Copper (Cu) is accumulating in agricultural soils worldwide creating concern for adverse impacts on soil microbial communities and associated ecosystem services. In order to evaluate the structural and functional resilience of soil microbial communities to increasing Cu levels, we compared a Cu-adapted and a corresponding non-adapted soil microbial community for their abilities to resist experimental Cu pollution. Laboratory soil microcosms were set-up with either High-Cu soil from Cu-amended field plots (63 g Cu m−2) or with Low-Cu control soil from the same five-year field experiment. Laboratory treatments consisted of Cu amendments in the presence or absence of pig manure. Microbial activities (soil respiration, substrate-induced respiration, [3H]leucine incorporation), bacterial community structure (terminal restriction fragment length polymorphism, T-RFLP), community-level physiological profiles, and pollution-induced bacterial community tolerance (PICT detected using the [3H]leucine incorporation technique) were monitored for 12 weeks. The High-Cu and Low-Cu soil microbial communities initially exhibited almost identical structure and function and could only be distinguished from each other by their differential Cu tolerance. Experimental Cu pollution inhibited microbial activities, affected bacterial community structure, and induced further bacterial community tolerance to Cu. However, Low-Cu and High-Cu soil microbial communities showed essentially identical responses. Manure amendment did not protect against Cu toxicity and slightly increased Cu bioavailability as measured by a Cu-specific whole-cell bacterial biosensor. Our results indicate convergence of bacterial community structure and function in the High-Cu and Low-Cu soils during the five-year field experiment. We conclude that soil bacterial communities can exhibit structural and functional resilience to a five-year Cu exposure by virtue of their ability to develop Cu tolerance without affecting overall community structure. The observed increased Cu tolerance may involve phenotypic adaptation or selection at the micro-diversity level, for example an increased proportion of Cu-resistant strains within each bacterial species, which go undetected by T-RFLP community fingerprinting. Finally, our results indicate that Cu-dissolved organic matter complexes contribute to microbial toxicity in manure-amended soils implying that free Cu may comprise a poor predictor of metal toxicity. 相似文献

The combination of high input costs and low commodity prices is forcing some farmers to consider reducing crop inputs like seed, fertilizer and herbicides. In a field trial in which different canola (Brassica napus L.) and barley (Hordeum vulgare L.) inputs were subtracted from a full package, or added to an empty package, we studied the effects of full or reduced fertilizer and herbicide inputs on soil microbiological characteristics at two sites from 2005 to 2008. The full package consisted of a high-yielding crop variety seeded at an optimum rate, with fertilizers and herbicides applied at recommended rates. The empty package consisted of a less expensive, low-yielding crop variety seeded at a low rate, with no fertilizer or herbicide applied. Between these two extremes were treatments in which fertilizers or herbicides were applied at 50% of recommended rates or not at all. Each treatment was repeated year after year in the same plot, i.e., treatment effects were cumulative. Fertilizer effects on soil microbial biomass C (MBC), β-glucosidase enzyme activity and bacterial functional diversity (based on community-level physiological profiles) were usually positive. Reduced fertilizer application rates reduced the beneficial fertilizer effects. Significant herbicide effects on soil microbiological properties occurred less often, were smaller in magnitude than fertilizer effects, and were mostly negative. Reduced herbicide rates reduced the deleterious herbicide effects. These significant fertilizer and herbicide effects were observed in canola more than barley, and mostly in the final year of the study, indicating the cumulative nature of treatment effects over time. Therefore, repeated applications of agricultural inputs like fertilizers and herbicides can have more significant effects on soil biology and biological processes than single applications indicate. 相似文献

