Two different strategies for sweeteners determination in tabletop samples by Fourier transform middle-infrared (FTIR) spectrometry, an off-line and a fully mechanized extraction of Aspartame and Acesulfame-K with different mixtures of chloroform and methanol, have been developed. The off-line method involves the extraction of both active principles by sonication of samples with 25:75 v/v CHCl3/CH3OH and direct measurement of the peak height values at 1751 cm(-1), corrected using a baseline defined at 1850 cm(-1) for Aspartame, and measurement of the peak height at 1170 cm(-1) in the first-order derivative spectra, corrected by using a horizontal baseline established at 1850 cm(-1), for Acesulfame-K. Limit of detection values of 0.10 and 0.9% w/w and relative standard deviations of 0.17 and 0.5% were found for Aspartame and Acesulfame-K, respectively. The time needed for the sweeteners determination is reduced from 35 min for the HPLC method to 7 min by FTIR. On the other hand, the fully mechanized on-line extraction avoids the contact of the operator with toxic solvents and differentiates between samples that contain Aspartame and Acesulfame-K and those that include only Aspartame, reducing the time needed for the analysis of the last kind of samples to 5 min. 相似文献

Several commercial soft drinks and respective plastic bottles were analyzed for their multielement contents employing the synchrotron radiation total reflection X-ray fluorescence spectrometry technique (SRTXRF). The SRTXRF method has been developed and validated, and about 20 elements were detected in the investigated samples, including some trace elements, which can be toxic for human beings, such as Ti, Cr, Sb, As, and Pb in soft drinks and Al, Sb, As, and Pb in poly(ethylene terephthalate) (PET) containers. Statistical analysis was performed using chemometric techniques (principal component analysis and cluster analysis), and similarities were verified in the multielement contents of the samples. The results demonstrated that the SRTXRF offers a good multielemental approach for the quality control of food products. Moreover, on the basis of enrichment factors, the possibility of the trace elements in the PET container may be leached to the beverages under normal commercial situations and other results were discussed. 相似文献

Several demographic parameters of Balearic shearwater (Puffinus mauretanicus) were estimated and used to model extinction probabilities for the species, which is an endemic seabird confined to a Mediterranean archipelago. Adult survival was analyzed through capture-recapture models at two colonies free of adult predators during 1997-2002. Extinction probabilities for the world breeding population were modelled using Monte Carlo simulations of population trajectories, introducing stochasticity both demographic and environmental. Adult survival estimate (0.780, SE=0.020, 95% Confidence Intervals: 0.739-0.816) was unusually low for a Procellariiform, suggesting that sources of mortality other than predators (e.g. fishing gears) also occur. A deterministic model showed a declining trend for the world breeding population (λ=0.952), and significantly less than 1. Population viability analysis showed that in the presence of environmental and demographic stochasticities, mean extinction time for the world population was estimated at 40.4 years (SE=0.2), and mean growth rate showed a 7.4% decrease per year. Feline immunodeficiency virus (FIV) is a naturally occurring lentivirus of domestic cats, and is the causative agent of feline AIDS. Similar to human immunodeficiency virus (HIV), the pathogenesis of FIV involves infection of lymphocytes and macrophages, and results in chronic progressive immune system collapse and death. Neuropathologic correlates of FIV infection have not yet been elucidated, and may be relevant to understanding HIV-associated neurologic disease (neuroAIDS). As in HIV, FIV strains have been shown to express differential tendencies towards development of clinical neuroAIDS. To interrogate viral genetic determinants that might contribute to neuropathogenicity, cats were exposed to two well-characterized FIV strains with divergent clinical phenotypes and a chimeric strain as follows: FIV(PPR) (PPR, relatively apathogenic but associated with neurologic manifestations), FIV(C36) (C36, immunopathogenic but without associated neurologic disease), and Pcenv (a chimeric virus consisting of a PPR backbone with substituted C36 env region). A sham inoculum control group was also included. Peripheral nerve conduction velocity, CNS imaging studies, viral loads and hematologic analysis were performed over a 12 month period. At termination of the study (350 days post-inoculation), brain sections were obtained from four anatomic locations known to be involved in human and primate lentiviral neuroAIDS. Histological and immunohistochemical evaluation with seven markers of inflammation revealed that Pcenv infection resulted in mild inflammation of the CNS, microglial activation, neuronal degeneration and apoptosis, while C36 and PPR strains induced minimal neuropathologic changes. Conduction velocity aberrations were noted peripherally in all three groups at 63 weeks post-infection. Pcenv viral load in this study was intermediate to the parental strains (C36 demonstrating the highest viral load and PPR the lowest). These results collectively suggest that (i) 3' C36 genomic elements contribute to viral replication characteristics, and (ii) 5' PPR genomic elements contribute to CNS manifestations. This study illustrates the potential for FIV to provide valuable information about neuroAIDS pathogenesis related to genotype and viral kinetics, as well as to identify strains useful to evaluation of therapeutic intervention. 相似文献

