Survey data from cotton farmers in six southeastern states of the USA were used to estimate the demand and willingness-to-pay (WTP) for either retrofitting yield monitors onto cotton pickers or to purchase a yield monitor as an option with a new cotton picker. ‘Don’t know’ responses were either omitted, combined with ‘no’ responses or included as a separate category for comparing WTP and estimates of the price elasticity of demand. Our results suggest that treating the ‘don’t know’ response as a separate category provides WTP estimates that are more consistent with expectations than the other approaches. The estimated price elasticities and demand curves indicate that previous users of precision technology are more responsive to changes in price of cotton yield monitors and would be more likely to adopt them when the price decreases. These demand and WTP estimates provide important information that can be used by those who sell cotton yield monitors, as well as policy-makers who may wish to subsidize this technology. Referendum contingent valuation was useful for evaluating the demand for any new technology.  相似文献

The tick-borne pathogen, Anaplasma marginale, has a complex life cycle involving ruminants and ixodid ticks. It causes bovine anaplasmosis, a disease with significant economic impact on cattle farming worldwide. The obligate intracellular growth requirement of the bacteria poses a challenging obstacle to their genetic manipulation, a problem shared with other prokaryotes in the genera Anaplasma, Ehrlichia, and Rickettsia. Following our successful transformation of the human anaplasmosis agent, A. phagocytophilum, we produced plasmid constructs (a transposon bearing plasmid, pHimarAm-trTurboGFP-SS, and a transposase expression plasmid, pET28Am-trA7) designed to mediate random insertion of the TurboGFP and spectinomycin/streptomycin resistance genes by the Himar1 allele A7 into the A. marginale chromosome. In these trans constructs, expression of the fluorescent and the selectable markers on the transposon, and expression of the transposase are under control of the A. marginale tr promoter. Constructs were co-electroporated into A. marginale St. Maries purified from tick cell culture, and bacteria incubated for 2 months under selection with a combination of spectinomycin and streptomycin. At that time, ≤1% of tick cells contained colonies of brightly fluorescent Anaplasma, which eventually increased to infect about 80–90% of the cells. Cloning of the insertion site in E. coli and DNA sequence analyses demonstrated insertion of the entire plasmid pHimarAm-trTurboGFP-SS encoding the transposon in frame into the native tr region of A. marginale in an apparent single homologous crossover event not mediated by the transposase. Transformants are fastidious and require longer subculture intervals than wild type A. marginale. This result suggests that A. marginale, as well as possibly other species of Anaplasma and Ehrlichia, can be transformed using a strategy of homologous recombination.  相似文献

The chemically different adjuvants ‘Agral’, ‘Bond’, ‘Codacide Oil’, Li 700, ‘Silwet L’-77, and ‘Headland Guard’ were assayed to determine their effects on the rainfastness of an emulsifiable concentrate formulation of the organophosphorus pesticide chlorpyrifos. Cabbage leaves were each treated with 200×0.25-μl droplets of diluted formulation using a hand-held microapplicator. Droplet deposits were left to air-dry for 1 h prior to exposure to simulated rainfall. Rain fastness was assessed using GC residue analyses of treated leaves after exposure to 10, 20 or 30 min simulated rainfall. The results indicated that the latex-based adjuvants ‘Bond’ and ‘Headland Guard’ induced a statistically significant increase in rainfastness, results for the other adjuvants assayed being either not significant or inconclusive. Diarrhoea due to enterotoxigenic Escherichia coli with fimbriae F4 (ETEC-F4) is an important problem in neonatal and just weaned piglets and hence for the pig farming industry. There is substantial evidence for a genetic basis for susceptibility to ETEC-F4 since not all piglets suffer from diarrhoea after an ETEC-F4 infection. It is assumed that the wild boar was originally ETEC-F4 resistant and that susceptibility towards ETEC arose after domestication. There are different phenotypes in the pig determined by which of the three existing F4 variants (F4ab, F4ac or F4ad) they are susceptible or resistant for. This suggests that several F4 receptors exist, expressed individually or in combination with each other on the brush border of the piglet’s small intestine. As such, the mucin-type glycoproteins (IMTGP) are described as F4ab/ac receptors, while the intestinal neutral glycospingolipid (IGLad) is proposed as an F4ad receptor. GP74 is a putative F4ab receptor. However, the specific genes that encode for the susceptibility are not yet known. In the past decades, linkage analyses revealed that the loci encoding for the receptor(s) for the two most frequent variants F4ab and F4ac were mapped to the 13th chromosome of the pig (Sus scrofa 13, SSC13). After fine mapping, the region of interest was mapped between two microsatellite markers, Sw207 and S0075, and interesting candidate genes surfaced. Numerous SNP analyses and a few expression studies on the three MUC-genes (MUC4, MUC13 and MUC20) and the transferrin receptor gene (TFRC) as well as on some other positional candidate genes have been performed in order to find the causative mutation for the ETEC-F4ab/ac receptor(s). However, until today, the exact mutation causing susceptibility to ETEC-F4 remains unknown.  相似文献

