Selamectin, a novel avermectin, was evaluated in two controlled studies (one in Beagles, one in domestic shorthaired cats) to determine an appropriate topical dose for efficacy against adult Ctenocephalides felis felis (C. felis) fleas on dogs and cats for 1 month. For each study, animals were allocated randomly to four treatments. One treatment consisted of the inert formulation ingredients (vehicle) administered as a negative control, and the other three treatments consisted of a single topical dosage of 3, 6, or 9mgkg(-1) of selamectin. In each study, selamectin was administered as a topical dose applied to the skin in a single spot at the base of the neck in front of the scapulae. Dogs and cats were infested with 100 viable unfed C. felis (50 males and 50 females) on days 4, 11, 18, and 27. Seventy-two hours (+/-2h) after each infestation, on days 7, 14, 21, and 30, a comb count to determine the number of viable fleas present on each animal was performed. Efficacy of selamectin on day 30 was used to select an appropriate dose. For dogs and cats, percentage reductions in geometric mean flea comb counts for the three selamectin treatments ranged from 94. 6 to 100% on days 7, 14, and 21, compared with the negative-control treatment. On day 30, reductions in flea comb counts were 81.5, 94.7, and 90.8% for dogs, and 79.8, 98.0, and 96.2% for cats treated with selamectin at 3, 6, or 9mgkg(-1), respectively. For day 30 flea comb counts for dogs and cats, analysis of variance showed that the three selamectin treatments resulted in significantly (P< or =0.05) lower counts than did the negative-control treatment. For dogs and cats, geometric mean flea counts for selamectin administered at a dosage of 3mgkg(-1) were significantly (P< or =0.05) higher than those for the 6 and 9mgkg(-1) treatment dosages combined. There were no significant differences in flea counts between the 6 and 9mgkg(-1) treatments. This analysis was confirmed by linear-plateau modeling. Thus, the optimal dose of selamectin for efficacy against adult fleas for both dogs and cats, as estimated by the turning point (plateau) in the dose response curve, was 6mgkg(-1).  相似文献

Gastrointestinal nematode parasitism is a concern to small ruminants worldwide. Productivity has been compromised because such nematodes, particularly Haemonchus contortus, have developed resistance to available anthelmintics. Some sheep breeds and lines within breeds are relatively resistant to infection, a trait that may be useful for developing control strategies. Suffolk sheep, which are susceptible to infection, were crossed with Gulf Coast Native sheep, which are more resistant to infection, to produce F1 progeny. F1 rams were bred to F1 ewes which produced 227 F2 offspring. These F2 offspring were evaluated for variability in infection levels, based on fecal egg count (FEC) and blood packed cell volume (PCV), under two natural infection conditions (one at weaning and another after a summer grazing period) and one experimental infection. The range of both FEC and PCV was large for all three infection periods with annual variation. Overall, the range for the three infection periods, respectively, were 167-149,933, 0-31,400 and 17-114,667 eggs per gram (EPG) of feces and 8.7-37.0%, 7.3-33.0% and 8.3-36.0%. This segregation of infection is what would be expected of F(2) progeny from susceptible and resistant parent breeds. Heritabilities of FEC and PCV for the three infection periods, respectively, were 0.15, 0.29 and 0.12, and 0.11, 0.22 and 0.12. Based on segregation of infection, larger heritabilities and maternal environment effects that declined after weaning, the summer natural infection was probably the best model for assessing genetic variation.  相似文献

Two experiments were conducted to examine how oocyte maturation and fertilization influence zygotic diversity in swine. In the first experiment, the distribution of oocyte maturation was compared to that of zygotic development. Oocytes were aspirated from follicles of 31 gilts and classified into stages of meiosis. Zygotes were flushed from oviducts of 19 additional gilts and classified into stages of meiosis and fertilization. The second experiment examined whether the time from ovulation to fertilization was constant among all oocytes. To test this premise, four to six oocytes from follicles of 10 mated gilts were aspirated just before or during ovulation, stained and transferred back into the oviducts of these same gilts. Zygotes were recovered 10 h later to determine whether the first oocytes ovulated were the more developed zygotes and, conversely, whether the last oocytes to be ovulated represented the lesser developed contemporaries. The skewed (P less than .05) distribution of oocyte maturation was similar to that of zygotic development. Regression of the frequency distribution describing early oocyte maturation resulted in a line with a slope (.59) that was similar to the slope (.58) of the regressed distribution of zygotic development. Likewise, the order of ovulation and order of subsequent stages of zygotic development were similar. These data suggest that variation in zygotic development in swine was due to variability in oogenesis; the time from ovulation to fertilization appeared to be constant.  相似文献