The Spanish type cultivar ‘Piel de Sapo’ (Cucumis melo L. var. saccharinus), has a limited compatibility with the Cucurbita maxima × Cucurbita moschata hybrids currently used as rootstocks. Double grafting can be used to improve compatibility between rootstock and scion by means of an intermediate rootstock compatible with both. Non-grafted, single, and double grafted melon plants of the cultivar Piel de Sapo were evaluated for water, nutrient absorption, photosynthesis activity, biomass production in early phases, as well as for yields and fruit quality in a long term trial. The hybrid ‘Shintoza’ (C. maxima × C. moschata) was used as rootstock, and the cantaloupe type melon cultivar ‘Sienne’ as an intermediate scion. Grafting did not affect net photosynthetic values, yet increased water use efficiency (35%). Double grafted plants increased aerial dry weights (66% and 31% with respect to non-grafted and simple graft plants, respectively), and also increased capacity for uptaking beneficial minerals (between 61% and 13% and particularly for NO3−, P, K, Ca, Mn, and Zn) with respect to non-grafted and single grafted plants. The quantum efficiency PSII photochemistry values increased in double grafted plants (12%) with respect to the control plants. Consequently, double grafting on a vigorous rootstock such as ‘Shintoza’, with an intermediate scion, results in improved mineral and water absorption and achieves an increase in ion influx to the scion – so enabling an increase in light photosynthetic reaction and biomass. Double grafted plants increased fruit yield when compared to simple grafted and non-grafted plants (12% and 56%, respectively) and did not affect fruit quality in terms of °Brix and colour. In conclusion, double grafting presents several beneficial aspects that are counter-balanced by the extra cost of the technique. The difference in yields reflects compatibility problems. 相似文献

Taste and aroma related compounds have been analyzed in a collection of four traditional varieties and two tomato hybrids, representing a wide variability in fruit shape and color, grown in different environments: screenhouse and open field. Protected cultivation tended to show lower sugar concentration (fructose and glucose) but similar acid contents (citric, malic, and glutamic acids). The decreased levels of sucrose equivalents and the similar ratios of sucrose equivalents to citric or glutamic acid contents indicated that protected cultivation, despite being useful to reduce the incidence of pests and viral diseases, reduces the organoleptic quality. Additionally, it doubles the interaccession variability and increased the level of intra-accession variability. In the case of aroma, the genotypic effect was considerably higher than the environmental component on the 12 main volatiles analyzed. Only hexanal and methyl salicylate were significantly affected by environment, while 10 out of 12 volatiles were affected by the genotype. Biplot analysis showed that, even in considerably different environments, it is possible to identify genotype-dependent main aroma profiles. In the case of 13 background volatiles, the environment showed no significant effects and the genotypic effect was lower, though it is possible to identify genotypic trends in background notes. 相似文献

In the Pacific Ocean, air and ocean temperatures, atmospheric carbon dioxide, landings of anchovies and sardines, and the productivity of coastal and open ocean ecosystems have varied over periods of about 50 years. In the mid-1970s, the Pacific changed from a cool "anchovy regime" to a warm "sardine regime." A shift back to an anchovy regime occurred in the middle to late 1990s.