Potassium channels are K+-selective protein pores in cell membrane. The selectivity filter is the functional unit that allows K+ channels to distinguish potassium (K+) and sodium (Na+) ions. The filter's structure depends on whether K+ or Na+ ions are bound inside it. We synthesized a K+ channel containing the d-enantiomer of alanine in place of a conserved glycine and found by x-ray crystallography that its filter maintains the K+ (conductive) structure in the presence of Na+ and very low concentrations of K+. This channel conducts Na+ in the absence of K+ but not in the presence of K+. These findings demonstrate that the ability of the channel to adapt its structure differently to K+ and Na+ is a fundamental aspect of ion selectivity, as is the ability of multiple K+ ions to compete effectively with Na+ for the conductive filter.  相似文献

An 8-year-old Somali cat presented with a 9-month history of inappetence, vomiting and weight loss. The disease progressed to involve neurological signs associated with a mass lesion at the level of the first lumbar vertebra. Histopathology identified the condition as malignant histiocytosis affecting the lungs, stomach, mesenteric lymph nodes, liver, spleen, brain and spinal cord. However, the presentation of this case differs from previously reported cases of malignant histiocytosis, and may therefore represent a variant form of histiocytic disease.  相似文献

Diameter distributions of juvenile loblolly pine (Pinus taeda L.) were characterized utilizing a two-parameter Weibull distribution to aid in forecasting and simulation of young stands. Juvenile diameter distributions were studied to gain insight into the effects of various stand-level factors. Results show that diameter distributions in juvenile loblolly pine stands can be successfully characterized with the two-parameter Weibull function. Repeated measures analysis detected significant planting density, age, and age by planting density interaction effects for the scale and shape parameter estimates from the two-parameter Weibull distribution. Using parameter recovery techniques, estimated diameter distributions were derived from easily attainable stand-level characteristics (i.e. basal area per hectare, planting density, age, and quadratic mean diameter). A thorough understanding of juvenile diameter distributions should prove especially useful for operational planning of stands on short rotations that require estimates of productivity at early ages.  相似文献

Variation among isolates of the Sirococcus shoot blight pathogen (attributed to Sirococcus conigenus, Sirococcus sp., or one of many synonyms) was studied. Inter‐simple‐sequence‐repeat‐anchored polymerase chain reaction fingerprints, nuclear rDNA internal transcribed spacers and 5.8S sequences, and 18S rDNA sequences were analysed. In addition, culture morphology as well as sizes and shapes of conidia produced on seed cone scales in culture were compared. Two main groups were distinguished based on both genetic markers and cultural characteristics. The P group consists of isolates mostly from pine and spruce from Europe and both eastern and western North America; the T group consists of isolates mostly from hemlock from western North America. Results provide a benchmark for comparison of other isolates of these fungi and indicate the need for additional sampling and comparative research of these pathogens.  相似文献
