivity of the abattoir inspection procedure introduced for Australian export beef in 1976 was compared to a detailed necropsy procedure for the detection of tuberculous lesions in cattle. In a sample of cattle that were reactors to the tuberculin test, abattoir inspection failed to detect an estimated 47% of cattle with lesions. The detailed necropsy examination of cattle with lesions of tuberculosis identified 21 sites of infection compared with 13 to 18 in cattle examined by routine meat inspection procedures. Of the lesions detected during detailed necropsy, 15.9% did not involve the thoracic cavity or the medial retropharyngeal lymph nodes. The failure to detect lesions during abattoir inspection has its greatest significance in an animal with a single lesion. If the 245 cattle found with single lesions during detailed necropsy had been examined by abattoir inspection using the 1976 or the 1986 procedures, 0.8 and 8.9%, respectively, of these animals would not have been detected because the diseased tissues would not have been examined. If meat inspection is to provide an effective means of monitoring the level of bovine tuberculosis during the final stages of eradication, a procedure no less sensitive than that introduced in 1976 should be used.  相似文献

A 21-month-old domestic Hanford pig (Sus scrofa domestica) in a 1-year study for experimental myocardial infarction was euthanized at the end of the study. One week earlier, the animal had symptoms and elevated clinical chemistry results suggestive of hepatobiliary disease, which resolved after medical therapy. At necropsy, the gallbladder was markedly enlarged, discolored, and had a thickened wall. Within the gallbladder, there was abundant friable green-brown material. A culture of the gallbladder luminal material yielded Clostridium perfringens type A. Histopathology of the gallbladder demonstrated multifocal areas of necrosis of varying depths, admixed with an inflammatory infiltrate that was also observed on the serosa and within the associated adipose tissue. Luminal material was composed of cellular debris and bile sludge admixed with numerous bacterial rods. Smooth-muscle hypertrophy of numerous small arterioles with narrowed lumina was observed in the gallbladder. A diagnosis of acalculous cholecystitis presumably because of ischemia of the gallbladder with secondary clostridial infection was made. To the authors' knowledge, this is the first reported case of acalculous cholecystitis with evidence of vascular compromise in a pig, as well as cholecystitis secondarily attributed to Clostridium perfringens type A.  相似文献

Various freshwater and marine algal toxins are known to affect plants, fishes, mammals, and invertebrates. During recent mortality events in Texas white shrimp aquaculture ponds, water and shrimp tissue samples were analyzed for cyanobacterial toxins and found to contain microcystin-LR. Cyanoprokaryota dominated the phytoplankton assemblage in water from the affected pond, particularly Microcystis aeruginosa and Anabaena sp. Water samples from the affected pond also contained high levels of microcystin-LR (45 μg/l), whereas adjacent ponds had a diatom-green algal assemblage and no measurable toxin. Unialgal isolates of M. aeruginosa from the affected pond produced microcystin-LR. Free microcystin-LR concentrations in dead shrimp hepatopancreas determined by HPLC were 55 μg/g total shrimp weight, whereas shrimp hepatopancreas from the adjacent pond without shrimp mortalities had no measurable toxin. Muscle toxin concentration was below 0.1 μg/g.  相似文献
