Tritrichomonas foetus is the causative agent of sexually transmitted trichomoniasis in cattle. In females, the infection can be associated with infertility, vaginitis, endometritis, abortion or pyometra, leading to significant economic losses in cattle raising. T. foetus is devoid of the ability to synthesize purine nucleotides de novo, depending instead on salvaging purines from the host environment. Ecto-5'-nucleotidase catalyzes the final step of extracellular nucleotide degradation, the hydrolysis of nucleoside 5'-monophosphates to the corresponding nucleosides and Pi. In this work we show that living, intact cells of T. foetus were able to hydrolyze 5'AMP at a rate of 12.57 ± 1.23 nmol Pi × h(-1) × 10(-7) cells at pH 7.2 and the 5'AMP hydrolysis is due to a plasma membrane-bound ecto-enzyme activity. The apparent K(m) for 5'AMP was 0.49 ± 0.06 mM. In addition to 5'AMP, the enzyme hydrolyzed all substrate monophosphates tested except 3'AMP. No divalent metals or metal chelators were able to modulate enzyme activity. Phosphatase inhibitors did not have an effect on ecto-5'-nucleotidase activity while ammonium molybdate did inhibit the activity in a dose dependent manner. The presence of adenosine in the culture medium negatively modulated the enzyme. These results indicate the existence of an ecto-5'-nucleotidase that may play a role in the salvage of purines. 相似文献

The aim of the present study was to evaluate the in vitro and in vivo effect of the addition of superoxide dismutase (SOD) and reduced glutathione (GSH) to ram semen freezing extender. Significant differences (p < 0.05) were detected between groups regarding total motility (TM), straightness (STR) and wobble (WOB), for which the GSH 7 mM group had lesser TM and better STR than the other groups and the GSH 5 and 7 mM groups had higher wobble values than the control, SOD 25 and 100 U/ml groups. The ultrastructural analysis revealed that the acrosome was better preserved after freezing in the SOD 100 U/ml and GSH 2 and 5 mM (p < 0.05) groups than the other groups, whereas mitochondria in both the control group and the 7 mM GSH group suffered the greatest damage. The plasma membrane remained preserved after freezing, regardless of the group. For in vivo fertilization, the SOD group achieved better results than the GSH group (p > 0.05). It can therefore be concluded that the addition of SOD 100 U/ml and GSH 2 and 5 mM preserves the acrosome integrity of frozen ram spermatozoa, while the addition of SOD 100 U/ml to Tris egg-yolk extender offers protection to the membranes of sperm cells after thawing. 相似文献

BACKGROUND
Tuta absoluta (Meyrick) is a major pest of tomato produced in glasshouses and open field, causing severe damages to crops, reducing the quality of tomato fruits. The current maintenance of the pest populations below the economic threshold is not achieved by natural and classical control, thus requiring the continuous application of biological control agents (BCAs), under an augmentative or inoculative approach. The present study aims to develop an economic and financial model to evaluate the commercial viability of a continuous mass production of Macrolophus pygmaeus (Rambur), a BCA commonly used against the tomato moth, Tuta absoluta, in protected culture. The estimations for our model were based on two approaches: the farm-level impact analysis and the benefit–cost analysis.RESULTS
The results of the farm-level analysis show that the adoption of a more sustainable biological control approach is profitable for farmers and the benefit–cost analysis provides evidence that the investment on a new factory dedicated to the mass rearing of M. pygmaeus to control tomato moth populations generates a positive net present value (NPV) of 7.2 million euros, corresponding to an internal rate of return (IRR) of 28.4% per year.CONCLUSION
Our results are in line with (i) the more recent European Commission proposals for a new Regulation on sustainable use of plant protection products, which includes the reduction of 50% the use and risk of chemical pesticides by 2030 and (ii) most of the existing literature which conclude that new projects on BCA production are worth investments. © 2023 The Authors. The effects of bovine leukemia virus (BLV) on the immune response have been extensively investigated; however, its effects on mammary gland immunity are only speculative. Although BLV has a tropism for B cells, it can affect both adaptive and innate immunities because these systems share many effector mechanisms. This scenario is the basis of this investigation of the effects of BLV on mammary gland immunity, which is largely dependent upon neutrophilic functions. Thus, the present study sought to examine neutrophilic functions and the lymphocyte profile in the milk of naturally BLV-infected cows. The viability of the milk neutrophils and the percentage of milk neutrophils that produced reactive oxygen species (ROS) or phagocytosed Staphylococcus aureus were similar between BLV-infected and BLV-uninfected dairy cows. Furthermore, the expression of CD62L and CD11b by the milk neutrophils and the percentage of milk neutrophils (CH138+ cells) that were obtained from the udder quarters of the BLV-infected cows were not altered. Conversely, the median fluorescence intensity (MFI) representing intracellular ROS production and the phagocytosis of S. aureus, the expression of CD44 by the milk neutrophils and the percentage of apoptotic B cells were lower in the milk cells from BLV-infected dairy cows, particularly those from animals with persistent lymphocytosis (PL). The lymphocyte subsets were not different among the groups, with the exception of the percentage of CD5−/CD11b− B cells, which was higher in the milk cells from BLV-infected cows, particularly those with PL. Cycas revoluta (Cycadaceae) is commonly cultivated as an ornamental tree in several countries. In April 2015, C. revoluta leaves showing brownish spots with a yellow halo that progressed to leaflet necrosis were observed in Belém, Pará state, Brazil. This study aimed to determine the aetiology of this disease on sago palm in Brazil. Based on DNA sequence-based phylogeny, morphology, and colony characterization, the fungus was identified as Pseudopestalotiopsis elaeidis. To the best of our knowledge, this is the first report of P. elaeidis associated with leaf spots on C. revoluta worldwide. 相似文献