Background
There is a discrepancy in the reproductive performance between different cattle breeds. Using abattoir-derived ovaries and data base information we studied the effects of breed on in vitro fertilization and early embryo development.Methods
The in vitro developmental competence of oocytes from cattle (n = 202) of Swedish Red (SR), Swedish Holstein (SH) and mixed beef breeds was compared, retrospectively tracing donors of abattoir-derived ovaries using a combination of the national animal databases and abattoir information. Age was significantly lower and carcass conformation score was higher in the beef breeds than in the dairy breeds.Cumulus oocyte complexes (n = 1351) were aspirated from abattoir-derived ovaries from animals of known breed (visual inspection confirmed through databases), age (databases), and abattoir information. Oocytes were matured, fertilized (frozen semen from two dairy bulls) and cultured according to conventional protocols. On day 8, blastocysts were graded and the number of nuclei determined.Results
Cleavage rate was not different between the breeds but was significantly different between bulls. The percentage of blastocysts on day 8 was significantly higher when the oocyte donor’s breed was beef or SR than SH. There was no significant difference in blastocyst grades or stages between the breeds, but the number of nuclei in day 8 blastocysts was significantly lower in SH compared to the beef.Conclusions
The use of abattoir-derived ovaries from animals whose background is traceable can be a valuable tool for research. Using this approach in the present study, oocyte donor breed was seen to affect early embryo development during in vitro embryo production, which may be a contributing factor to the declining fertility in some dairy breeds seen today. 相似文献197.

Agroforestry is a promising method for enhancing land-use sustainability in the Bolivian Andes. However, its benefits in terms of rural development are under-recognized due to gaps in understanding users?? perceptions while taking into consideration both local and global environmental goals. Our study aimed to narrow these gaps by developing an analytical framework for analyzing the site-specific socio-ecological factors and interactions related to local woody species and assessing their ecological, economic, and socio-cultural plant values in order to identify the most promising agroforestry species. The framework was then tested in an indigenous community at 2,760?C3,830?m?a.s.l., incorporating vegetation surveys, environmental studies, and interviews on plant functions. Ecological, economic, and socio-cultural values and the ecological apparency of plants were calculated, and detrended correspondence and principal component analyses helped to reveal the socio-ecological context of significant factors for plant distribution and uses. Results showed dominating seral woody species along an altitudinal gradient. Although shrubs were more ecologically apparent than trees, trees were perceived to be more valuable as the usefulness and cultural importance of species increased with plant height and timber availability. Phytosociological factors played a minor but still significant role in perceived usefulness. Schinus molle and Prosopis laevigata (<3,200?m a.s.l.), Polylepis subtusalbida (>3,200?m a.s.l.), and Baccharis dracunculifolia (both zones) were evaluated as most promising for agroforestry use. In conclusion, our analytical framework proved to be a valuable tool for context-specific agroforestry plant selection. Nonetheless, economic, technical, and socio-cultural limitations of cultivating native agroforestry species were revealed as well. The effects of floor heating at parturition on sow and piglet behaviour in pens were examined in a Danish production herd. A total of 50 gilts and sows were split into two groups; one (n = 25) experienced no treatment; one (n = 25) was exposed to floor heating in the area of the sow around parturition. The sows and piglets were recorded on video 24 h a day for behavioural analyses.Floor heating tended to improve piglet weight at weaning (P = 0.05) but did not affect average daily gain. Piglets in pens with floor heating were more often in proximity of the sow at posture changes than piglets from pens without floor heating on day one postpartum (P = 0.01). OBJECTIVE: To compare obese horses with insulin resistance (IR) with nonobese horses and determine whether blood resting glucose, insulin, leptin, and lipid concentrations differed between groups and were correlated with combined glucose-insulin test (CGIT) results. ANIMALS: 7 obese adult horses with IR (OB-IR group) and 5 nonobese mares. PROCEDURES: Physical measurements were taken, and blood samples were collected after horses had acclimated to the hospital for 3 days. Response to insulin was assessed by use of the CGIT, and maintenance of plasma glucose concentrations greater than the preinjection value for > or = 45 minutes was used to define IR. Area under the curve values for glucose (AUC(g)) and insulin (AUC(i)) concentrations were calculated. RESULTS: Morgan, Paso Fino, Quarter Horse, and Tennessee Walking Horse breeds were represented in the OB-IR group. Mean neck circumference and BCS differed significantly between groups and were positively correlated with AUC values. Resting insulin and leptin concentrations were 6 and 14 times as high, respectively, in the OB-IR group, compared with the nonobese group, and were significantly correlated with AUC(g) and AUC(i). Plasma nonesterified fatty acid, very low-density lipoprotein, and high-density lipoprotein-cholesterol (HDL-C) concentrations were significantly higher (86%, 104%, and 29%, respectively) in OB-IR horses, and HDL-C concentrations were positively correlated with AUC values. CONCLUSIONS AND CLINICAL RELEVANCE: Measurements of neck circumference and resting insulin and leptin concentrations can be used to screen obese horses for IR.