In 2011, following severe flooding in Eastern Australia, an unprecedented epidemic of equine encephalitis occurred in South-Eastern Australia, caused by Murray Valley encephalitis virus (MVEV) and a new variant strain of Kunjin virus, a subtype of West Nile virus (WNVKUN). This prompted us to assess whether a delta inulin-adjuvanted, inactivated cell culture-derived Japanese encephalitis virus (JEV) vaccine (JE-ADVAX™) could be used in horses, including pregnant mares and foals, to not only induce immunity to JEV, but also elicit cross-protective antibodies against MVEV and WNVKUN. Foals, 74–152 days old, received two injections of JE-ADVAX™. The vaccine was safe and well-tolerated and induced a strong JEV-neutralizing antibody response in all foals. MVEV and WNVKUN antibody cross-reactivity was seen in 33% and 42% of the immunized foals, respectively. JE-ADVAX™ was also safe and well-tolerated in pregnant mares and induced high JEV-neutralizing titers. The neutralizing activity was passively transferred to their foals via colostrum. Foals that acquired passive immunity to JEV via maternal antibodies then were immunized with JE-ADVAX™ at 36–83 days of age, showed evidence of maternal antibody interference with low peak antibody titers post-immunization when compared to immunized foals of JEV-naïve dams. Nevertheless, when given a single JE-ADVAX™ booster immunization as yearlings, these animals developed a rapid and robust JEV-neutralizing antibody response, indicating that they were successfully primed to JEV when immunized as foals, despite the presence of maternal antibodies. Overall, JE-ADVAX™ appears safe and well-tolerated in pregnant mares and young foals and induces protective levels of JEV neutralizing antibodies with partial cross-neutralization of MVEV and WNVKUN.  相似文献

The addition of the organosilicone surfactant ‘Silwet L77’ at 1-5 ml litre?1 to formulated glyphosate gave complete surface wetting on application to the adaxial leaf surface of perennial ryegrass (Lolium perenne L.). The wetting characteristics of the solution were associated with rapid foliar uptake and near maximal uptake in 3 h compared to greater than 5 h in the absence of ‘Silwet L77’. Evidence is presented showing that solutions containing ‘Silwet L77’ rapidly infiltrate stomata. Rapid uptake did not occur after application to the astomatous abaxial surface of perennial ryegrass leaves. The rapid rate of glyphosate uptake reduced the critical rainfall period to 2 h or less, compared to up to 10 h in the absence of ‘Silwet L77’. The use of ‘Silwet L77’ has major practical implications for the use of glyphosate in regions with unpredictable rainfall or high rainfall frequency.  相似文献

ABSTRACT Local and regional employment growth is generally studied either by searching for local qualitative explanatory factors such as governance, synergy between firms, and milieu effects, or by searching for general growth factors using statistical techniques. The body of work that relies on this approach has tended, in keeping with economics’ nomothetic tradition, to assume that local and regional growth factors are constant over space. The focus of this paper is on exploring the spatial stationarity of employment growth factors in Canada, but it also seeks to clarify some of the broad principles behind spatial regression techniques in order to provide a point of entry and a conceptual framework for empirical researchers. To do so, we apply a recently developed technique, Geographically Weighted Regression (GWR), and we explore the method's advantages and limits for answering our research question. We find evidence that growth factors differ across Canada, but we also conclude that the GWR technique, given the number and shape of regions available for our analysis and given certain limitations that are currently inherent to the method, can only provide tentative and exploratory results.  相似文献

It is often necessary to predict the distribution of mineral particles in soil between size fractions, given observations at sample sites. Because the contents in each fraction necessarily sum to 100%, these values constitute a composition, which we may assume is drawn from a random compositional variate. Elements of a D‐component composition are subject to non‐stochastic constraints; they are constrained to lie on a D– 1 dimensional simplex. This means we cannot treat them as realizations of unbounded random variables such as the multivariate Gaussian. For this reason, there are theoretical reasons not to use ordinary cokriging (or ordinary kriging) to map particle size distributions. Despite this, the compositional constraints on data on particle size fractions are not always accounted for by soil scientists. The additive log‐ratio (alr) transform can be used to transform data from a compositional variate into a form that can be treated as a realization of an unbounded random variable. Until now, while soil scientists have made use of the alr transform for the spatial prediction of particle size, there has been concern that the simple back‐transform of the optimal estimate of the alr‐transformed variables does not yield the optimal estimate of the composition. A numerical approximation to the conditional expectation of the composition has been proposed, but we are not aware of examples of its application and it has not been used in soil science. In this paper, we report two case studies in which we predicted clay, silt and sand contents of the soil at test sites by ordinary cokriging of the alr‐transformed data followed by both the direct (biased) back‐transform of the estimates and the unbiased back‐transform. We also computed estimates by ordinary cokriging of the untransformed data (which ignores the compositional constraints on the variables) for comparison. In one of our case studies, the benefit of using the alr transform was apparent, although there was no consistent advantage in using the unbiased back‐transform. In the other case study, there was no consistent advantage in using the alr transform, although the bias of the simple back‐transform was apparent. The differences between these case studies could be explained with respect to the distribution on the simplex of the particle size fractions at the two sites.  相似文献