在系统分析能值—生态足迹模型缺陷的基础上,通过增加人工投入指标,统一能值密度以及引入折能/折标系数对其进行了修正,并采用修正后的模型对江苏省南通市1996—2010年的可持续发展水平进行了测算。结果表明:(1)1996—2010年间南通市人均生态足迹由0.65hm2增长到0.90hm2,主要是由能源消费量的增加引起的;(2)同期南通市人均生态承载力由0.51hm2增长到0.81hm2,这主要是由农、畜产品产量提高和水电开发力度加大引起的,而通过劳务和资金投入提高承载力的空间很大;(3)1996—2010年南通市一直处于生态赤字状态,可持续性评价指数也都在0.5以上,区域发展处于不可持续状态。为进一步缩小生态承载力与生态足迹之间的差距,应采取以下措施:引导重点行业和企业节能降耗,大力发展新能源;加大农业科技投入,继续提高农产品产量;重视人力资源的投入,促使经济发展方式向技术密集型和资本密集型转变。 相似文献

Background: The current study evaluated the subcutaneous fatty acid(FA) composition of calf- and yearling-fed stee with or without growth promoting implants. Crossbred steers(n = 112; 267 ± 5.0 kg) of the same contemporary group were allocated to one of four production system and implant strategy based treatments in a completely randomized design with a 2 × 2 factorial arrangement of treatments.Results: There were no interactions(P 0.05) between production systems and growth promoting implants for the total and individual subcutaneous FA. Yearling as opposed to calf finishing reduced(P 0.05) subcutaneous proportions of C20:3n-6, trans(t)12-18:1, C14:0, several minor cis-monounsaturated FA(c-MUFA; c9-14:1, c11-16:1,c11-18:1, c12-18:1, c13-18:1, c9-20:1 and c11-20:1), and increased(P 0.05) subcutaneous proportions of t11c15-18:2,total and individual branched-chain FA. Subcutaneous fat from steers implanted with growth promotants had higher(P 0.05) proportions of total polyunsaturated FA(PUFA), total n-6 PUFA, C18:2n-6 and individual t-18:1isomers(t6 to t10) compared to non-implanted steers.Conclusions: Overall, current findings show that production systems and growth promotants led to only minor differences in subcutaneous FA composition of beef steers. 相似文献

Litopenaeus schmitti juveniles (total length 15 ± 0.4 cm) were exposed to different concentrations of nitrite using the static renewal method at different salinity levels (5‰, 20‰ and 35‰) at pH 8.0 and 20 °C. The 24, 48, 72 and 96 h LC50 values of nitrite in L. schmitti juveniles were 40.72, 32.63, 24.63, and 19.12 mg L−1 at 5‰; 53.52, 38.60, 27.76, 25.55 mg L−1 at 20‰; 54.32, 47.87, 41.67 and 38.88 mg L−1 at 35‰ salinity. As the salinity decreased from 35‰ to 5‰, susceptibility to nitrite increased by 33.4%, 46.7%, 69.2% and 103.3%, after 24, 48, 72 and 96 h of exposure respectively. Furthermore, we found that exposure of shrimp to nitrite caused an increase in oxygen consumption by 137.3%, 99.2% and 81.4% and an increase in the ammonia excretion level by 112.5%, 87% and 64.3% with respect to the control with decreasing salinity levels. 相似文献
